Abstract
Un conjugado de un polímero y al menos un resto activo seleccionado de un resto terapéutico, un resto seleccionador de una diana, y un resto de diagnóstico, comprendiendo dicho polímero unidades que se repiten que están representadas por la siguiente fórmula (I): -[N(R1)-(CHR2)m]- (I) en la que: - R1 es R3-(CHR4)n-CONH-R5; - R2 se selecciona de H y alquilo de C1-5 opcionalmente sustituido; - R3 es CO, C(O)O, C(O)NH o C(S)NH; - R4 se selecciona de H y alquilo de C1-5 opcionalmente sustituido; - R5 es H; un alquilo de C1-5; arilo; o un resto que comprende un grupo funcional que se puede usar para conjugación, seleccionándose dicho grupo funcional de un grupo que comprende un átomo de nitrógeno que está enlazado covalentemente a al menos un carbono, hidrógeno o heteroátomo; un tiol; una fosfina; un alquino; un alqueno; un arilo; un aldehído; una cetona; un acetal; un éster; un ácido carboxílico; un carbonato; un cloroformiato; un hidroxilo; un éter; una vinilsulfona; un epóxido; sulfonato; ácido haloacético y anhídrido; - m es 2 o 3; y - n es 1-5.
Description
tal grupo funcional es que tales aminas terciarias son catiónicas cuando están presentes en una disolución acuosa, lo que les permite interaccionar con ARN y ADN de, por ejemplo, células tumorales. El grupo funcional, que comprende un átomo de nitrógeno que está enlazado covalentemente a al menos un carbono, hidrógeno o heteroátomo, se selecciona preferiblemente de maleimida, isocianato, isotiocianato, disulfuro de ortopiridilo y haloacetamida.
En una realización preferida de tal polímero, R5 representa R6NR7’R7”, en el que R6 representa un enlace covalente o un alquileno de C1-5 opcionalmente sustituido; y R7’ y R7” se seleccionan independientemente de H y C1-5.
Según una realización particularmente preferida, R6 se selecciona de un enlace covalente, metileno, etileno y propileno, debido a que estos polímeros presentan la solubilidad más elevada en agua.
Según otra realización preferida, R5 es H o alquilo de C1-3. Incluso más preferiblemente, R5 es H o metilo, lo más preferible, R5 es metilo.
En una realización preferida adicional de la presente invención, el número entero m en la fórmula (I) es 2.
El polímero según la presente invención tiene típicamente un peso molecular de al menos 1.000 g/mol. Incluso más preferiblemente, el polímero tiene un peso molecular en el intervalo de 2.000-100.000 g/mol, lo más preferible, de 5.000-50.000 g/mol.
En el caso de que R5 sea un resto que comprende un grupo funcional como se especifica aquí, las unidades que se repiten representadas por la fórmula (I) representan típicamente al menos 2% en moles, más preferiblemente 5-70% en moles, y lo más preferible, 10-30% en moles del polímero.
En el caso de que R5 sea H o alquilo de C1-5, las unidades que se repiten representadas por la fórmula (I) representan típicamente al menos 10-100% en moles, más preferiblemente 20-100% en moles, y lo más preferible, 40-100% en moles del polímero.
Además de las unidades que se repiten (monoméricas) representadas por la fórmula (I), el presente polímero puede contener adecuadamente otras unidades que se repiten, principalmente otras unidades de iminoéter cíclico, tales como 2-oxazolinas y 2-oxazinas. En una realización particularmente preferida de la presente invención, el polímero contiene 0-98% en moles, más preferiblemente 30-95% en moles, y lo más preferible, 70-90% en moles de unidades que se repiten representadas por la fórmula (Y)
-[N(Ra)-(CHR2)m]-(fórmula Y)
en la que las unidades que se repiten representadas por la fórmula (Y) son diferentes de las unidades que se repiten representadas por la fórmula (I); y en la que Ra representa C(O)H, C(O)Rk o Rk, y en el que Rk representa alquilo de C1-22 opcionalmente sustituido, cicloalquilo opcionalmente sustituido, aralquilo opcionalmente sustituido, o arilo opcionalmente sustituido, y en la que el número entero m es 2 o 3.
Preferiblemente, Rk, representa un alquilo de C1-22 opcionalmente sustituido, más preferiblemente alquilo de C1-10, y lo más preferible, alquilo de C1-5.
Las unidades que se repiten representadas por la fórmula (I) y las unidades que se repiten representadas por la fórmula (Y) representan juntas típicamente al menos 50% en peso del presente polímero. Incluso más preferiblemente, dichas unidades que se repiten representan juntas al menos 80% en peso, y lo más preferible, 100% en peso de las unidades que se repiten contenidas en el presente polímero.
El polímero según la presente invención puede ser un homopolímero o un copolímero. Lo más preferible, el presente polímero es un copolímero de polioxazolina.
Se prefiere además que el polímero según la presente invención sea un polímero lineal.
Un segundo aspecto de la presente invención se refiere a un conjugado de un polímero y al menos un resto activo seleccionado de un resto terapéutico, un resto seleccionador de una diana, y un resto de diagnóstico, comprendiendo dicho polímero unidades que se repiten que están representadas por la siguiente fórmula (I):

El número entero n es preferiblemente 1-4, y lo más preferible, 1-2.
Preferiblemente, el conjugado se obtiene permitiendo que el resto activo, tal como un resto terapéutico, un resto seleccionador de una diana, y/o un resto de diagnóstico, reacciona con al menos uno de los grupos funcionales en R5 del polímero. Ventajosamente, el resto activo está enlazado covalentemente al polímero.
Como alternativa, el conjugado es un montaje no covalente basado en múltiples interacciones no covalentes tales como enlazamiento de hidrógeno o interacciones electrostáticas (por ejemplo, que implican aminas terciarias en el polímero), con enlazamiento de hidrógeno complementario o con macromoléculas cargadas opuestamente, incluyendo polímeros sintéticos, biopolímeros y sustancias biofarmacéuticas. En una realización preferida, el conjugado es un montaje capa a capa del polímero y una macromolécula. En otra realización preferida, el conjugado es un montaje en forma de un vector no viral para el suministro de genes.
El resto terapéutico que está contenido en el presente conjugado es capaz preferiblemente de inhibir una cascada de transducción de señales en un sistema celular, interfiere con el citoesqueleto, o se intercala en la doble hélice del ADN. Se prefiere además que el resto terapéutico, unido a o separado del polímero, tenga actividad antifibrótica, antiangiogénica, antitumoral, inmunoestimulante o inductora de la apoptosis. Los restos terapéuticos particularmente preferidos son doxorrubicina, daunorrubicina, epirrubicina, paclitaxel, mesoclorina, 5-fluorouracilo y cis-platino.
El resto seleccionador de una diana que está contenido en el conjugado comprende preferiblemente un grupo folato, puesto que se sabe que el folato tiene una elevada afinidad natural por la proteína receptora de folato, que es expresada habitualmente en la superficie de muchos cánceres humanos. Otro resto seleccionador de una diana preferido es la secuencia de arginina-glicina-aspartato (RGD), ya que selecciona como diana a la integrina, que también está sobreexpresada en tejidos cancerosos. Los restos seleccionadores de una diana según la presente invención pueden comprender también anticuerpos, fragmentos de la unión de fragmento-antígeno (FABs), anticuerpos de un solo dominio (nanocuerpos), péptidos o nucleótidos, tales como ARN, ADN y ARNpi.
El resto de diagnóstico se selecciona preferiblemente de radiomarcadores (radionúclidos), agentes que se pueden presentar en imágenes mediante PET, agentes que se pueden presentar en imágenes mediante SPECT, agentes que se pueden presentar en imágenes mediante MRI, y sondas fluorescentes.
Un tercer aspecto de la presente invención se refiere al conjugado mencionado anteriormente para uso en el tratamiento terapéutico, tratamiento profiláctico o diagnóstico de enfermedades o trastornos, particularmente cáncer.
Según una realización particularmente preferida de dicho uso, el conjugado se administra intravenosamente, subcutáneamente u oralmente. Lo más preferible, el conjugado se administra subcutáneamente u oralmente.
Un cuarto aspecto de la presente invención se refiere a un método para preparar un polímero como se describe anteriormente, en el que el método comprende:

Se preparó un polímero de polioxazolina según la presente invención mediante un método que comprende las siguientes etapas.
Poli(2-etil-2-oxazolina) (PEtOx, Mw 10.000, grado de polimerización 100), preparada mediante polimerización de apertura de anillo catiónica viviente bajo irradiación de microondas (140ºC, concentración de monómero 4 M en acetonitrilo, tosilato de metilo como iniciador), se sometió a hidrólisis ácida controlada para producir un copolímero de poli[2-etil-2-oxazolina/etilenimina]. Se hidrolizó dieciocho por ciento de las unidades de 2-etil-oxazolina iniciales.
Esta poli[2-etil-2-oxazolina/etilenimina] se funcionalizó en un enfoque de dos etapas mediante el acoplamiento en primer lugar 3-isocianatopropionato de metilo (3 eq; reacción llevada a cabo en cloroformo seco durante 5 minutos a 150ºC en un horno de microondas), produciendo el copolímero funcionalizado de cadena lateral de éster metílico, en el que la cadena lateral está unida a la cadena principal vía un enlace de urea. La espectroscopía infrarroja (IR) reveló claramente la aparición de la banda característica de éster metílico en 1732 cm-1 , mientras que la espectroscopía de resonancia magnética nuclear de protón (RMN 1H) mostró claramente la señal de éster metílico en 3,7 ppm. SEC reveló un Mw de 1,98 x 104, Mn de 1.83 x 104 y una Ð de 1.1.
Este polímero se puede modificar además mediante reacción del éster metílico con diversas aminas, como se ejemplifica en el Ejemplo 6. Esto se demostró mediante reacción del polímero con etilamina disolviendo 0,3 g de polímero en 2 ml de etilamina, y calentando toda la noche a 70ºC. El éxito de la reacción se demostró mediante espectroscopía de RMN 1H en DMSO-d6, que revela la señal de amida específica en 7,9 ppm. La espectroscopía IR también confirmó la reacción por la desaparición de la banda en 1732 cm-1. SEC también confirmó un incremento en la masa molar tras la reacción hasta un Mw de 2,14 x 104, Mn de 1,98 x 104 y una Ð de 1,1.
Ejemplo 8
Se preparó un polímero de polioxazolina según la presente invención mediante un método que comprende las siguientes etapas.
Poli(2-etil-2-oxazolina) (PEtOx, Mw 10.000, grado de polimerización 100), preparada mediante polimerización de apertura de anillo catiónica viviente bajo irradiación de microondas (140ºC, concentración de monómero 4 M en acetonitrilo, tosilato de metilo como iniciador), se sometió a hidrólisis ácida controlada para producir un copolímero de poli[2-etil-2-oxazolina/etilenimina]. Se hidrolizó dieciocho por ciento de las unidades de 2-etil-oxazolina iniciales.
Esta poli[2-etil-2-oxazolina/etilenimina] se funcionalizó en un enfoque de dos etapas mediante el acoplamiento en primer lugar 3-isotiocianatopropanoato de metilo (3 eq; reacción llevada a cabo en cloroformo seco durante 5 minutos a 150ºC en un horno de microondas), produciendo el copolímero funcionalizado de cadena lateral de éster metílico, en el que la cadena lateral está unida a la cadena principal vía un enlace de urea. La espectroscopía infrarroja (IR) reveló claramente la aparición de la banda característica de éster metílico en 1732 cm-1, mientras que la espectroscopía de resonancia magnética nuclear de protón (RMN 1H) mostró claramente la señal de éster metílico en 3,7 ppm. SEC reveló un Mw de 2,28 x 104, Mn de 2,05 x 104 y una Ð de 1,1.
Este polímero se puede modificar además mediante reacción del éster metílico con diversas aminas, como se ejemplifica en el Ejemplo 6. Esto se demostró mediante reacción del polímero con etilamina disolviendo 0,3 g de polímero en 2 ml de etilamina, y calentando toda la noche a 70ºC. El éxito de la reacción se demostró mediante espectroscopía de RMN 1H en DMSO-d6, que revela la señal de amida específica en 8,1 ppm. La espectroscopía IR también confirmó la reacción por la desaparición de la banda en 1732 cm-1. SEC también confirmó un incremento en la masa molar tras la reacción hasta un Mw de 2,26 x 104, Mn de 2,08 x 104 y una Ð de 1,1.

El homopolímero de poli(2-(2-metoxicarboniletil)-2-oxazolina), preparado en el ejemplo 4, se hizo reaccionar con hidrazina para producir el homopolímero de poli(hidrazona etil-2-oxazolina).
