LA INCAPATIZACION TIENE LUGAR ELIMINANDO, AL MENOS, PARTE DE DICHOS ELEMENTOS, POR LIGADURA DE LOS EXTREMOS ROMOS DE FRAGMENTOS DE DNA PRODUCIDOS POR DIGESTIONES CON ENZIMAS DE RESTRICCION, Y DONDE LOS FRAGMENTOS SE HACEN DE EXTREMOS ROMOS POR USO DE LA ENZIMA DNA POLIMERASA I; LAS COLAS 3'' DE HEBRA UNICA SE ELIMINAN, Y LAS COLAS 5'' DE HEBRA UNICA SE VUELVEN A EMPAREJAR EN FORMA DE DOBLE HEBRA.
