Abstract
2-Acylphenothiazines of the general formula: <FORM:0900755/IV (b)/1> in which A is an alkylene radical containing 2-5 carbon atoms, R is an alkyl radical containing 1-4 carbon atoms and R1 is an hydroxy substituted alkyl radical containing 1-4 carbon atoms are prepared by reacting a phenothiazine of the general formula: <FORM:0900755/IV (b)/2> either with an alkylene halide or haloalkyl aryl sulphonate and condensing the 2 2-acyl-10-(haloalkyl)-phenothiazine so produced with an N-hydroxyalkyl piperazine, or with piperazine and reacting the 2-acyl-10-(piperazinylalkyl) phenothiazine with an alkylene halohydrin, an alkylene oxide or an alkylene carbonate or reacting the above 2-acylphenothiazine with either an N-haloalkylpiperazine and reacting the resulting 2-acyl-10-(alkylpiperazinyl)-pheno-thiazine with an alkylene halohydrin, alkylene oxide or alkylene carbonate, or with an N-haloalkyl-N1-hydroxyalkylpiperazine, the condensation involving the 10-position of the 2-acylphenothiazine in each of the aforesaid reactions taking place in the presence of sodium hydride dissolved in dimethyl formamide or dimethyl acetamide. The products may be converted to acid addition salts. Tranquillising compositions comprise the 2-acylphenothiazines or p acid addition salts thereof produced by the process of the invention and an inert carrier. Salts specified include ascorbates and nicotinates. Reference has been directed by the Comptroller to Specification 819,886
