Abstract
Guanine and derivatives thereof having the formula <FORM:1066325/C2/1> or <FORM:1066325/C2/2> wherein R1 and R2 represent H or C1- 6 alkyl groups, are prepared by reacting 2,4-diamino-6-hydroxy -5-nitrosopyrimidine or a derivative thereof of the formula <FORM:1066325/C2/3> or <FORM:1066325/C2/4> with formamide under reducing conditions and with heating to complete ring closure, without isolation of intermediate products. The starting materials may be prepared in situ by a nitrosation reaction. Reducing conditions may be provided by hydrogen and a hydrogenation catalyst or by means of a reducing agent with or without hydrogen. Various solvents may also be present during the reaction. The examples describe the preparation of guanine and 3-methylguanine hydrochlorides.
