Ejemplo 5:
Farmacocinética de conjugado de rFVIIa-PSA en ratas normales
Se preparó un conjugado de rFVIIa-PSA según el ejemplo 2 usando una concentración de MAL-FMS-OSU de 0,05 mg/mg de proteína. Se anestesiaron 8 ratas normales (4 machos, 4 hembras) y se aplicó el conjugado de rFVIIa-PSA en tampón (glicilglicina 1,3 g/l, cloruro de sodio 3 g/l, manitol 30 g/l, CaCl2 x 2H2O 1,5 g/l, Tween 80 0,1 g/l, pH 5,5) mediante inyección intravenosa en la vena de la cola en una dosis de volumen de 10 ml por kg (1200 µg de proteína/kg). Se usó rFVIIa no modificado en una dosis de 1200 µg de proteína/kg como control en 8 ratas normales (4 machos, 4 hembras). Se tomaron muestras de sangre de la arteria de la cola 5 minutos, 1 hora, 2, 4, 7, 10 y 24 horas tras la aplicación de la sustancia y se preparó plasma con citrato y se congeló para su análisis adicional.
Se midió la actividad de FVIIa en plasma con un ensayo de coagulación (Staclot, Diagnostica Stago, Asnières, Francia), se determinó el antígeno de FVII con un ELISA (anticuerpo policlonal anti-FVII humano). Se evaluaron los resultados estadísticamente. Para la actividad de coagulación de FVIIa, el área bajo la curva (AUC) ajustada a la dosis fue de 0,014 para rFVIIa no modificado y aumentó hasta 0,015 para el conjugado de rFVIIa (0-infinito). La semivida terminal aumentó desde 2,3 hasta 4,4 horas y el tiempo de residencia medio (MRT) desde 1,4 hasta 2,4 horas. Para el antígeno, el AUC ajustada a la dosis (0-infinito) aumentó desde 0,010 (rFVIIa no modificado) hasta 0,014 (conjugado de rFVIIa), la semivida terminal aumentó desde 1,4 hasta 2,3 horas y el MRT desde 1,5 hasta 2,2 horas. Se llevaron a cabo todos los cálculos mediante el uso de un programa estadístico (program R: A language and environment for statistical computing. R Foundation for Statistical Computing, Viena, Austria, ISBN 3-900051-070, URL http://www.R-project.org). Los resultados de farmacocinética se ilustran en las figuras 3 y 4.
Ejemplo 6:
Farmacocinética de conjugado de rFVIIa-trímero-PSA en ratas normales
Se preparó conjugado de rFVIIa-trímero-PSA según el ejemplo 3 usando una concentración de MAL-FMS-OSU de 0,05 mg/mg de proteína. Se anestesiaron 6 ratas normales (3 machos, 3 hembras) y se aplicó el conjugado de rFVIIa-trímero-PSA en tampón (glicilglicina 1,3 g/l, cloruro de sodio 3 g/l, manitol 30 g/l, CaCl2 x 2H2O 1,5 g/l, Tween 80 0,1 g/l, pH 5,5) mediante inyección intravenosa en la vena de la cola en una dosis de volumen de 10 ml por kg (1200 µg de proteína/kg). Se usó rFVIIa no modificado en una dosis de 1200 µg de proteína/kg como control en 6 ratas normales (3 machos, 3 hembras). Se tomaron muestras de sangre de la arteria de la cola 5 minutos, 1 hora, 2, 4, 7, 10 y 24 horas tras la aplicación de la sustancia y se preparó plasma con citrato y se congeló para su análisis adicional.
Se midió la actividad de FVIIa en plasma con un ensayo de coagulación (Staclot, Diagnostica Stago, Asnières, Francia) y se construyó la curva de eliminación. La farmacocinética mejorada del conjugado de rFVIIa-trímero-PSA se ilustra en la figura 5.
Ejemplo 7:
Conjugación de rFIX con PSA usando el conector MAL-FMS-OSU
A 0,6 ml de una disolución de FIX recombinante (8 mg/ml) en tampón Hepes 20 mM, pH 7,4, se le añadió el conector bifuncional MAL-FMS-OSU (preparado tal como explican resumidamente Tsubery et al., J Biol Chem. 279, 3811838124 (2004)) (concentración: 0,07 mg/mg de proteína) y se incubó a T.A. durante 30 min. Se preparó PSA derivatizado que contenía un grupo SH terminal según el ejemplo 1. Se añadió a la mezcla el derivado de PSA (concentración: 32 mg de PSA-SH/mg de proteína – exceso molar de 100 veces) y se incubó durante 2 horas adicionales a T.A. Se detuvo la reacción añadiendo una disolución acuosa de glicina 0,1 M (concentración final 10 mM) y cisteína 5 mM (concentración final 0,5 mM). Se separaron los reactivos libres del conjugado de rFIX-PSA mediante cromatografía de interacción hidrófoba usando una columna de Butyl-Sepharose con relleno previo (HiTrap Butyl FF 5 ml, GE Healthcare). Se añadió un tampón que contenía NaCl 5 M (tampón Hepes 50 mM, NaCl 5 M, Tween 80 al 0,01%, CaCl2 6,7 mM, pH 6,9) a la disolución que contenía PSA-rFIX para dar una concentración final de NaCl 3 M. Se aplicó esta mezcla a la columna, que se lavó posteriormente con 10 VC de tampón de equilibrado (tampón Hepes 50 mM, NaCl 3 M, Tween 80 al 0,01%, CaCl2 6,7 mM, pH 6,9) y se llevó a cabo la elución del conjugado de rFIX-PSA con tampón Hepes 50 mM, pH 7,4, que contenía CaCl2 6,7 mM. Tras la elución del conjugado, se ajustó el pH a pH 6,9. El eluato contenía 0,24 mg/ml de proteína tal como se mide mediante el ensayo de BCA, se demostró la evidencia de PSA unido en el conjugado mediante el ensayo con resorcinol (Svennerholm, Biochim Biophys Acta 24, 604-611 (1957)). En una etapa final, se concentró el eluato 10 veces mediante ultrafiltración/diafiltración (UF/DF) usando una membrana de 30 kD (celulosa regenerada/Millipore) frente a Hepes 20 mM, NaCl 50 mM, CaCl2 1 mM, pH 7,4.
