Abstract

Método de elaboración de una cerveza que comprende la adición de una proteasa termoestable a un mosto después del filtrado pero antes del hervido del mosto, donde termoestable significa que la actividad de la proteasa es al menos el 70% de la actividad de referencia cuando es evaluada según el siguiente método: La proteasa es diluida a 1mg/ml en un tampón de ensayo que comprende 100 mM ácido succínico, 100 mM HEPES, 100 mM CHES, 100 mM CABS, 1 mM CaCl2, 150 mM KCL, 0.01% Tritón X-100, pH ajustado a 5.5 con NaOH. La proteasa es entonces preincubada en i) hielo, ii) 10 min a 70°C. El sustrato para el que la proteasa tiene actividad es suspendido en 0.01% Tritón X-100. Para iniciar la reacción, se añaden 20 μl al tubo y se incuba en un termomezclador Eppendorf a 70°C, 1400 r.p.m. durante 15 min. La reacción se detiene colocando los tubos en hielo. Las muestras son centrifugadas en frío a 14000 g durante 3 min, y se mide el OD590 en el sobrenadante. El valor OD590 de las muestras en blanco es sustraído de los valores correspondientes de las muestras tratadas con proteasa. La termoestabilidad de la proteasa se determina calculando la actividad porcentual de las muestras preincubadas a 70° en comparación con la actividad de referencia, que es la actividad de la proteasa de las muestras incubadas en hielo.
