Abstract
A pyrogen-free aqueous solution of a sodium or potassium salt of a calcium chelate compound of an alkylene-polyamine polyacetic acid at pH 7.2-7.4 is used for medical injection. Diamine tetra-acetic acids mentioned are ethylene diamine tetra-acetic acid, trimethylene diamine tetra-acetic acid, propylene diamine tetra-acetic acid, 1,2-cyclohexylene diamino N,N-tetra-acetic acid, diethyl ether b,b1-diamino N,N1-tetra-acetic acid, ethylene glycol 1,2-ethylene ether o ,o 1, diamino N,N1, tetra-acetic acid. In the preparation of the sodium calcium complex of ethylene diamine tetraacetic acid, a solution in pyrogen-free water of the repeatedly crystallized acid is reacted with a pyrogen-free solution of 2 mols. sodium hydroxide with stirring and gentle heating, and 1 mol. calcium chloride, acetate, oxide, hydroxide carbonate &c. is then added with agitation and heating. The pH of the solution is finally brought to 7.2 to 7.4 by addition of sodium or potassium hydroxide, or of hydrochloric, citric or acetic acid.ALSO:A preparation for reducing, by base exchange, the content of toxic metal ions which may be present in the system consists of a pyrogen-free aqueous solution of an alkali metal, particularly the disodium or dipotassium, salt of a calcium chelate compound of a diamine N, N1 tetra acetic acid of pH 7.2-7.4. The solution may be injected, or taken into the body by ingestion or by skin absorption. Thus, the solution may be evaporated to dryness, and the solid incorporated in a tablet with dextrose, or gelatin-coated. Or the solution may be mixed with a water-miscible ointment to form an emulsion containing 5 per cent of the active ingredient. Diamine tetra acetic acids mentioned are ethylene diamine tetra acetic acid, trimethylene diamine tetra acetic acid, proplyene diamine tetra acetic acid, 1, 2 cyclohexylene diamino N, N1 tetra acetic acid, diethyl ether b, b1 diamino N, N1 tetra acetic acid, ethylene glycol 1, 2 ethylene ether o 1 o 1 diamino N, N1 tetra acetic acid. The preparation of the complex of ethylene diamine tetra acetic acid is described, (see Group IV(b)), the pH of the solution being finally brought to 7.2-7.4 by addition of sodium or potassium hydroxide, or of hydrochloric, citric or acetic acid.
