Abstract

Alkyl esters of a -formylphenaceturic acid are prepared by reacting an alkyl ester of phenaceturic acid with an alkyl formate in the presence of an alkali metal alkoxide at a temperature not in excess of 100 DEG C. The reaction may take place in the presence of an inert solvent such as ether or alcohol and suitable alkyl formates are methyl-, ethyl-, propyl- or butylformates. In the examples, (1) ethyl formate and sodium ethoxide in ether are reacted with butylphenaceturic acid prepared by treating phenacetylchloride with glycine and potassium hydroxide and esterifying the phenaceturic acid formed with n-butanol; (2) ethyl a -formylphenaceturate is prepared from ethylphenaceturate and ethylformate and sodium- or potassium ethoxide in alcohol or ether. The esters of a -formylphenaceturic acid may be (a) condensed with cysteine, or its salts or esters to give esters of a -[2-(4-carboxy) thiazolindyl] phenaceturic acid and (b) reduced with aluminium amalgam and subjected to acid hydrolysis to give aminohydroxypropionic acid (serine).
