1,216,667. Antibiotic diumycin. E. R. SQUIBB & SONS Inc. 20 Dec., 1967 [27 Dec., 1966; 15 Nov., 1967], No. 57923/67. Heading C2A. An antibiotic diumycin and its components A and B, which are effective against gram positive bacteria, yeasts and fungi are coproduced with antibiotic umbrinomycin (Specification 1,216,668) by the cultivation of Streptomyces umbrinus ATCC 15972, or a mutant thereof, in an aqueous nutrient medium under aerobic conditions. The mycelium is separated from the broth at pH 3À0, is extracted with methanol and the methanol is removed at pH 6 to 8 to leave an aqueous suspension of the antibiotics. The aqueous suspension is extracted with butanol at pH 8À0 to remove umbrinomycin and the residual aqueous layer is extracted with water-saturated butanol at pH 3À0 and then back-extracted with water at pH 7À0. The aqueous extracts are concentrated to dryness, the residue dissolved in CH 3 OH, the insolubles removed and acetone added to precipitate the diumycin. It is purified by chromatography and countercurrent distribution. Components A and B are obtained by the countercurrent distribution of diumycin complex using the solvent system n-propanol, n-butanol, 0À5N. NH 4 OH. The diumycins have growth-promoting properties and may be added to animal feeds. They have general disinfecting properties.
