Abstract

Oxidised steroids are prepared by the action of mycelium forming microorganisms with oxidisable steroids in which an aqueous liquid containing oxygen and the steroid to be oxidised are pushed through a stationary mass of the mycelium. It is preferable to use a static layer of the microorganisms in pellet rather than fibrous form, and the aqueous liquid may be the culture medium for the microorganisms. It is possible to use a continuous circulation process in which a part of the aqueous liquid is separated from the mycelium while circulation is continued, the volume of liquid separated being replaced by further liquid, e.g. fresh nutrient culture medium, pure water or physiological saline, and the further liquid may contain additional oxidisable steroid, e.g. of the pregnane series which may be unsubstituted in the 11-position. The process is particularly suitable for the introduction of an oxygen function at the 11-position such as an 11a -hydroxy group by means of Rhizopur nigricans or an 11b -hydroxy group by means of Cunninghametta blakesleeana. The introduction of oxygen into the system may be used to circulate the liquid. In the examples 11a -hydroxy-progesterone and hydrocortisone are prepared from the corresponding 11-desoxy compounds.
