Die Ausführung des Verfahrens gestaltet sich beispielsweise wie folgt:
Man versetzt die wässrige Lösung des Hydrochlorids eines Lysergsäure- oder Dihydrolysergsäure-hydrazids der Formel 1I mit der berechneten Menge Nitrit, z. B. Natriumnitritlösung, und lässt hierauf unter Rühren bei tiefer Temperatur, vorzugsweise bei 0 bis -S , wässrige Salzsäure zu tropfen.
Aus der durch Zusatz von Natriumhydrogen- carbonat alkalisch gemachten Reaktionsmischung extrahiert man das entstandene Azid mit einem inerten, mit Wasser nicht mischbaren Lösungsmittel, vorzugsweise Benzol oder Toluol. Die so erhaltene Lösung des Azids der Formel III wird getrocknet, vorzugsweise mit Kaliumcarbonat, und hierauf so rasch als möglich auf eine Temperatur von 80-120 erhitzt.
Man lässt bis zu 10 Minuten bei der ange gebenen Temperatur stehen und fügt hierauf die Aminoverbindung der Formel IV hinzu. Für das Gelingen der Reaktion ist die Zugabe der theoretisch erforderlichen Menge einer Aminoverbindung der Formel IV ausreichend, wobei indessen zur Ver besserung der Ausbeute die Verwendung eines ge ringen überschusses zweckmässig ist. Das Gemisch wird während kurzer Zeit erwärmt und hierauf zur Trockne verdampft.

Die neuen Harnstoffverbindungen der Lyserg- säure-Reihe sind bei Raumtemperatur feste kristalli sierte Verbindungen. In Wasser sind sie sehr wenig, in organischen Lösungsmitteln mässig bis gut löslich. Mit dem Keller-Reagens [Eisen-(III)-chlorid enthal tender Eisessig und konz. Schwefelsäure] geben sie eine Farbreaktion, deren Nuance von der Art des Substituenten R1 abhängig ist.
Die nach dem vorliegenden Verfahren hergestell ten, neuen Harnstoffverbindungen sollen therapeu tisch verwendet werden. Sie wirken ausgesprochen serotoninhemmend. überdies wirken sie sedativ auf motorische und psychische Funktionen und beein flussen den Tonus der Gefässe. Einige haben eine spezifisch blutdrucksenkende Wirkung.
Die erfindungsgemäss hergestellten Verbindungen sind überdies wertvolle Zwischenprodukte für die Herstellung von Medikamenten.
Die als Ausgangsmaterialien verwendeten Hy- drazide der Lysergsäure- und Dihydrolysergsäure- Reihe können z. B. hergestellt werden, indem man in 1-Stellung substituierte Derivate der natürlichen Mutterkomalkaloide oder ihrer Dihydro-Derivate nach bekannten Methoden mit wasserfreiem Hydrazin bei erhöhter Temperatur spaltet.
In den nachfolgenden Beispielen erfolgen alle Temperaturangaben in Celsiusgraden. Die Schmelz punkte sind unkorrigiert. <I>Beispiel 1</I> 1,6-Dimethyl-ergolyl-(8)-carbamidsäure-anilid Man versetzt eine Lösung von 4,235 g 1-Me- thyl-9,10-dihydro-D-lysergsäure-hydrazid in 145 cm 0,1n Salzsäure mit 14,5 cm, In Natriumnitritlösung und lässt unter Rühren bei einer Temperatur von 2 bis 5 20 cm3 In Salzsäure zutropfen. Man entfernt anschliessend das Eisbad,
rührt weitere 15 Minuten und schüttelt hierauf rasch zwischen eiskalter Na- triumcarbonatlösung und Benzol aus. Die über Ka- liumcarbonat getrocknete benzolische Lösung erhitzt man möglichst rasch zum Sieden und hält sie wäh rend 5 Minuten bei dieser Temperatur.
Anschlie ssend versetzt man die Lösung mit 5 g Anilin, er hitzt weitere 5 Minuten und versetzt die auf ein kleines Volumen eingeengte Lösung mit 500 cm3 Petroläther, wobei sich reines 1,6-Dimethyl-ergolyl- (8)-carbamidsäure-anilid ausscheidet, das abfiltriert wird. Die Verbindung wird nach Umkristallisieren aus Benzol und Petroläther als mikrokristalline, gallertartige Substanz erhalten.
Smp.: Keine Schmelze bis 300 , ab etwa 250 Zersetzung. [a]D = -80 (c - 0,5 in Pyridin). Kellersche Farbreaktion: blau. Das als Ausgangsmaterial zur Verwendung ge langende 1-Methyl - 9,10 - dihydro-D-lysergsäure-hy- drazid wird hergestellt, indem man 1-Methyl-dihydro- ergokryptin mit wasserfreiem Hydrazin erhitzt. Smp. 259-262 .
[a] D = -118 (c = 0,5 in Pyridin). Kellersche Farbreaktion: blau. <I>Beispiel 2</I> 1,6-Dimethyl-ergolyl-(8)-carbamidsäure- diäthylamid 4,235 g 1- Methyl - 9,10 - dihydro-D-lysergsäure- hydrazid werden wie in Beispiel 1 beschrieben in das Azid übergeführt und dessen benzolische Lösung möglichst rasch zum Sieden erhitzt und während 5 Minuten bei dieser Temperatur gehalten.
Anschlie ssend versetzt man die Lösung mit 5 cm3 Diäthyl- amin, erhitzt noch 5 Minuten zum Sieden und ver dampft die Lösung zur Trockne. Der Rückstand wird aus Benzol unter Zusatz von Petroläther umkristalli siert. Prismen. Smp. 142-143 .
[a]D = -60 (c = 0,5 in Pyridin). <I>Beispiel 3</I> 1,6-Dimethyl-ergolenyl-(8)-carbamidsäure- diäthylamid und 1,6-Dimethyl-isoergolenyl- (8)-carbamidsäure-diäthylamid Eine Lösung von 1,8 g 1-Methyl-D-lysergsäure- hydrazid in 61,5 cm3 0,1n Salzsäure versetzt man mit 6,15 cm3 In Natriumnitritlösung, kühlt auf 0-5 ab und lässt bei dieser Temperatur unter Rühren 8,
5 cm3 In Salzsäure zutropfen. Nach 15 Minuten macht man das Reaktionsgemisch mit Na- triumhydrogencarbonat alkalisch und schüttelt rasch mit Benzol aus. Die über Kahumcarbonat scharf ge trocknete benzolische Lösung erhitzt man möglichst rasch zum Sieden und hält sie während 5 Minuten bei dieser Temperatur. Anschliessend versetzt man die Lösung mit 2,5 g Diäthylamin. Man erhitzt einige Minuten weiter und verdampft sodann die Lösung zur Trockne.
Der Rückstand wird an einer Säule von 55g Aluminiumoxyd chromatographiert. Beim Entwickeln des Chromatogramms mit Chloro form wird zuerst das 1,6-Dimethyl-isoergolenyl-(8)- carbamidsäure-diäthylamid ins Filtrat gewaschen. Farblose Prismen aus Essigsäureäthylester vom Smp. 114-116 . [a120 = + 290c (c = 0,5 in Pyridin). Kellersche Farbreaktion: blau.
Das 1,6 - Dimethyl - ergolenyl-(8)-carbamidsäure- diäthylamid wird anschliessend mit Chloroform + 1/@ 3ö Athanol ins Filtrat gewaschen. Es bildet aus Methanol ein in Nadeln kristallisierendes Bimaleinat, das bei 176-179 schmilzt. [cc] D = +<B>70'</B> (c = 0,5 in Wasser). Kellersche Farbreaktion: blau.
Zu den gleichen zwei Verbindungen gelangt man auch, wenn man, anstatt 1-Methyl-D lysergsäure- hydrazid, das 1-Methyl-D-isolysergsäure-hydrazid als Ausgangssubstanz verwendet.
Die als Ausgangssubstanzen verwendeten Hy- drazide können hergestellt werden, indem man 1-Me- thyl-ergotamin-hydrochlorid mit Hydrazin auf etwa <B>901</B> erwärmt und nach Abdestillieren des Hydrazins das gebildete Gemisch aus 1-Methyl-D lysergsäure- hydrazid und 1-Methyl-D-isolysergsäure-hydrazid an Aluminiumoxyd chromatographiert. Das 1-Methyl- D-isolysergsäure-hydrazid wird mit
Chloroform. + Äthanol ins Filtrat gewaschen und kristallisiert aus Äthanol in Blättchen vom Smp. 201-204 . [a]20 = + 440 (e = 0,5 in Pyridin).
Das 1-Methyl-D-lysergsäure-hydrazid wird mit Chloroform + 2 ö Äthanol ins Filtrat gewaschen und kristallisiert aus Äthanol in Prismen vom Sm_p. 194 bis<B>195'.</B>
[ü] D =<B>125'</B> (c = 0,5 in Chloroform). Beispiel <I>4</I> 1,6-Dimethyl-ergolenyl (8)-carbamidsäure- (+)-butanolamid-(2') Man versetzt eine Lösung von 1,25 g 1-Methyl- D-lysergsäure-hydrazid in 42,5 cm3 1/1()n Salzsäure bei<B>51</B> mit 4,
5 cm-' 1n Natriumnitritlösung und tropft unter Rühren bei einer Temperatur von 5 bis <B>10'</B> 6 em3 1n Salzsäure hinzu, rührt noch 15 Mi nuten bei Raumtemperatur, alkalisiert dann unter Eiskühlung mit Natriumbicarbonat und schüttelt mit Benzol aus.
Die dreimal über Pottasche getrocknete Benzollösung wird so rasch als möglich zum Sieden erhitzt und 5 Minuten bei Siedetemperatur gehalten, und hierauf werden; 2 cm3 (+) Butanolamin-(2) hinzugefügt.
Nach weiteren 5 Minuten wird am Vakuum auf etwa 50 cm3 eingeengt und mit 200 cm3 Petroläther versetzt. Man filtriert die ausgeschiedene flockige Fällung ab und kristallisiert aus einem Ge misch von Chloroform/Methanol, dem man vor sichtig Äther zufügt, in flache farblose Nadeln. Smp. 227 .
[a] D = + 211 (c = 0,5 in Pyridin). Kellersche Farbreaktion: blau. Das Bitartrat kristallisiert in Nädelchen vom Smp. 191-192 .

Das als Ausgangsmaterial verwendete 1 Methyl- D lysergsäure-hydrazid kann z. B. hergestellt werden durch Erhitzen eines 1-Methyl-D-lysergsäure-amids, z.
B. von 1-Methyl-ergotamin, mit Hydrazin in Gegenwart von Protonen, und Chromatographie des dabei sich bildenden Gemisches von 1-Methyl-D- lysergsäure-hydrazid und 1-Methyl-D-isolysergsäure- hydrazid. Prismen aus Methanol, Smp. 194-195 .

und das sich spontan gallertig ausscheidende Butanol- amid wird abgenutscht und aus einem Gemisch von Chloroform/Methanol unter Ätherzusatz kristalli siert. Uncharakteristisches Mikrokristallisat vom Smp. > 350 (Zersetzung).

Das als Ausgangsmaterial verwendete 1-Methyl- 9,10-dihydro-D-lysergsäure-hydrazid kann z. B. her gestellt werden durch Erhitzen von 1-Methyl-9,10- dihydro-ergocristin mit Hydrazin, worauf sich beim Erkalten das 1-Methyl-9,10-dihydro-D-lysergsäure- hydrazid in Spiessen vom Smp. 259-262 abscheidet. Beisviel <I>6</I> 1 Allyl-6-methyl-ergolenyl (8)-carbamidsäure- diäthylamid und 1-Allyl-6-methyl-isoergolenyl-(8)-carbamidsäure- diäthylamid 3,25 g 1-Allyl-D-isolysergsäure-hydrazid (Smp. 129-130 ) werden wie in Beispiel 3 beschrieben in das Azid übergeführt und dessen benzolische Lösung möglichst rasch zum Sieden erhitzt und während 5 Minuten bei dieser Temperatur gehalten.
Anschlie ssend versetzt man die Lösung mit etwa 10 cm3 Di- äthylamin, lässt noch 30 Minuten bei Raumtempera tur stehen und arbeitet das erhaltene Isomeren- gemisch wie in Beispiel 3 auf.

Das als Ausgangsmaterial benötigte 1-Allyl-D- isolysergsäure-hydrazid kann z. B. wie folgt her gestellt werden: D-Lysergsäure wird in flüssigem Ammoniak in Gegenwart von tert. Natriumbutylat mit Allylbromid alkyliert, die so gewonnene 1-Allyl- D-lysergsäure (Smp. 209-211 ) mit Diazomethag verestert,
der rohe Methylester mit Propanol und Hydrazin in Gegenwart von 1 Mol Hydrazinchlor-- hydrat auf 40-90 erwärmt und das erhaltene Ge misch von 1-Allyl - D - lysergsäure-hydrazid und 1 Allyl-D-isolysergsäure-hydrazid chromatographisch in seine Komponenten zerlegt. Letztere Verbindung kristallisiert aus Alkohol/Äther. Smp. 129-130 .
[a] D = + 399 (c = 0,5 in Pyridin). <I>Beispiel 7</I> 1,6-Dimethyl-ergolenyl-(8)-carbamidsäure- amid und 1,6-Dimethyl-isoergolenyl-(8)-carbamidsäure- amid 5 g 1-Methyl=D-lysergsäure-hydrazid werden wie in Beispiel 1 beschrieben in das Azid übergeführt, dessen benzolische Lösung rasch zum Sieden erhitzt und 5 Minuten lang am Sieden gehalten wird.
An schliessend leitet man bei Raumtemperatur während 30 Minuten gasförmigen Ammoniak durch die Lö sung, verdampft zur Trockne und zerlegt das zu ungefähr gleichen Teilen aus 1,6-Dimethyl-ergolenyl- (8)-carbamidsäure-amid und 1,6-Dimethyl-isoergo- lenyl-(8)-carbamidsäure-amid bestehende Rohprodukt durch fraktionierte Kristallisation der Bimaleinate in die Einzelkomponenten:
Man versetzt eine Lösung des Rohproduktes in Äther/Methanol zuerst mit 1/2 Mol Maleinsäure, wo bei sich schon ziemlich reines 1,6-Dimethyl-isoergo- lenyl - (8) - carbamidsäure - amid-bimaleinat kristallin abscheidet, das man durch Umkristallisation aus Methanol/Äther von beigemengtem 1,6-Dimethyl- ergolenyl-(8)-carbamidsäure-amid-bimaleinat abtrennt.
Die reine Verbindung bildet Prismen aus Methanol vom Smp. 215-218'.

Aus der Mutterlauge der vorstehend beschrie benen Isoverbindung setzt man die Basen wieder frei und versetzt sie dann in methanolischer Lösung mit 1 Mol Maleinsäure, wobei fast reines 1,6-Di- methyl-ergolenyl - (8) -carbamidsäure-amid-bimaleinat auskristallisiert, das man durch Kristallisation aus demselben Lösungsmittel von wenig beigemengter Isoverbindung abtrennt.
Die reine Verbindung bildet feine Nadeln aus Methanol/Äther, die oberhalb etwa 110 langsam zusammensintern. [a] D = + 61 (c = 0,5 in Wasser). <I>Beispiel 8</I> 1,6-Dimethyl-ergolenyl-(8)-carbamidsäure- äthylamid 10 g 1-Methyl-D-lysergsäure-hydrazid werden wie in Beispiel 1 beschrieben in das Azid übergeführt, dessen benzolische Lösung rasch zum Sieden erhitzt und 5 Minuten bei dieser Temperatur gehalten wird.
Anschliessend versetzt man die Lösung mit 10 em3 Athylamin und verdampft das Gemisch nach 1stün digem Stehen bei Raumtemperatur zur Trockne. Der Rückstand wird aus Benzol/Methanol umkristalli siert. Nadeln vom Smp. 250-252 .
Bimaleinat: Nadeln aus Methanol. Add 5 cm3 in hydrochloric acid. After 15 minutes, the reaction mixture is made alkaline with sodium hydrogen carbonate and quickly extracted with benzene. The benzene solution, which has been sharply dried over potassium carbonate, is heated to the boil as quickly as possible and kept at this temperature for 5 minutes. The solution is then mixed with 2.5 g of diethylamine. Heating is continued for a few minutes and the solution is then evaporated to dryness.
The residue is chromatographed on a column of 55 g of aluminum oxide. When developing the chromatogram with chloro form, the 1,6-dimethyl-isoergolenyl- (8) -carbamic acid diethylamide is first washed into the filtrate. Colorless prisms made from ethyl acetate with a melting point of 114-116. [a120 = + 290c (c = 0.5 in pyridine). Keller's color reaction: blue.
The 1,6-dimethyl-ergolenyl- (8) -carbamidsäure- diethylamide is then washed into the filtrate with chloroform + 1 / @ 3ö ethanol. From methanol it forms a bimaleate which crystallizes in needles and melts at 176-179. [cc] D = + 70 '(c = 0.5 in water). Keller's color reaction: blue.
The same two compounds are obtained if, instead of 1-methyl-D-lysergic acid hydrazide, 1-methyl-D-isolysergic acid hydrazide is used as the starting substance.
The hydrazides used as starting substances can be prepared by heating 1-methylergotamine hydrochloride with hydrazine to about 901 and, after the hydrazine has been distilled off, the mixture formed of 1-methyl-dysergic acid - hydrazide and 1-methyl-D-isolysergic acid hydrazide chromatographed on aluminum oxide. The 1-methyl-D-isolysergic acid hydrazide is with
Chloroform. + Ethanol washed into the filtrate and crystallized from ethanol in leaflets with a melting point of 201-204. [a] 20 = + 440 (e = 0.5 in pyridine).
The 1-methyl-D-lysergic acid hydrazide is washed into the filtrate with chloroform + 2 ö ethanol and crystallizes from ethanol in prisms of Sm_p. 194 to <B> 195 '. </B>

5 cm- '1N sodium nitrite solution and dropwise with stirring at a temperature of 5 to <B> 10' </B> 6 em3 1N hydrochloric acid, stir for another 15 minutes at room temperature, then alkalize with sodium bicarbonate while cooling with ice and shake out with benzene .
The benzene solution, dried three times over potash, is heated to boiling as quickly as possible and kept at the boiling temperature for 5 minutes, and then; 2 cm3 (+) butanolamine- (2) added.
After a further 5 minutes, the mixture is concentrated to about 50 cm3 in vacuo and 200 cm3 of petroleum ether are added. The flaky precipitate which has separated out is filtered off and crystallized from a mixture of chloroform / methanol, to which ether is carefully added, into flat, colorless needles. M.p. 227.

The 1 methyl D lysergic acid hydrazide used as starting material can, for. B. be prepared by heating a 1-methyl-D-lysergic acid amide, e.g.
B. of 1-methyl-ergotamine, with hydrazine in the presence of protons, and chromatography of the resulting mixture of 1-methyl-D-lysergic acid hydrazide and 1-methyl-D-isolysergic acid hydrazide. Methanol prisms, m.p. 194-195.

B. be made by heating 1-methyl-9,10-dihydro-ergocristine with hydrazine, whereupon the 1-methyl-9,10-dihydro-D-lysergic acid hydrazide in skewers of melting point 259-262 on cooling separates. [a] D = -118 '(c = 0.5 in pyridine).
As much <I> 6 </I> 1 allyl-6-methyl-ergolenyl (8) -carbamic acid diethylamide and 1-allyl-6-methyl-isoergolenyl- (8) -carbamic acid diethylamide 3.25 g 1-allyl D-isolysergic acid hydrazide (melting point 129-130) are converted into the azide as described in Example 3 and its benzene solution is heated to boiling as quickly as possible and kept at this temperature for 5 minutes.
The solution is then mixed with about 10 cm 3 of diethylamine, left to stand for a further 30 minutes at room temperature and the isomer mixture obtained is worked up as in Example 3.
Bimaleate of 1-allyl-6-methyl-ergolenyl- (8) -carbamic acid diethylamide: prisms of m.p. 176-177.

Sodium butylate alkylated with allyl bromide, the 1-allyl-D-lysergic acid obtained in this way (melting point 209-211) is esterified with diazomethag,
the crude methyl ester with propanol and hydrazine in the presence of 1 mol of hydrazine chlorohydrate is heated to 40-90 and the resulting mixture of 1-allyl-D-lysergic acid hydrazide and 1 allyl-D-isolysergic acid hydrazide is broken down into its components by chromatography . The latter compound crystallizes from alcohol / ether. M.p. 129-130.
[a] D = + 399 (c = 0.5 in pyridine). <I> Example 7 </I> 1,6-Dimethyl-ergolenyl- (8) -carbamic acid amide and 1,6-dimethyl-isoergolenyl- (8) -carbamic acid amide 5 g 1-methyl = D-lysergic acid- hydrazide are converted into the azide as described in Example 1, the benzene solution of which is rapidly heated to the boil and kept boiling for 5 minutes.
Subsequently, gaseous ammonia is passed through the solution at room temperature for 30 minutes, evaporated to dryness and split into approximately equal parts from 1,6-dimethyl-ergolenyl- (8) -carbamic acid amide and 1,6-dimethyl isoergo - lenyl- (8) -carbamic acid amide existing crude product through fractional crystallization of the bimaleinates into the individual components:
A solution of the crude product in ether / methanol is first mixed with 1/2 mole of maleic acid, where already fairly pure 1,6-dimethyl-isoergolenyl- (8) -carbamic acid-amide-bimaleate precipitates in crystalline form, which is precipitated by recrystallization separated from methanol / ether of added 1,6-dimethylergolenyl- (8) -carbamic acid amide bimaleate.
The pure compound forms prisms from methanol with a melting point of 215-218 '.
[a120 = + 222 (c = 0.5 in water).
The bases are released again from the mother liquor of the isocompound described above and then 1 mol of maleic acid is added to them in methanolic solution, with almost pure 1,6-dimethylergolenyl- (8) -carbamic acid amide-bimaleate crystallizing out, which is separated from a little added iso-compound by crystallization from the same solvent.
The pure compound forms fine needles made of methanol / ether, which slowly sinter together above about 110. [a] D = + 61 (c = 0.5 in water). <I> Example 8 </I> 1,6-Dimethyl-ergolenyl- (8) -carbamic acid ethylamide 10 g of 1-methyl-D-lysergic acid hydrazide are converted into the azide as described in Example 1, and its benzene solution is rapidly converted heated to boiling and held at this temperature for 5 minutes.
The solution is then mixed with 10 em3 ethylamine and the mixture is evaporated to dryness after standing for 1 minute at room temperature. The residue is recrystallized from benzene / methanol. Needles of m.p. 250-252.
Bimaleate: needles made from methanol. Mp. 177 to <B> 179 '. </B> [a] 220 + 55 (c = 0.5 in water). <I> Example 9 </I> 1,6-Dixethyl-ergolyl- (8) -carbamic acid amide 2.96 g of 1-methyl-9,10-dihydro-D-lysergic acid hydrazide are converted into the hydrazide as in Example 1 Azide passed over, the benzene solution of which is rapidly heated to boiling and then left to cool in a stream of ammonia.
The amide which has crystallized out is recrystallized from a mixture of methanol / chloroform / petroleum: ether. Finest colorless needles with a melting point of over 350. [a] D = -86 (c-0.25 in pyridine).
