Abstract

The invention comprises aroylalkyl piperidinols of the formula <FORM:0881893/IV (b)/1> and acid addition salts and non-toxic quaternary salts thereof wherein Ar is a halophenyl (e.g. fluoro-, chloro-, bromo-oriodo-phenyl), methoxy-, ethoxy- or dimethoxy-phenyl or thienyl radical or a monocyclic aromatic hydrocarbon radical of less than 9 carbon atoms, Ar1 is a halophenyl radical or a monocyclic aromatic hydrocarbon radical of less than 9 carbon atoms (e.g. phenyl, tolyl or xylyl), and Alk is a lower alkylene radical of 3-6 carbon atoms, e.g. propyl, and their preparation by reacting a compound of the formula Ar-CO-Alk-Halogen with a 4-Ar1-piperidin-4-ol. The compounds are used pharmaceutically, suitably in the form of salts such as the hydrohalides, sulphates, phosphates, sulphamates, citrates, lactates, maleates, malates, succinates, tartrates, cinnamates, acetates, benzoates, gluconates and ascorbates. They also form quaternary salts with quaternating agents such as alkyl, aralkyl, alkenyl and hydroxyalkyl halides, sulphates and aryl-sulphonates. Aryl haloalkyl ketones of the formula Ar CO.Alk.Halogen used as starting materials are made either by treating benzene or a substituted benzene, with a haloalkanoyl halide, by the Friedel-Crafts reaction, using aluminium chloride or a milder catalyst, such as stannic chloride, or by reacting an aryl magnesium bromide Grignard reagent with a haloalkanonitrile, followed by acid hydrolysis of the adduct. 4-Aryl piperidin-4-ols, in which the aryl group may be substituted according to the definition of the group Ar1 above, are made by treating a 4-aryl-1:2:3:6-tetrahydropyridine, obtainable by condensing an a -methylstyrene with ammonium chloride and formaldehyde, with hydrogen bromide to give a 4-bromo-4-aryl-piperidine which is hydrolysed with aqueous sodium hydroxide. a -Methylstyrenes of the formula Ar1C(CH3) : CH2, such as p-fluoro-a -methylstyrene, are made by treating the corresponding acetophenone with methyl magnesium bromide and dehydrating the product or by treating the Grignard reagent derived from an aryl halide; e.g. p-chlorophenyl magnesium bromide with acetone and dehydrating the resulting carbinol. The Provinional Specification relates to compounds of the above general formula wherein Ar is a phenyl, alkylphenyl, halophenyl, alkoxyphenol or thienyl radical, Alk is a 1:3 propylene radical and Ar1 is a phenyl, halophenyl or alkyphenyl radical.
