Abstract

A test composition in dry solid form for the detection of peroxidase activity due to occult blood in body fluids comprises an organic hydroperoxide and an indicator material which changes colour in the presence of said organic hydroperoxide and blood. The organic hydroperoxide and the indicator material may be impregnated into a bibulous carrier, for example, a filter-paper strip, or may be in the form of tablets. The organic hydroperoxide is preferably first encapsulated within microspherical bubbles of a colloidal substance such as polyvinyl alcohol, gelatin, gum arabic, or carboxy vinyl polymer. Specified organic hydroperoxides are ethyl hydroperoxide, 1-cyclohexyl hydroperoxide-1, cumene hydroperoxide, diisopropyl benzene hydroperoxide, para-menthane hydroperoxide, 2,5-dimethyl hexane-2.5-dihydroperoxide, and titanium potassium oxalate hydroperoxide. Specified indicators are aniline, o-tolidine, o-toluidine, p-toluidine, o-phenylenediamine, N, N1-dimethylp-phenylenediamine, N, N1-diethyl-p-phenylenediamine, benzidine, di-anisidine, o-cresol, m-cresol, p-cresol, a - and b -naphthols, catechol, guaicol, and pyrogallol. The composition is preferably buffered to a pH of 4 to 7, e.g. with citrates, tartrates, phosphates, phthalates, or acetates. In the case of acetates, the buffer may comprise calcium, sodium, potassium, or lithium acetate and an acid, such as citric, fumaric, itaconic, maleic, malic, malonic, or mandelic acid, which on hydrolysis will react with the acetate to form acetic acid. The compositions may also contain stabilizers such as gelatin, algin, carogein, casein, or albumin. Test tablets may comprise excipients, such as lactose, starch, or flour, to facilitate the formation of granules. A background dye may be included so that its colour contrasts strongly with the indicator colour. Examples of such dyes are D and C Red No. 35, D and C Yellow No. 11, and D and C Orange No. 15.
