Abstract

The invention comprises 2-methyl-4-amino-5-acetaminomethyl-pyrimidine and the preparation thereof by reacting a propionitrile of the general formula <FORM:0772256/IV(b)/1> wherein R represents the same or different alkyl radical or aralkyl radical such as the benzyl or phenethyl radical, with acetamidine or with acetimidoether and ammonia, and the resulting 2,7 - dimethyl - 5,6 - dihydropyrinido-(4,5-d) pyrimidine of the formula <FORM:0772256/IV(b)/2> is hydrolysed under weak hydrolysis conditions, e.g. by boiling with water or heating in a dilute aqueous alkaline or acidic solution. The reaction is preferably carried out in the presence of an inert solvent for both reactants, e.g. a lower aliphatic alcohol such as methanol, ethanol or propanol, or a mixture thereof with benzene, toluene, xylene, ether or petroleum ether. An alkaline condensing agent may also be used in the reaction, e.g. an alkali or alkaline earth metal hydroxide or carbonate. In the examples, 2 - methyl - 4 - amino - 5 - acetaminomethyl-pyrimidine is prepared by the above process using a -diethoxy-methyl-b -ethoxypropionitrile and the corresponding a -dibenzyloxy-methyl compound as reactants.
