Abstract

A mixed inorganic acid salt of dihydrostreptomycin is prepared by adding to an aqueous solution of a single acid salt of dihydrostreptomycin at least one molar equivalent, and preferably about two, of a water-soluble salt having a different inorganic anion and a cation which is capable of producing a water-soluble inorganic salt with the acid of the dihydrostreptomycin. The mixed acid salt of dihydrostreptomycin may contain two or three anions, one of which may be trivalent, e.g. phosphate, the others are mono- or divalent. A salt containing a monovalent anion and a divalent anion is preferred. Specified anions are sulphate, sulphite, hydrosulphite, thiosulphate, chloride, bromide, iodide, chlorate, bromate, nitrate, nitrite, and thiocyanate. Preferred water-soluble salts are the alkali metal, ammonium and organic ammonium salts of the above anions. The initial aqueous solution has a potency of at least 80,000 mcg./ml. and preferably at least 200,000 mcg./ml. Excess water-soluble salt is used to reduce the solubility in water of the mixed acid salt or dihydrostreptomycin. Crystallization is enhanced by cooling and seeding. The mixed acid salt may also be formed by adding the chosen anions, as acids, to the base dihydrostreptomycin. The mixed acid salts of streptomycin or mannosido-dihydrostreptomycin are not formed in the above manner. A crude dihydrostreptomycin obtained by adsorbing streptomycin from a filtered culture broth on to an ion-exchange resin, eluting and hydrogenating with Raney nickel catalyst is purified by forming its mixed acid salt in the above manner. Among specific salts are dihydrostreptomycin sulphate-iodide, dihydrostreptomycin sulphitebromide and dihydrostreptomycin sulphiteiodide.
