Die erfindungsgemässe Herstellung der Amide kann beispielsweise wie folgt durchgeführt werden: Man lässt ein Pyrazol der Formel 1I mehrere Stunden mit einem primären Amin der Formel III bei Raum temperatur oder erhöhter Temperatur stehen. Nach Entfernen von überschüssigem Amin und von durch die Reaktion abgespaltenem Pyrazol wird der Rück stand durch Kristallisation und/oder Chromatogra- phie gereinigt.
Die Reaktion wird durch Erhöhung der Tempe ratur beschleunigt. Falls es der Siedepunkt des zu verwendenden Amins erlaubt, arbeitet man zweck mässigerweise bei einer Temperatur von etwa 100 .
Zusatz von Lösungsmitteln erübrigt sich, wenn ein grösserer überschuss an Amin (5-20 Teile pro Teil Pyrazol-Derivat) verwendet wird. Wird jedoch nur ein geringer überschuss eingesetzt, dann ist es zweckmässig, die Umsetzung in Gegenwart eines in differenten Lösungsmittels, beispielsweise Chloro- form, vorzunehmen.
Es hat sich gezeigt, dass die Herstellung von, Amiden der Lysergsäure-Reihe mehr oder weniger von einer Umlagerung in die Iso-Form, oder umge kehrt, begleitet ist. So wird beispielsweise aus 1-(D-Lysergyl)-3,5-dimethyl-pyrazol und (+)-Butanol- amin-(2') sowohl D-Lysergsäure-(+)-butanolamid- (2') als auch D-Isolysergsäure-(+)-butanolamid-(2'1 erhalten.
Die Amide der Lyserg- und der Dihydrolyserg- säure-Reihe der Formel I spielen wegen ihrer spezi fischen pharmakodynamischen Eigenschaften in der Therapie eine wichtige Rolle.
Zu ihrer Synthese war man bis jetzt darauf angewiesen, sie aus den entsprechenden Säurehydraziden, die an und für sich durch Behandlung der natürlichen Mutterkornalka- loide mit Hydrazin relativ leicht in guter Ausbeute rein gewonnen werden können, auf dem Weg über die Säureazide herzustellen. Diese Synthesemethode war aber kompliziert und unbefriedigend, da die Azide nicht leicht zu handhabende, empfindliche Ausgangsmaterialien sind.
Das vorliegende neue Verfahren hat demgegen über den besonderen Vorteil, dass es sich unempfind licher Ausgangsmaterialien bedient, die bei Raum temperatur stabil und deshalb ohne besondere Vor sichtsmassnahmen zu handhaben sind. Diese Aus gangsmaterialien, das heisst die Pyrazole der Formel II, sind neu und können z.
B. hergestellt werden, indem man ein Hydrazid der Lyserg- oder Dihydro- lysergsäure#-Reihe der Formel

und ein 1,3-Diketon der Formel R,-CO-CH2-COR3 V in schwach saurer Lösung bei Raumtemperatur einige Zeit sich selbst überlässt.
Ein weiterer Vorteil dieses Verfahrens besteht darin, dass bei der Umsetzung keinerlei Nebenreak tionen auftreten, und das Endprodukt nach Entfer nen von überschüssigem Amin und abgespaltenem Pyrazol in reiner Form isoliert werden kann. Dieser technische Fortschritt wirkt sich ganz besonders auf die Wirtschaftlichkeit des Verfahrens aus. Dieses ist daher in hohem Masse für die techniche Herstellung der Verfahrensprodukte geeignet und gestattet die rationelle Herstellung von grossen Mengen dieser Verbindungen.
In den nachfolgenden Beispielen erfolgen alle Temperaturangaben in Celsiusgraden und sind un- korrigiert.
<I>Beispiel 1</I> 9,10-Dihydro-D-lysergsäure-äthylamid Man lässt 100 mg 1-(9',10'-Dihydro-D-Ixsergyl)- 3,5-dimethyl-pyrazol mit 2 cm3 Äthylamin 15 Stun- den bei Raumtemperatur stehen, verdampft hierauf zur Trockne, erwärmt den Rückstand einige Minuten im Vakuum auf 100 und kristallisiert das verblei bende 9,10-Dihydro-D lysergsäure-äthylamid aus Chloroform/Petroläther um.
Blättchen vom Smp. 257-2631. [ajD = -136 (c = 0,5 in Pyridin). Das als Ausgangsmaterial zur Verwendung ge langende 1-(9',10'-Dihydro-D-lysergyl)-3,5-dimethyl- pyrazol wird wie folgt hergestellt:
Eine Lösung von 1,42 g 9,10 - Dihydro - D - lysergsäure - hydrazid in 5,5 cm3 n Salzsäure, 5 cm3 Wasser und 5 cm3 Äthanol lässt man mit 1 g Acetylaceton 1 Stunde bei Raumtemperatur stehen, neutralisiert hierauf genau durch Zusatz von 5,5 em3 n Natronlauge, filtriert das ausgeschiedene Pyrazol-Derivat und wäscht es mit Wasser nach. Das im Vakuum bei 60 getrock nete Rohprodukt wird hierauf aus Chloroform unter Zusatz von Äther kristallisiert.

ver dampft anschliessend das nicht umgesetzte Butanol- amin und das Dimethyl-pyrazol durch Erhitzen im Hochvakuum auf 110 und kristallisiert das zurück bleibende 9,1(@-Dihydro-D-lysergsäure-(+)-butanol- amid-(2') aus einem Gemisch von Chloroform/Me- thanol unter Zusatz von Petroläther.

[a] D = - l49 (c = 0,5 in Pyridin). <I>Beispiel 3</I> D-Lysergsäure-(+)-butanolamid-(2') und D-Isolysergsäure-(+)-butanolamid-(2') Man lässt 200 mg 1-(D-Lysergyl)-3,5-dimethyl- pyrazol mit 3 cm3 (+)-Butanolamin-(2') 6 Stunden bei Raumtemperatur stehen, wobei langsam voll ständige Lösung eintritt.
Anschliessend wird das überschüssige Butanolamin und das abgespaltene 3,5-Dimethyl-pyrazol durch Erhitzen im Hoch vakuum auf 110 entfernt, und das verbleibende Gemisch von D-Lysergsäure-(+)-butanolamid-(2') und D-Isolysergsäure-(+)-butanolamid-(2') an einer Säule von 8 g Aluminiumoxyd chromatographiert. D - Lysergsäure - (+) - butanolamid - (2') wird mit Chloroform + 0,2 % Äthanol ins Filtrat gewaschen. Aus Chloroform Nadeln vom Smp. 160-170 .

Das als Ausgangsmaterial zur Verwendung ge langende 1-(D-Lysergyl)-3,5-dimethyl-pyrazol wird wie folgt hergestellt: Eine Lösung von 1,41g D-Ly- sergsäure-hydrazid in 5,5 cm3 n Salzsäure, 5 cm3 Wasser und 3,5 cm3 Dimethoxy-äthan lässt man mit 0,75 g Acetylaceton 15 Minuten bei Raumtemperatur stehen, neutralisiert hierauf durch Zusatz von 5,5 cm3 n Natronlauge,
filtriert das ausgeschiedene Pyrazol- Derivat ab und wäscht es mit Wasser nach. Das im Vakuum bei 60 getrocknete Rohprodukt wird hierauf aus Chloroform unter Zusatz von Äther kristallisiert. Prismen vom Smp. 175-l77 .
