1,126,214. Antiprotozoal and anthelmintic antibiotic. HINDUSTAN ANTIBIOTICS Ltd. 5 Oct., 1965, No. 42265/65. Heading C2A. An antiprotozoal and anthelmintic antibiotic, antiamoebin, is prepared by cultivating an antiamoebin-producing strain of Emericellopsis synnematicola, e.g. ATCC 16540, E. poonensis, e.g. ATCC 16411, E. minima, E. humicola or Cephalosporium pimprina, in a culture medium under submerged or surface aerobic cultivation. Antiamoeba is isolated from a fermentation broth by (a) solvent extraction, (b) adsorption. Adsorption is on activated C or magnesium aluminium silicate, followed by elution with an alcohol, e.g. methanol. Extraction may be of the clarified broth or mycelium at e.g. pH 5À0 using waterimmiscible, polar solvents for the broth e.g. n-butanol, and aliphatic alcohols, ketones, and aliphatic mono-alkyl ethers of ethylene glycol for the mycelium. The antibiotic is precipitated from the extract by the addition of a non- solvent. Further purification is by crystallization from solvents. Antiamoebin is a neutral polypeptide, forming white needles, melting point 219-220‹ C., [α]D<SP>D5</SP> + 10 (C = 1À02% CH 3 OH); soluble in CH 3 OH, C 2 H 5 OH, C 3 H 7 OH, pyridine, glacial acetic acid, dimethyl formamide, methyl ether of ethylene glycol; insoluble in water, chloroform, ether, petroleum ether, ethyl acetate, butyl acetate, acetone, benzene, methyl chloride and ethylene glycol; acid hydrolysis produces the amino acids proline, α-amino isobutyric acid, phenylalanine, valine, leucine, hydroxyproline, lysine and glutamic acid; elementary analysis C = 56À11%, H = 7À56% and N = 13À56%; U.V. (Fig. 1, not shown); I.R. Figs. 2 and 3. A therapeutic oral preparation for the treatment of protozoal and helminth infections in man and animals comprises the antibiotic antiamoebin.
