1341921 Preparation of penicillins FABRICA DE ANTIBIOTICE IASI 4 Jan 1971 353/71 Heading C2A A process for the preparation of semisynthetic penicillins of Formulµ I and II: wherein in Formula I R and R 1 are methoxy groups or hydrogen, and R 2 is a nitro group or hydrogen, and in Formula II R is an alkyl, aralkyl or aryl group, by acylating 6-aminopenicillanic acid with the appropriate acid chloride in the case of penicillins of Formula I or with the appropriate mixed anhydride of an N-protected alpha-aminoacid in the case of penicillins of Formula II, wherein the acylation is carried out in solution in an organic solvent using the 6-aminopenicillanic acid in the form of its triethylammonium salt and the triethylammonium salt of the resulting penicillin is extracted with water from the organic solvent. The aqueous extract containing the triethylammonium salt may be acidified with mineral acid in the presence of dichloromethane and the resulting organic phase containing the penicillin in its acid form is separated from the aqueous phase. The separated organic phase containing the penicillin in its acid form may be treated with an aqueous solution of an alkali metal hydroxide and the aqueous phase containing the penicillin in the form of its alkali metal salt is separated from the organic phase. The alkali metal salt of the penicillin may be recovered by spray drying the aqueous solution thereof.
