An apparatus for use in preparing pure cultures of living micro organisms at the place of application, comprises a bottle for receiving the culture medium, a thin-walled, domed, flexible elastic stopper of rubber or other elastomer closing said bottle, and an elongated container for the micro-organisms in their stationary state, the elongated container passing in air-tight manner through said stopper and dipping into said culture medium, the elongated container having within the bottle a portion which can readily be ruptured without the formation of splinters, the stopper being sufficiently flexible to allow of manipulating the container from outside the bottle so as to rupture the said readily rupturable portion and thus place the inside of the container in communication with the culture medium. As shown, a U-tube a for bacteria in their stationary state is passed through a rubber stopper b so that its U-shaped lower half projects into a flask c filled with culture liquid g, the lower part of the U -tube a being scratched at d to facilitate opening when the legs e, which project through the stopper b, are pressed together at the time of preparation of the fresh culture. The flask also has a closure cap f. To facilitate withdrawal of the prepared culture the stopper b may have a lateral outlet h which can be sealed at i after the culture medium has been sterilized. The stopper may be provided with a filter r. In use, the culture medium is filled into the flask e, which is closed by the rubber stopper provided with U-tube a and with outlet tube h open. After sterilization the outlet tube h is sealed at i. One leg of the U-tube is opened under sterile conditions and the bacteria introduced. The tube is evacuated or filled with nitrogen and the opened leg sealed. In modifications, the container for the bacteria in a stationary state is (a) a glass tube slidably held in a rubber stopper having a cap n adapted to be punctured when the tube is thrust onto a spike o on the bottom of the flask (Fig. 2, not shown), and (b) a glass tube having its lower end bent at an obtuse angle (Fig. 3, not shown).
