Abstract
Bacitracin is purified by passing an aqueous solution of bacitracin having a pH of 2 to 10 through a strongly acidic cation exchange resin and a weakly basic anion exchange resin, the bacitracin being neither absorbed on said resins nor eluted therefrom, and drying the resulting solution to obtain solid bacitracin. A fermentation broth is clarified and the pH adjusted first to pH 3,5 and then to pH 7,5 to precipitate impurities. The exchange resins are in the free acid and free base form. The aqueous solution of bacitracin may be passed down a column of exchange resin in any order or simultaneously i.e. a column of mixed resins may be used. The cation exchange resins may be those which contain nuclear sulphonic acid groups or methylene sulphonic acid groups e.g. phenol formaldehyde resins containing nuclear sulphonic acid groups or sulphonated aromatic hydrocarbon polymers such as sulphonated polystyrene resins e.g. " Amberlite " (Registered Trade Mark) 1R-120. The anion exchange resins are weak amine or mixed amine type phenol formaldehyde resins e.g. Amberlite 1R-4B and Amberlite 1R-45. Mixed resins are Amberlite MB-4, a mixture of Amberlite 1R-120 and Amberlite 1R-45.
