Abstract

Weakly alkaline protease ("Seaprose S") produced from moulds, esp. from strains of Aspergillus. Anti-inflammatory, useful in surgical and orthopaedic treatments. Seaprose S is a white powder soluble in water, insoluble in acetone and alcohol. It shows maximum activity at pH 7.0-8.5, and maximum stability at pH 5.0-9.0. Its m.wt. is 30400 and its activity is 2000 units/mg. The active site is one serine, and the molecule contains 17 other amino acids. It is prepared from cultures of Aspergillus, esp. A.melleus, A.oryzae, or A.sojae. The resultant enzymes are extracted with water and filtered. The filtrate is treated with 3 times its volume of ethanol and allowed to stand to precipitate the enzyme sought. This is purified by passage through an anion exchange resin in the acetate form. The enzyme is precipitated from aqueous ethanol, then from aqueous acetone and is obtained in 70% yield.
