Abstract
3-Keto-D 1,4-steroids are prepared by dehydrogenation of 3-keto-D 4-steroids having from 18 to 21 atoms in the carbon skeleton by contact with oxidizing enzymes produced by an organism of the genus Nocardia or the genus Corynebacterium in the presence of a steroid production-enhancing amount of at least one antibacterial antibiotic. Suitable antibiotics include tetracyclines, neomycin and streptomycins, and salts thereof, used in amounts varying from 0.125 mcg. to 200 mcg. per milliliter of reaction mixture, although others such as penicillin, tyrothricin, polymixin, chloramphenicol, erythromycin, carbomycin, bacitracin, viomycin, gramicidin, novobiocin, dextromycin and oleandomycin may also be used. Examples describe the preparation of prednisolone and the A1-dehydrogenation of Compound S, 9a -fluoro-hydrocortisone, D 4:6-androstadiene-3;17-dione, 11b :17a -dihydroxyprogesterone, D 14-Compound S, androst-4-ene-3:17-dione, 19-nor-testosterone, 17a -hydroxyprogesterone, 16-dehydroprogesterone, 11-ketoprogesterone, D 9(11)-dehydro-Compound S, 14a :15a - epoxy - Compound S, 14a - hydroxyhydrocortisone and 14a -hydroxy-cortisone. Other starting materials referred to are cortisone, corticosterone, 11-epi-hydrocortisone, 14a - hydroxy - Compound S, deoxycorticosterone, testosterone and progesterone. The preparation of hydrocortisone by 11b -hydroxylation of Compound S with Curvularia lurata is also described. Specifications 769,998, 770,000, 789,363 and 829,260 are referred to.
