A process for the isolation of a fibrinolytic enzyme from mould fungus cultures, wherein a crude enzyme mixture from Aspergillus ochraceus is extracted with an aqueous buffer solution, the extract containing the active material is filtered through a diethylaminoethyl-groupcontaining cross-linked dextran gel anion exchanger or through a diethylaminoethyl-cellulose anion exchanger, the active material precipitated from the filtrate obtained by means of a water-miscible organic solvent, the precipitate obtained separated and dried in a vacuum, again dissolved in water and subjected to gel chromatography on cross-linked dextran gel and the protease-containing fractions obtained then lyophilised. The cross-linked dextran gel must be one capable of fractionating substrates of M.Wt. 1000 to 100,000 and have a water regain of 7À5-10; suitable commercially available types are mentioned.
