Abstract
1,223,924. Purification of antibiotic 833A. MERCK & CO. Inc. 12 Sept., 1968 [22 Jan., 1968], No. 43438/68. Heading C2A. Antibiotic 833A, i.e. (-) (cis-1,2-epoxypropyl)- phosphonic acid, obtained according to Specification 1,223,923, is purified by stirring a solution of the antibiotic containing impurities with acid-washed alumina, filtering the resulting mixture, eluting the filtered alumina adsorbate with a basic aqueous or aqueous alcoholic solution and concentrating the eluate to produce a purified solution of the antibiotic. Antibiotic 833A may be in the form of an alkali metal, alkaline earth metal or ammonium salt or a salt with an organic base. Further purification is obtained by adding a C 1 -C 5 alkanol to the purified solution to precipitate insoluble material, filtering off the insolubles, passing the filtrate through a column of acidwashed alumina, eluting with a C 1 -C 5 alkanol gradient containing 2N ammonia and C 1 -C 5 alkanol, and finally ammonia, and lyophilizing the active fractions. Further purification is by dissolving solids in methanol, adding butanol, evaporating off the methanol and recovering the antibiotic from the butanol extract.
