Abstract

Di- and tri-iodo derivatives of acylamino acids are prepared by reacting aromatic or heterocyclic carboxylic acid halides containing two or three iodine atoms in the nucleus with amino-carboxylic acids in the presence of alkalies or other bases. The products, usually in the form of their salts with alkalies or organic bases such as alkylamines and alkoxy-amines, possess therapeutic properties. Examples are given of the preparation of (1) 3 - 5- or 2 : 4-diiodohippuric acid from 3 : 5- or 2 : 4-diiodobenzoyl chloride and glycocoll; (2) 3 : 4 : 5-triiodohippuric acid from 3 : 4 : 5-triiodobenzoyl chloride and glycocoll; (3) 3 : 5-diiodobenzoyla -aminobutyric acid from 3 : 5-diiodobenzoyl chloride and a -aminobutyric acid; (4) 3 : 5-diiodobenzoyl - b - alanine from 3 : 5-diiodobenzoyl chloride and b -alanine; (5) 3 : 4 : 5 - triiodobenzoyl - a - aminobutyric acid and 3 : 4 : 5-triiodobenzoyl - sarcosine by treating 3 : 4 : 5-triiodobenzoyl chloride with a -aminobutyric acid and sarcosine respectively. Iodobenzoyl chlorides containing other substituents such as hydroxy, alkoxy and amino groups, may also be used, as well as iodo-substituted heterocyclic carboxylic acid halides such as diiodo-hydroxypyridine - carboxylic acid halides. As amino-carboxylic acids, aromatic or heterocyclic compounds too may be employed, e.g. aminobenzoic acid and tryptophane. Iodobenzoyl chlorides.--The acid chlorides of 2 : 4- and 3 : 5-diiodobenzoic acid and 3 : 4 : 5-triiodobenzoic acid are prepared by treating the acids with thionyl chloride.
