1450885 D-homosteroids F HOFFMANNLA ROCHE & CO AG 25 Sept 1974 [26 Sept 1973] 7642/76 Divided out of 1450884 Heading C2U The invention comprises 3,17a#-dihydroxy-D- homo-17aα-pregn-5-ene-21-carboxylic acid γ- lactone (Ia) and its 5,16-diene analogue (Ib). Compound Ia is prepared from 3#-acetoxy- 17aα - ethynyl - 17a# - hydroxy - D - homoandrost-5-ene by reaction of its Li salt with CO 2 to give 3#,17a#- dihydroxy - D - homo - 17aα- pregn - 5 - en - 20 - yne - 21 - carboxylic acid which by catalytic hydrogenation and acidification yields 3,17a# - dihydroxy - D - homo - 17aα- pregna - 5,20 - diene - 21 - carboxylic acid γ- lactone, catalytic hydrogenation of which yields Ia. Compound Ib is prepared from 3#-acetoxy-D- homoandrost - 5 - en - 17a - one by bromination, dehydrobromination and acetylation to give 3# - acetoxy - D - homoandrosta - 5,16 - dien- 17a-one which by Grignard reaction gives 3#- acetoxy - 17aα - (3,3 - ethylenedioxypropyl)- 17a# - hydroxy - D - homoandrosta - 5,16 - diene which by ketal hydrolysis, Jones oxidation, 3-saponification, and acidification yields Ib.
