Abstract
The invention comprises 21-substituted polycarboxylic acid esters of hydrocortisone, the metal salts thereof and the salts thereof with ethylene diamine derivatives having the structure R1(CH2)xNHCH2CH2NH(CH2)y R2 wherein R1 and R2 are aryl groups and x and y are 1, 2 or 3, and a process for the preparation of a hydrocortisone acid ester or salt thereof which comprises contacting hydrocortisone with a polycarboxylic acid acylating agent such as an acid anhydride, acid halide or the acid itself and if desired, converting the 21-substituted acid ester of hydrocortisone thus produced, to a salt. As esterification agents there may be used maleic anhydride, glutaric anhydride, phthalic anhydride, phthaloyl chloride, maleic acid, trimethyl citrate, tartaric acid, succinic anhydride and pyromellitic acid. The esterification is catalysed by ion-exchange resins and minor amounts of strong acids and alkalis. Examples disclose the production of hydrocortisone phthalate, succinate, maleate, sodium hydrocortisone succinate, and the dibenzylethylenediamine salt of hydrocortisone succinate.ALSO:Sodium hydrocortisone succinate is obtained by adding hydrocortisone succinate to water containing sodium bicarbonate and the product isolated by freeze-drying. An aqueous solution of the sodium salt is suitable for injection. Saline may be employed to form an isotonic solution.
