Abstract

The invention comprises trinicotinoyl glycer ine, and acid addition salts thereof. The compound is prepared by reacting nicotinic acid, or its metal salts, esters, anhydrides or halides, or acid addition salts, with glycerine (or partially nicotinoylated glycerine) or reactive derivatives of glycerine, such as a boric acid ester, or a glycerine halohydrin. The trinicotinoyl ester, when prepared, may be converted into its acid addition salts by a sufficient amount of acid, e.g. hydrohalic acids, sulphuric, phosphoric, toluene-sulphonic, acetic, oxalic, malonic, succinic, malic, maleic, and tartaric acids. The trinicotinyl ester may also be prepared from derivatives of nicotinic acid from which can be liberated a nicotinoyl residue, e.g. pyridine-a ,b -dicarboxylic acid. Two examples are given: (1) nicotinic acid is converted to the chloride-hydrochloride by reaction with thionyl chloride; this is reacted with glycerine in the presence of triethylamine, in dimethyl formamide as a solvent, to give, after working up, trinicotinoyl glycerine; (2) nicotinic acid anhydride and glycerine are reacted in the presence of pyridine, and the product worked up to give trinicotinoyl glycerine. The compound is for use as a vasodilating and fibrinolytic agent (see Division A5).ALSO:Vasodilator compositions for external and internal application, comprise trinicotinoyl glycerine, or an acid addition salt thereof, together with a pharmaceutically acceptable carrier. The composition also may contain a xanthine base, such as theophylline or a water-soluble derivative thereof, theobromine, or caffeine. The composition may be applied externally, as liniments or ointments; orally as tablets, pills, coated tablets, powders, or capsules; rectally, as retention suppositories; or parenterally in a suitable solution. Examples are given for preparation of tablets, with conventional exceipients; suppositories; and an injectable solution of the active agent in polyoxyethylene glycol/water, or propylene glycol/water, optimally with 2% of benzyl alcohol.
