Abstract

L-Glutamic acid is obtained by inoculating bacteria, characterized by the fact that they belong to a specific growth-factor-requiring mutant strain of the species Microbacterium ammoniaphilum ATCC 15354, said mutant strain being selected from a hypoxanthine-requiring mutant strain, a L-histidine-requiring mutant strain, a hypoxanthine- and L-histidine-requiring mutant strain, or a L-arginine-requiring mutant strain, into a culture medium containing carbohydrate material, nitrogen source, inorganic salts, biotin, and the growth factor required by the bacteria, the amount of biotin being more than that required for the maximum growth of the bacteria and the amount of the growth factor being less than that required for the maximum growth of the bacteria, and culturing the medium under aerobic conditions to accumulate L-glutamic acid therein. Specified carbohydrate materials are beet sugar molasses, cane sugar molasses, their waste molasses, crude sugar, starch hydrolysate liquids, glucose, fructose, maltose and sucrose. Specified nitrogen sources are inorganic and organic ammonium salts, urea, ammonia, meat extract, peptone, yeast extract, corn steep liquor and various protein hydrolysates. Examples are given.
