Description
inhiben NHE, el compuesto de inhibición de NHE (es decir, un compuesto de Fórmula (I)) que posee propiedades fisicoquímicas generales que lo hacen sustancialmente impermeable o sustancialmente no biodisponible de manera sistémica. El resto Núcleo puede estar unido a esencialmente cualquier posición sobre, o dentro del resto de molécula pequeña inhibidor de NHE, siempre que la instalación del mismo no afecte significativamente de manera adversa a la actividad inhibidora de NHE.
En otra realización, se proporciona un compuesto que tiene la estructura de Fórmula (II):

Por lo tanto, los compuestos de la presente invención pueden existir en forma de un hidrato, incluyendo un monohidrato, dihidrato, hemihidrato, sesquihidrato, trihidrato, tetrahidrato, y similares, así como las formas solvatadas correspondientes. Una "composición farmacéutica" se refiere a una formulación de un compuesto de la invención y un medio generalmente aceptado en la técnica para la administración del compuesto biológicamente activo a mamíferos, por ejemplo, seres humanos. La expresión "lumen gastrointestinal" se usa de forma intercambiable en el presente documento con el término "lumen", para referirse al espacio o cavidad dentro de un tracto gastrointestinal (tracto Gl, que también puede denominarse el intestino), delimitado por la membrana apical de células epiteliales del Gl del sujeto. En algunas realizaciones, los compuestos no se absorben a través de la capa de células epiteliales del tracto Gl (también conocido como el epitelio del Gl). "Mucosa gastrointestinal" se refiere a la capa o capas de células que separan el lumen gastrointestinal del resto del cuerpo e incluye mucosa gástrica e intestinal, tal como la mucosa del intestino delgado. En tales casos, la localización en el lumen gastrointestinal se refiere a

Inhibición de la absorción intestinal de sodio
5 La concentración de sodio urinario y la forma fecal se midieron para evaluar la capacidad de los compuestos de ejemplo seleccionados para inhibir la absorción de sodio del lumen intestinal. Se compraron ratas Sprague-Dawley de ocho semanas a través de Charles River Laboratories (Hollister, CA), se alojaron 2 por jaula, y se aclimataron durante al menos 3 días antes del inicio del estudio. Se alimentó a los animales con comida para roedores Harlan
10 Teklad Global 2018 (Indianápolis, IN) y agua ad libitum a lo largo del estudio y se mantuvieron en un ciclo de luz oscuridad estándar de 6 AM a 6 PM. El día del estudio, entre 4 PM y 5 PM, se dosificó a un grupo de ratas (n = 6) mediante alimentación oral forzada con un compuesto de ensayo a la dosis indicada o vehículo (agua) a un volumen de 10 ml/kg. Después de la administración de la dosis se puso a los animales en jaulas metabólicas individuales donde se les alimentó con el mismo pienso en forma de comida y con agua a voluntad. A las 16 h después de la
15 dosis, se recogieron las muestras de orina y se determinó la forma fecal mediante dos observaciones

independientes. Las formas fecales se puntuaron de acuerdo con una escala común asociada al aumento de agua fecal a la observación más húmeda en el embudo de recolección de la jaula (1, deposición normal; 2, deposición que se adhiere a los lados del embudo de recolección debido a la hidratación; 3, pérdida de la forma normal de la deposición; 4, pérdida completa de la forma con un patrón de coagulación; 5, evidentes flujos de fecales). Se 5 determinó una puntuación de forma fecal de rata (FFS) promediando ambas puntuaciones observacionales para todas las ratas dentro de un grupo (n = 6). En cada estudio, el promedio del grupo de vehículo fue 1. Estos promedios se comunican en la Tabla 5. Para las muestras de orina, se determinaron los volúmenes gravimétricamente y se centrifugaron a 3.600 x g. Los sobrenadantes se diluyeron 100 veces en agua MilliQ desionizada, después se filtraron a través de una placa de filtro GHP Pall AcroPrep de 0,2 µm (Pall Life Sciences, 10 Ann Arbor, MI) antes del análisis por cromatografía iónica. Se inyectaron diez microlitos de cada extracto filtrado en un sistema de cromatografía iónica Dionex ICS-3000 (Dionex, Sunnyvale, CA). Los cationes se separaron mediante un método isocrático usando ácido metanosulfónico 25 mM como eluyente en una columna lonPac CS12A 2 mm d.i. x 250 mm, tamaño de partícula 8 µm, de intercambio catiónico (Dionex). El sodio se cuantificó usando patrones preparados a partir de una mezcla patrón de cationes que contenía Li+, Na+, NH4+, K+, Mg2+, y Ca2+ (Dionex). La 15 masa media de sodio orinado para cada grupo en el periodo de 16 h se determinó con el grupo de vehículo orinando normalmente aproximadamente 21 mg de sodio. 5 Se midió la capacidad de los compuestos para inhibir el antipuerto de H+ dependiente de Na+ mediado por NHE-3 después de la aplicación y lavado usando una modificación del método de tinción sensible a pH descrito anteriormente en el Ejemplo 74. Se obtuvieron células renales de zarigüeya (KO) a partir de la ATCC y se propagaron siguiendo sus instrucciones. El gen de NHE-3 de rata se introdujo en células OK mediante

Evaluación farmacocinética en la bilis
5 Se dosificó a ratas Sprague-Dawley canuladas en el conducto biliar (BDC) con compuestos de ensayo mediante alimentación oral forzada y se recogió una única alícuota de bilis a través de la cánula en las 24 horas posteriores a la dosificación. Las muestras de bilis se trataron con acetonitrilo y las proteínas precipitadas se retiraron mediante centrifugación. Algunos compuestos necesitaron extracción líquido-líquido usando MTBE. Tras la centrifugación, se
10 diluyeron las muestras según fue necesario en fase móvil y se analizaron mediante LC-MS/MS. Las concentraciones de los compuestos en la bilis se determinaron mediante interpolación a partir de una curva patrón de calibración preparada en bilis de rata de ratas BDC no tratadas. La recuperación precisa de muestras de control de calidad se confirmó para aceptar cada ciclo analítico. La Tabla 9 ilustra los datos de la exposición biliar de compuestos de ejemplo seleccionados. La concentración en bilis se comunica en nM y representa el resultado medio de n = 3 ratas.
