Abstract

1,053,269. Phenylaminophenyl-alkane carboxylic acids. SOC ANON RHONE-POULENC. Nov. 25, 1965 [Nov. 27, 1964; Oct. 15. 1965], No. 50187/65. Heading C2C. The invention comprises compounds of the formula wherein R represents a hydrogen atom or a C 1-4 alkyl groups; and salts thereof. The compounds are prepared by (a) decarboxylating a compound of the formula by heating; or (b) hydrolysing in an alkaline medium a compound of the formula wherein R 1 represents a hydrogen atom, a methyl group or an optionally substituted phenyl group and R 2 represents a C 1-4 alkyl group; or (c) subjecting 4-acetyldiphenylamine to the Willgerodt reaction or the Kindler modification of it, and hydrolysing by known methods the amide or thio amide produced to (4-phenylamino-phenyl) acetic acid; or (d) C- alkylating the compound of Formula I in which R represents a hydrogen atom. Anthranitic acid derivatives of Formula II are prepared by reacting a halogeno-benzoic acid of the formula with an aniline derivative of the formula Compounds of Formula III, wherein R 1 represents a hydrogen atom or a methyl group, are prepared by reacting a, halogenobenzene of the formula wherein Hal represents a halogen atom. with an aniline derivative of the formula wherein R 3 represents a hydrogen atom or a methyl group, itself prepared by acylating an amine of the formula Compounds of Formula III, wherein R 1 represents an optionally substituted phenyl group, are prepared by the Chapman rearrangement of an imino ether of the formula wherein R 4 represents an optionally substituted phenyl group, the imino ethers themselves being retained by reacting an alkali metal derivative of phenol with an imidoyl chloride of the formula or by reacting an alkali metal derivative of a phenol of the formula with an imidoyl chloride of the formula Imidoyl chlorides of Formula XI or XIII are prepared by the action of phosphorus pentachloride or thionyl chloride on an amide of the formula the amides themselves being obtained by aeylation of amines of Formula VIII, or of aniline. Pharmaceutical compositions having antiinflammatory, anti-rheumatic and analgesic activity, for oral, parenteral, rectal or topical administration, comprise a compound of the invention together with a pharmaceutical carrier.
