Abstract

3 - Pyrrolidino - and 3 - piperidino - 1 - cyclohexyl-1-phenylpropan-1-ol are manufactured by catalytic hydrogenation of the corresponding 1 : 1-diphenyl compounds, the reaction being stopped when a small excess over 3 molecular proportions of hydrogen are absorbed. The products are useful pharmaceutically, particularly in the form of their salts with acid which are non-toxic in the dosages employed. In examples: (1) 3-piperidino-1 : 1-diphenylpropan-1-ol dissolved in glacial acetic acid is shaken with hydrogen in the presence of platinum oxide; (2) the acetic acid in (1) is diluted with water; (3) aqueous ethanolic hydrochloric acid is used as solvent and the hydrogenation effected at an initial hydrogen pressure of 29 atmospheres; (4) the hydrogenation in (1) is effected under an initial hydrogen pressure of 67 atmospheres; (5) 3-pyrrolidino - 1 : 1 - diphenylpropan - 1 - ol is hydrogenated as in (1). Specification 646,198 is referred to. The first Provisional Specification comprises additionally, with examples, the hydrogenation of 3-piperidino-1 : 1-diphenyl- or -1-phenyl-1-cyclohexylpropan-1-ol to 3-piperidino-1 : 1-dicyclohexylpropan-1-ol. The second Provisional Specification comprises generally the preparation of compounds of the general formula <FORM:0708805/IV (b)/1> wherein R represents a dimethylamino, diethylamino, methylethylamino, morpholino, pyrrolidino or piperidino group, and R1 represents hydrogen or an alkyl group of 1-3 carbon atoms, by partial hydrogenation of the corresponding 1 : 1-diphenyl compounds (obtainable by the process of Specification 624,118), additional examples describing the preparation of 3-dimethylamino-, 3-diethylamino- and 3-morpholino - 1 - cyclohexyl - 1 - phenylpropan - 1-ol, 3-piperidino- and 3-dimethylamino-1-cyclohexyl-1-phenylbutan-1-ol, and 3-piperidino- and 3 - dimethylamino - 1 - cyclohexyl - 1-phenylhexan-1-ol.
