Abstract
Bis-nor - cholenic acid and acyl derivatives thereof are produced by the oxidation by means of ozone of crude mixtures of o-acyl derivatives of phytosterols such as the sterols of soya bean oil or calabar bean oil, the nuclear double bonds of the sterols being intermediately protected preferably by bromination. The preferred embodiment of the process comprises the acylation of the crude mixture of phytosterols by acetic acid followed by bromination and treatment with ozone. After oxidation the protective bromine is removed and the nuclear double bond re-established by treatment with zinc and acetic acid. In an example a 5 per cent ethereal solution of phytosterol acetate derived from soya bean oil is treated with bromine and then ozonized. The bromine is removed with zinc and acetic acid, water is added and the product taken up with ether. Acetic acid is removed by washing with water and the sodium salt of 3-acetoxy-bis-norcholenic acid is precipitated on the addition of caustic soda. The sodium salt is suspended in water, acidified, and extracted with ether. On removal of the ether and recrystallization from aqueous acetone pure 3-acetoxy-bis-norcholenic acid is obtained. Pure sitosterol acetate may be recovered from the first ether extract. Specification 453,773 is referred to.
