Abstract
Highly purified fibrinolytic enzyme for dissolving arterial and venous thrombi is prepared by extracting crude enzyme mixture from Aspergillus ochraceus with aqs. buffer solution (pref. with 0.05-0.1 M tris-HCl buffer or 0.05-0.1 M NH4 acetate buffer of pH 6.5-7.5, partic. 7-7.2); filtering the extract containing active ingredient over an anion exchanger DEAE-Sephadex or cellulose; precipitating the active ingredient from the filtrate with water-miscible organic solvents; re-dissolving the precipitate with water after drying in vacuo and subjecting it to gel chromatography on cross-linked dextran gel Sephadex G-75 or Sephadex G-100 and freeze drying the protease-containing fractions.
