Abstract

A complex of ferric iron and monosaccharide phosphate ester is prepared by reacting a colloidal aqueous solution of ferric hydroxide and an aqueous solution of an alkali metal salt of the said ester, in such proportions that the atomic ratio of iron to phosphorus does not exceed 10.7:1. The complex may be precipitated from the resulting solution by addition of about 0.5 volumes of ethanol, the complex containing iron and phosphorus in the said atomic ratio and any excess ester remaining in solution. Also described is a preparation, from glucose-1-phosphate, of the potassium salt using an anion exchange resin. The monosaccharide esters may be fructose-1-phosphate or glucose-1-phosphate. The aqueous solution of the complex is for use in pharmaceutical compositions (see Division A5).ALSO:Intravenously administrable iron compositions comprise an aqueous solution of the complex formed by reacting a colloidal aqueous solution of ferric hydroxide with an aqueous solution of an alkali metal salt of a monosaccharide phosphate ester, in such proportions that the atomic ratio of iron to phosphorus does not exceed 10.7:1. The esters may be fructose-1-phosphate or glucose-1-phosphate preferably as sodium or potassium salts. The ferric hydroxide solution is prepared in known manner by adding ammonia solution to ferric chloride solution and dialysing against water; it should contain about 5% Fe2O3. The reaction is preferably performed by adding the ferric hydroxide solution to the sugar phosphate solution, rather than vice-versa, to avoid forming and redissolving a precipitate. The ratio of iron to phosphorus is preferably below 10.7:1, so that unreacted sugar phosphate is present in the composition for its stabilising effect. The final solution may be further stabilised by boiling after preparation, and also by addition thereto of sodium citrate.
