Abstract
The invention comprises 2 : 17-dimethyl-3-hydroxymorphinane of the formula <FORM:0731787/IV (b)/1> and its acid addition salts, particularly the hydrochloride. The compound is made by treating 3 - hydroxy - 17 - methylmorphinane with formaldehyde (or compound yielding it) and a secondary amine in a Mannich reaction, and catalytically hydrogenating the resulting 2 - (N : N - disubstituted - aminomethyl) - 3 - hydroxy - 17 - methylmorphinane. The secondary amine is preferably aliphatic such as dimethylamine, diethylamine, dipropylamine or dibutylamine. In the examples (1) 3-hydroxy-17-methylmorphinane is heated with paraformaldehyde and diethylamine in ethanol and the 2-diethylaminomethyl compound isolated as hydrobromide; the free base of the latter is hydrogenated at elevated temperature and pressure in the presence of palladium charcoal and the 2 : 17-dimethyl-3-hydroxymorphinane converted into its hydrochloride; (2) as in (1) using dibutylamine; (3) as in (1) using the optically active (-)-form of the starting material, the activity being maintained in the final product. Salts may also be formed with hydrobromic, sulphuric, phosphoric and tartaric acids.
