Abstract

The invention comprises saturated and unsaturated 20-hydroxy pregnanes and allopregnanes which may be nuclearly substituted and contain in the 20-position a radical of formula: <FORM:0869007/IV (b)/1> in which R1 represents hydrogen or an alkyl group containing one or two carbon atoms, and R2 represents a pyridyl or piperidyl residue bound in the 2-position to the carbon atom shown in the above formula, their esters, ketals, N-lower alkyl and N-lower acyl derivatives and salts thereof. The compounds are prepared by reacting a 20-oxo-allopregnane or pregnane with a 1-pyridyl-(21)-alkyl lithium compound, and if necessary reducing the resulting pyridyl compound to the piperidyl analogue. Examples detail the preparation of D 5-3b ,20-dihydroxy-20-methyl-21-pyridyl-(21) -pregnene, 3b ,20-dihydroxy-20-methyl-21-piperidyl-(21)-allopregnane their esters and salts, N-methyl-3b -acetoxy-20-hydroxy- 20- methyl- 21- piperidyl- (21)- allopregnane and salts thereof, together with the corresponding 20,21 -dimethyl analogues of the foregoing compounds.ALSO:Pharmaceutical compositions having a coronary dilatory action comprise saturated or unsaturated 20-hydroxypregnanes or allopregnanes containing in position 20 the grouping -CHR1R2, in which R1 is hydrogen or an alkyl group of one or two carbon atoms, and R2 is a pyridyl or piperidyl residue bound in the 2-position to the carbon atom shown in the above formula, their esters, ketals, N-lower alkyl or N-lower acyl derivatives or salts thereof, together with a pharmaceutical carrier suitable for enteral or parenteral administration. The preparations may be in the form of tablets, dragees, solution, suspensions or emulsions and may contain auxiliaries such as preserving, stabilizing, wetting, emulsifying and buffering agents or other therapeutic substances.
