Abstract
Antibiotic 106-7 is purified by contacting a solution thereof having a pH of 5 to 12 with a strongly basic quaternary ammonium anion-exchange resin deriving its capacity from quaternary amine groups on the hydroxyl cycle to adsorb said antibiotic and eluting the adsorbed antibiotic with a solution containing anions, the antibiotic being recovered from the eluate. Examples of suitable resins are "Amberlites" (Registered Trade Mark) IRA400, IRA140, XE98 and XE58, "Permutit" (Registered Trade Mark)-S "Ionac" (Registered Trade Mark), A-293 and Dowex 2. They are converted to the hydroxyl cycle by washing with an alkaline solution such as aqueous sodium hydroxide followed by a water wash. Elution is by means of (a) solutions of organic or inorganic acids, e.g. aqueous solutions of acetic, propionic, carbonic, hydrochloric and sulphuric acids; (b) basic solutions, e.g. aqueous quaternary ammonium hydroxides, aqueous sodium hydroxide, potassium hydroxide and ammonium hydroxide; and (c) aqueous solutions of salts such as sodium chloride, sodium sulphate, potassium chloride and calcium chloride. The antibiotic is recovered from eluate fractions of pH 7 to 4.5 in the case of acid solutions, and pH 7-14 in the case of basic solutions. The resin containing the adsorbed antibiotic 106-7 may be washed and dried directly to yield an antibiotic-resin-complex useful as a therapeutic agent or animal feed supplement. Specifications 757,089, 757,090 and 758,500 are referred to.ALSO:As an animal feed supplement is used a complex of antibiotic 106-7 and a strongly basic quaternary ammonium type of anion exchange resin deriving its capacity from quaternary amine groups present in the polymer and prepared by mixing the resin with a solution of the antibiotic, or by passing a solution of the antibiotic down a column of the resin, separating the resin-complex, washing and drying (see Group IV (b)). Examples of the resins are "Amberlites" (Registered Trade Mark) IRA-400, IRA-410, XE-98, XE-75, XE-58, "Permutit" (Registered Trade Mark) S, "Ionac" (Registered Trade Mark) A-293 and Dowex 2. Specifications 757,089, 757,090 and 758,500, [all in Group IV (b)], are referred to.
