Abstract
Glutamine is obtained by heating a solution of the corresponding optical form of g -glutamyl hydrazide in a solvent containing at least 65 per cent by volume of water in the presence of Raney nickel at a pH greater than 9. The process may be carried out between 50 DEG C. and reflux temperature for 1-3 hours, especially at 65-75 DEG C. Preferably 1-4 parts of catalyst are used per part of glutamyl hydrazide. After the reaction the catalyst is filtered off, any nickel in solution is precipitated with dimethylglyoxime or hydrogen sulphide and the glutamine crystallized out. Examples are given of the production of L-glutamine using as solvents water, aqueous ammonia, methanol, ethanol, isopropanol and dioxane. Other specified solvents are water and butanols, pentanol, tetrahydrofurfuryl alcohol and dimethylformamide. The starting material is made by esterifying glutamic acid at low temperatures (with a sulphuric acid catalyst) by means of an alcohol or phenol to give e.g. the methyl, ethyl, propyl or butyl ester and treating this with hydrazine in aqueous or aqueous-alcoholic solution. Examples are given of these steps.
