Abstract

L'invention concerne un procédé de stabilisation d'un coenzyme et d'un enzyme labiles. Des co-enzymes labiles tels que le nicotinamide-adénine-dinucléotide (NAD) sont stabilisés par traitement d'un solvant organique en milieu aqueux dans un polymère convenable n'inhibant pas l'activité enzymatique. Un azoture peut être ajouté à la solution, non seulement comme agent bactériostatique, mais aussi comme agent stabilisant. Un second enzyme tel que le triphosphate d'adénosine peut aussi être stabilisé dans la même solution. Un ou plusieurs enzymes tels que l'hexokinase ou la glucose-6-phosphate-déshydrogénase peuvent également être stabilisés contre une dénaturation dans la même solution. Le procédé donne une composition très stable qui peut être utilisée dans des dosages pour le diagnostic biologique.The invention relates to a method of stabilizing a labile coenzyme and an enzyme. Labile co-enzymes such as nicotinamide-adenine-dinucleotide (NAD) are stabilized by treatment of an organic solvent in aqueous medium in a suitable polymer which does not inhibit the enzymatic activity. Azide can be added to the solution, not only as a bacteriostatic agent, but also as a stabilizing agent. A second enzyme such as ATP can also be stabilized in the same solution. One or more enzymes such as hexokinase or glucose-6-phosphate-dehydrogenase can also be stabilized against denaturation in the same solution. The method gives a very stable composition which can be used in assays for biological diagnosis.
