Description
La resina se desprotegió y se escindió de la resina a través de un tratamiento con 5 ml del siguiente reactivo: 5% de TIS, 2% de agua, 5% (p/v) de ditiotreitol (DTT), 88% de TFA y se mezcló durante 3,5 horas. El material filtrado se recogió en 45 ml de éter etílico anhidro frío. El precipitado sedimentó centrifugando durante 10 minutos a 3500 rpm en una centrífuga refrigerada. El éter se decantó, y el péptido se resuspendió en éter de nuevo aporte. El tratamiento 5 con éter se llevó a cabo un total de 2 veces. Después del último lavado con éter, el péptido se dejó secar al aire para eliminar el éter residual. El sedimento de péptido se resuspendió en 8 ml de acetonitrilo (Acn), seguido de 8 ml de agua desionizada y se permitió que se disolviera completamente. A continuación, la solución de péptido se analizó por espectrometría de masas. Con el análisis de masas que empleaba ionización por electronebulización se identificó un producto principal que contenía una masa de 5205,1 Dalton; que se correspondía al producto lineal 10 deseado. El producto crudo (aproximadamente 500 mg) se analizó mediante HPLC, empleando una columna de 250 x 4,6 mm C18 (Phenomenex; Torrance, CA, EE.UU.) empleando un gradiente de 2-80% de acetonitrilo (0,1% de TFA) durante 30 minutos. La HPLC analítica identificó un producto con una pureza del 50%. A continuación, el péptido se purificó en una HPLC preparativa equipada con una columna C18, usando un gradiente de elución similar. El producto purificado se volvió a analizar por HPLC para estudiar la pureza (97,40%) y espectrometría de
15 masas (5204,6 Dalton) y posteriormente se liofilizó. Después de la liofilización, se obtuvieron 6,2 mg de producto purificado que representaba un rendimiento del 1,2%.
Los compuestos GIP PEGilados descritos en este documento se pueden sintetizar sustancialmente según el procedimiento descrito para la síntesis del compuesto del Ejemplo 15, mediante el uso de PEG-maleimida como material de partida, en lugar de N-propilmaleimida utilizada en el Ejemplo 15.
20 Una persona de experiencia ordinaria en la técnica puede preparar otros péptidos de la presente memoria, usando procedimientos sintéticos análogos a los descritos en los ejemplos anteriores. Ensayos funcionales
A. Ensayo de unión del receptor de hGIP in vitro
Las membranas para los ensayos de unión del receptor in vitro se prepararon mediante homogeneización de las células clonales CHO-K1 que expresaban el receptor de GIP humano recombinante, con un aparato Brinkman Polytron (ajuste 6,15 s), Tris-HCI 50 mM enfriado con hielo y luego se sometieron a dos centrifugaciones a 39.000 g durante 10 minutos, con una resuspensión intermedia en tampón de nuevo aporte. Para el ensayo, se incubaron partes alícuotas de las preparaciones de membranas lavadas (100 minutos a 25°C con [125I]GIP 0,05 nM

(aproximadamente 2200 Ci/mmol) en Tris-HCl 50 mM, 0,1 mg/ml de bacitracina y 0,1% de BSA. El volumen final del ensayo fue de 0,5 ml. Las incubaciones se terminaron por filtración rápida a través de filtros GF/C (empapados previamente en polietilenimina al 0,5%) usando un colector de filtración Brandel. Cada tubo y filtro se lavó después tres veces con partes alícuotas de 5 ml de tampón enfriado con hielo. La unión específica se definió como el
5 radioligando total unido menos el unido en presencia de GIP 1000 nM. Los datos de unión in vitro del receptor de hGIP para los compuestos ejemplificados en la presente memoria, se proporcionan en la Tabla 2.
B. Ensayo de la semivida plasmática humana y de rata
El péptido GIP (50 µL, 1 mg/ml) se añadió a 450 µL de plasma (humano o de rata), se sometió brevemente a vórtex y se incubó a 37°C. Se retiraron 50 µL en diversos momentos, como al cabo de 0, 1, 2, 3, 4, 8, 24, 32, 48, 56, 72 10 horas, se mezclaron con 5 µL de ácido fórmico y 150 µL de acetonitrilo en un tubo de microcentrífuga, se sometieron a vórtex y se centrifugaron durante 10 minutos a 10K rpm. El material sobrenadante se transfirió a un vial de inyección y se analizó por LC-MS. El sistema LC-MS consistió en un espectrómetro de masas API4000 con una sonda ESI. Se utilizó el modo de iones positivos y detección con barrido completo. La separación por HPLC se llevó a cabo en una columna Luna de 3 μ C8 (2), de 2 x 30 mm con un gradiente desde 90% de A a 90% de B en 10
15 minutos, con un caudal de 0,3 ml/min. El tampón A era 1% de ácido fórmico en agua y el tampón B era 1% de acetonitrilo y ácido fórmico. Los datos de la semivida plasmática humana y de rata para los compuestos ejemplificados en la presente memoria, se proporcionan en la Tabla 2.

C. Determinación de la estimulación con AMP cíclico
Se sembraron 1 x 105 células CHO-K1 que expresaban el receptor de GIP recombinante humano o células de
insulinoma RIN-5F, durante la noche en placas de cultivo celular de 24 pocillos (Coming Incorporate, Corning, NY,
5 EE.UU.). Para el ensayo, las células se incubaron previamente en 500 µl de solución salina equilibrada con Hanks
(Sigma, St. Louis, MO, EE.UU.) con IBMX 0,55 mM (Sigma, St. Louis, MO, EE.UU.), ajustado a pH 7,3 durante 10
minutos. A continuación, se añadió GIP o sus análogos a una concentración de 100 nM. Después de una incubación
con hielo para detener la reacción. Se recogieron los contenidos de los pocillos, se centrifugaron a 2700 g durante 10 20 minutos a 4°C para eliminar los desechos celulares. Los niveles de AMPc en el material sobrenadante se
determinaron por radioinmunoensayo (New England Nuclear, Boston, MA, EE.UU.).

D. Determinación de la secreción de insulina in vivo en ratas normales
Ratas macho Sprague Dawley con un peso corporal de aproximadamente 275-300 g se utilizaron como sujetos experimentales. El día antes del tratamiento, se implantaron cánulas en la aurícula derecha a través de la vena yugular bajo clorhidrato. Cada cánula se llenó con 100 u/ml de solución salina con heparina y se sujetó. Las ratas se 5 mantuvieron en ayunas durante aproximadamente 18 horas antes de la dosificación con el compuesto o el vehículo (solución salina/0-25% de BSA). El día del experimento, se descongelaron partes alícuotas del compuesto, se llevaron a temperatura ambiente y se agitaron a fondo. Se realizó una revisión cuidadosa en busca de cualquier señal del compuesto partiendo de la solución. Diez minutos antes de la inyección de compuesto/glucosa, se recogió una muestra de 500 µl de sangre y se reemplazó con un volumen igual de solución salina heparinizada (10 u/ml). En 10 el tiempo 0, se recogió una muestra de 500 µl de sangre a través de la cánula. A continuación, se inyectó o bien el vehículo o la dosis apropiada de compuesto en la cánula y se presionó hacia dentro con la glucosa (1 g/kg) o la solución de vehículo. Finalmente, se utilizaron 500 µl de volumen de solución salina heparinizada (10 u/ml) para presionar la glucosa restante a través de la cánula. Muestras adicionales de sangre de 500 µl se recogieron al cabo de 2,5, 5, 10 y 20 minutos de la dosificación post-glucosa; cada una seguida inmediatamente por un bolo, inyección
15 iv de 500 µl de solución salina heparinizada (10 u/ml) a través de la cánula. El plasma se recogió de las muestras de sangre mediante centrifugación, y se almacenó a -20°C hasta el ensayo para estudiar el contenido en insulina.
La FIG. 1 muestra los efectos in vivo de los compuestos de los Ejemplos 1-7 y el GIP natural sobre la liberación de insulina de ratas Sprague Dawley. Los valores numéricos de la secreción de insulina total que se muestran en la FIG. 1 se resumen en la Tabla 3.
