Abstract
Método de selección de una célula eucariótica expresante de una inmunoglobulina, en el que el método comprende: a) transfectar una célula eucariótica con un ácido nucleico que comprende, en dirección 5' a 3', i) un primer ácido nucleico codificante de por lo menos un fragmento de un dominio CH3 o CH4 de cadena pesada de inmunoglobulina sin codón de parada traduccional dentro del marco, ii) un segundo ácido nucleico que se inicia con el sitio 5' donante de procesamiento de un dominio CH3 o CH4 de cadena pesada de inmunoglobulina y que termina en el sitio 3' aceptor de procesamiento del exón M1 de dominio transmembranal de cadena pesada de inmunoglobulina posterior que comprende un codón de parada traduccional dentro del marco y una señal de poliadenilación, iii) un tercer ácido nucleico codificante de: iiia) por lo menos un fragmento de un dominio transmembranal de inmunoglobulina, b) cultivar dicha célula transfectada bajo condiciones adecuadas a la producción de pre-ARNm de dicho ácido nucleico, procesar dicho pre-ARNm, y traducción de dicho ARNm procesado en una inmunoglobulina, en la que dicha célula transfectada produce inmunoglobulina soluble e inmunoglobulina unida a membrana plasmática mediante procesamiento alternativo de dicho pre-ARNm y c) seleccionar una célula con inmunoglobulina unida a membrana plasmática que será la célula expresante de una inmunoglobulina.
