Abstract

1,223,763. Chymotrypsin B. L'OPOCHIMIE. 29 Oct., 1968 [3 Nov., 1967], No. 51160/68. Heading C3H. In a process for obtaining chymotrypsin B by a fractionated precipitation of the proteins contained in a pulpy pancreas mixture followed by dissolution of the final cake in cold water, activation with trypsin and chromatographic purification, a first amount of ammonium sulphate is added to reach a degree of saturation of about 0À2, the mixture obtained is then filtered free of proteins of very high molecular weight and non-proteinaceous impurities, a second amount of ammonium sulphate is added to the filtrate to reach a degree of saturation comprised between 0À4 and 0À5 and the resulting mixture is filtered, giving a final cake the weight of which is approximately equal to 20% of that of the raw mixture of precipitable proteins contained in the aqueous extract before adding thereto said second amount of ammonium sulphate, said cake is redissolved in water, trypsin is added to the resulting solution in a quantity substantially equal to 1/40 of the weight of proteins contained in said solution, as determined from spectrophotometric readings, and the chymotrypsin B contained in the trypsinactivated solution is separated from other enzymes and proteins by a single chromatography. The cake may be dialysed to remove ammonium sulphate before the activation step.
