Abstract

Die Erfindung betrifft ein optisches System, welches eine Lichtquelle (1), einen Detektor (6) und eine Probenaufnahme aufweist. Ein erster Strahlengang (2) verläuft von der Lichtquelle (1) zur Probenaufnahme und ein zweiter Strahlengang (5), der vom ersten Strahlengang (2) getrennt ist, verläuft von der Probenaufnahme zum Detektor (6). Das optische System weist zudem einen Filterträger (9, 14, 20, 21, 22, 23) auf, der mehrere Filter (10a, 10b, 11a, 11b, 12a, 12b, 13a, 13b) aufweist. Jeweils zwei Filter (10a, 10b, 11a, 11b, 12a, 12b, 13a, 13b) bilden ein Filterpaar. Der Filterträger (9, 14, 20, 21, 22, 23) ist so im optischen System angeordnet, dass der erste Strahlengang (2) durch einen Filter (10a, 11a, 12a, 13a) eines Filterpaars verläuft und der zweite Strahlengang (5) durch den anderen Filter (10b, 11b, 12b, 13b) desselben Filterpaars verläuft.

Die vorliegende Erfindung betrifft ein optisches System mit einem Filterträger, welcher mehrere in Filterpaaren zusammengeschlossene Filter aufweist sowie eine Verwendung des Filterträgers in einem optischen System.
Stand der Technik
In optischen Systemen werden im Betrieb einzelne Komponenten wie optische Filter automatisch getauscht. Hierfür werden mechanische Wechseleinheiten verwendet, welche die Filter vor einer Apertur auswechseln. Ein Beispiel hierfür sind Filterräder, bei denen die Filter auf einer drehbaren Scheibe angeordnet sind und vor die Apertur gedreht werden. In vielen optischen Systemen sind zwei Aperturen zweier unterschiedlicher Strahlengänge vorgesehen. Die beiden Filter werden abhängig voneinander gewählt und bilden ein Filterpaar. Eine triviale Lösung sieht vor, zwei Filterräder zu verwenden, wobei je ein Filterrad eine Apertur bedeckt und auf jedem Filterrad nur die Filter für die entsprechende Apertur angeordnet sind. Die beiden Filterräder können dann mit einem gemeinsamen Antrieb versehen sein.
In der US 6,313,960 B2 ist vorgesehen, alle Filter auf einem gemeinsamen Filterrad anzuordnen. Die Filter sind entlang eines Kreises um das Drehzentrum des Filterrads in gleichem Abstand zu diesem angeordnet. Die beiden Filter eines Filterpaars sind einander gegenüberliegend auf einer Linie mit dem Drehzentrum angeordnet. In einem Gehäuse sind die zwei Aperturen so angeordnet, dass diese jeweils um den Radius des Kreises der Filter vom Drehzentrum beabstandet sind und ebenfalls auf einer Linie mit dem Drehzentrum angeordnet sind. Ein Filterpaar besetzt somit gleichzeitig beide Aperturen. Das Filterrad wird hier für zwei identische Strahlengänge verwendet, Aus der Fluoreszenzmikroskopie sind zudem sogenannte „Filtercubes“ bekannt, die zwei Filter und einen dichroitischen Spiegel bzw. einen Strahlteiler aufweisen. Die Filtercubes sind auf einem Rad montiert, wodurch sich beide Filter gleichzeitig wechseln lassen. Ein Filter ist im Strahlengang von der Quelle bis zum dichroitischen Spiegel angeordnet und ein weiterer Filter ist im Strahlengang zwischen dem dichroitischen Spiegel und dem Detektor angeordnet. Vom dichroitischen Spiegel zur Probe wird ein gemeinsamer Strahlengang verwendet.
Offenbarung der Erfindung
Es wird ein optisches System vorgeschlagen, das eine Lichtquelle, einen Detektor und eine Probenaufnahme aufweist. Ein erster Strahlengang verläuft von der Lichtquelle zur Probenaufnahme und ein zweiter Strahlengang verläuft von der Probenaufnahme zum Detektor. Der erste und der zweite Strahlengang treffen sich nur an der Probenaufnahme, sind ansonsten aber räumlich getrennt. Das Licht von der Lichtquelle und das Licht zum Detektor laufen also entlang separater Pfade.
Das optische System weist zudem einen Filterträger auf, der mehrere optische Filter aufweist. Jeweils zwei Filter werden für eine jeweilige Anwendung zueinander passend gewählt und bilden ein Filterpaar. Die Filter des Filterpaars können vorzugsweise unterschiedliche Frequenzbereiche sperren. Der Filterträger ist so im optischen System angeordnet ist, dass der erste Strahlengang durch einen Filter eines Filterpaars verläuft und der zweite Strahlengang durch den anderen Filter desselben Filterpaars verläuft. Somit ist eine Seite des Filterpaars dem ersten Strahlengang von der Lichtquelle zur Probe zugeordnet und die andere Seite des Filterpaars ist dem zweiten Strahlengang von der Probe zum Detektor zugeordnet. Unter einer Seite der Filterpaare soll hierbei verstanden werden, dass diese Filter dem gleichen Strahlengang und somit der gleichen Apertur zugeordnet sind und kein gemeinsames Filterpaar untereinander bilden. Durch den Filterträger kann in einfacher Weise zwischen den Filterpaaren gewechselt werden und es werden jeweils die zusammengehörigen und zueinander passend gewählten Filter eines Filterpaars in die beiden komplett voneinander getrennten Strahlengänge gebracht. Insbesondere ist das optische System ein Fluoreszenzmesssystem. Ein Anregungslicht von der Lichtquelle verläuft entlang des ersten Strahlengangs und durch den ersten Filter des Filterpaars bis zur Probe. Der Filter kann ein Bandpassfilter sein, der nur das Anregungslicht passieren lässt. Das Anregungslicht regt eine Fluoreszenz in der Probe an, wodurch diese Fluoreszenzlicht abstrahlt. Das Fluoreszenzlicht wird in den zweiten Strahlengang eingeleitet und verläuft getrennt vom Anregungslicht entlang des zweiten Strahlengangs durch den zweiten Filter des Filterpaars zum Detektor, der das Fluoreszenzlicht aufnimmt. Der zweite Filter lässt das Fluoreszenzlicht passieren und sperrt die Frequenzen des Anregungslichts, sodass eventuell gestreutes Anregungslicht nicht in den Detektor dringt. Bei einem Fluoreszenzmesssystem können die Filterpaare abhängig vom jeweiligen Anregungslicht gewählt werden. Hierbei ist Verwendung des Filterträgers von Vorteil, da bei Änderung des Anregungslicht das Filterpaar in einfacher Weise gewechselt werden kann.
In einer Variante kann der Filterträger ein Filterrad sein. Das Filterrad ist bevorzugt eine Scheibe, insbesondere eine Kreisscheibe, und die Filter sind auf der Stirnseite angeordnet. Insbesondere weisen die Filter eine quadratische Form auf. Eine solche quadratische Form ist einfacher herzustellen als beispielsweise eine runde Form. Das Filterrad kann um ein Drehzentrum, welches sich bevorzugt im Mittelpunkt des Filterrads befindet, gedreht werden. Dadurch werden die Filter bewegt und können mit einer entsprechenden Apertur ausgerichtet werden. Alle Filter, die einer Seite der Filterpaare angehören, sind im gleichen Abstand zum Drehzentrum angeordnet. Dies gilt jeweils für beide Seiten der Filterpaare. Unter einer Seite der Filterpaare soll hierbei verstanden werden, dass diese Filter dem gleichen Strahlengang und somit der gleichen Apertur zugeordnet sind und kein gemeinsames Filterpaar untereinander bilden. Für jede Seite wird die entsprechende Apertur im selben Abstand wie die Filter zum Drehzentrum angeordnet, sodass die Filter dieser Seite mit der zugehörigen Apertur ausgerichtet werden können. Vorzugsweise sind die Filter zu ihren Nachbarn der gleichen Seite gleich weit beabstandet. Dadurch kann das Filterrad beim Wechseln der Filter um den gleichen Winkel gedreht werden.
Vorzugsweise weisen die beiden Filter eines Filterpaars indes nicht den gleichen Abstand zum Drehzentrum auf. Somit liegen die Filter des Filterpaars nicht auf einem Kreis mit gleichem Radius um das Drehzentrum, sondern können auf dem Filterrad verteilt werden. Alternativ oder zusätzlich kann vorgesehen sein, dass die beiden Filter eines Filterpaars und das Drehzentrum nicht auf einer gemeinsamen Geraden liegen. Somit sind die Filter des Filterpaars in Relation zum Drehzentrum versetzt angeordnet. Im Ergebnis wird auf beide Weisen die Fläche des Filterrads für die Anordnung der Filter besser ausgenutzt. Demnach kann der Durchmesser des Filterrads kleiner gewählt werden, was besonders hinsichtlich des geringen Bauraums, aber auch der geringeren Materialkosten von Vorteil ist. Eine Kombination der beiden Weisen verbessert die Ausnutzung der Fläche zusätzlich. Die Aperturen werden so angeordnet, dass sie bei zumindest einer Stellung des Filterrads mit einem Filterpaar ausgerichtet sind. Da, wie oben beschrieben, die Filter einer Seite der Filterpaare für jede der beiden Seiten im gleichen Abstand zum Drehzentrum angeordnet sind, werden die jeweiligen Filter durch Drehen des Filterrads mit den Aperturen ausgerichtet.
Eine bevorzugte Anordnung der Filter auf dem Filterrad stellen zwei konzentrische Kreise mit unterschiedlichen Radien um das Drehzentrum dar. Die Filter einer Seite der Filterpaare sind entlang eines Kreises angeordnet und die Filter der anderen Seite sind entlang des anderen Kreises angeordnet. Da die beiden Kreise unterschiedliche Radien aufweisen, weisen die Filter eines Filterpaars unterschiedliche Abstände zum Drehzentrum auf. Zusätzlich können die Filter so auf den Kreisen angeordnet sein, dass die Filter eines Filterpaars mit dem Drehzentrum keine gemeinsame Linie bilden.
Beispielsweise können acht Filter, die vier Filterpaare bilden, vorgesehen sein. Eine besonders bevorzugte Anordnung von vier Filterpaaren auf dem Filterrad stellt ein Quadrat dar, dessen Mittelpunkt im Drehzentrum des Filterrads liegt. Die Filter sind entlang der Seiten, einschließlich der Ecken, des Quadrats angeordnet und vorzugsweise äquidistant auf den Seiten verteilt. Insbesondere sind die Filter einer Seite der Filterpaare an den Ecken des Quadrats angeordnet. Somit verläuft einer der vorstehend beschriebenen Kreise durch die Ecken des Quadrats. Die Filter der anderen Seite der Filterpaare sind dann an jeweils einem Punkt auf der Seite des Quadrats, beispielsweise dem Mittelpunkt der jeweiligen Seite, angeordnet. Somit verläuft der andere Kreis durch diese Punkte. Das Quadrat bietet aufgrund seiner Symmetrie eine einfache Möglichkeit, die Filter eines Filterpaars in unterschiedlichen Abständen zum Drehzentrum anzuordnen und gleichzeitig die Filter einer Seite der Filterpaare im gleichen Abstand zum Drehzentrum anzuordnen. Vor allem bei quadratischen Filtern kann durch diese Anordnung gegenüber einer Anordnung entlang nur eines Kreises Fläche auf dem Filterrad eingespart werden. Die acht Filter können vorteilhaft entlang der Seiten des Quadrats angeordnet werden, indem jeweils einer der Filter des Filterpaars in einer Ecke des Quadrats angeordnet ist und der zugehörige andere Filter auf einer Seite des Quadrats, besonders bevorzugt im Mittelpunkt der Seite, angeordnet ist. Demnach sind alle Filter einer Seite der Filterpaare in den Ecken des Quadrats angeordnet und alle Filter der anderen Seite der Filterpaare auf den Seiten des Quadrats, besonders bevorzugt im Mittelpunkt der Seiten, angeordnet. Der Abstand vom Drehzentrum zu einer Ecke des Quadrats ist der Größte und aufgrund der Symmetrie für jede Ecke gleich. Der Abstand zu einer Seite des Quadrats und insbesondere zum Mittelpunkt der Seite ist geringer und kann aufgrund der Symmetrie ebenfalls für alle Seiten gleich gewählt werden. Zudem liegen ausgehend von einer Ecke nur die gegenüberliegende Ecke des Quadrats und keine anderen Punkte auf den Seiten des Quadrats auf einer gemeinsamen Linie mit dem Drehzentrum.
Beispielsweise können zehn Filter, die fünf Filterpaare bilden, vorgesehen sein, wobei fünf Filter einer Seite der Filterpaare entlang eines inneren Kreises auf einem Fünfeck angeordnet sind und weitere fünf Filter der anderen Seite der Filterpaare entlang eines äußeren Kreises auf einem Fünfeck angeordnet sind. Somit bilden die fünf Filter auf dem inneren Kreis ein Fünfeck und die fünf Filter auf dem äußeren Kreis bilden ein weiteres Fünfeck, welches das erstgenannte Fünfeck umgibt.
Verallgemeinert gilt, dass bei n (n e N) Filterpaaren die Filter vorzugsweise auf n- Ecken angeordnet sind.
In einer weiteren Variante kann der Filterträger ein Filterschieber sein, auf dem alle Filter angeordnet sind. Der Filterschieber ist entlang einer Geraden in dem optischen System verschiebbar. Die Filter jeder Seite des Filterpaars sind in Reihen hintereinander angeordnet. Dabei weisen die Filter jeweils den gleichen Abstand zueinander auf, und zwar für jede Seite des Filterpaars und innerhalb der Filterpaare. Die Reihen jeder Seite des Filterpaars sind jeweils in Richtung der Geraden, entlang der der Filterschieber verschoben wird, angeordnet. Durch den Filterschieber können die Filter eines Filterpaars gemeinsam verschoben werden und in die beiden Strahlengänge gebracht werden. Da die Filter alle auf demselben Filterschieber angeordnet sind, muss beim Wechseln nur dieser eine Schieber bewegt werden.
In einer weiteren Variante kann der Filterträger mehrere in Richtung der Strahlengänge hintereinander angeordnete Einzelschieber aufweisen, die gemeinsam als ein Filterträger fungieren und auch als solcher angesehen werden. In jedem Einzelschieber sind nur die beiden Filter eines einzelnen Filterpaars angeordnet. Außerdem sind in jedem Einzelschieber zwei Öffnungen vorgesehen. Jeder Einzelschieber kann eine erste Position geschoben werden, in der die Filter des Filterpaars in die beiden Strahlengänge gebracht werden. Zudem kann jeder Einzelschieber in eine zweite Position geschoben werden, in der die beiden Öffnungen in den Strahlengängen angeordnet sind. Der Einzelschieber mit dem gewählten Filterpaar wird in die erste Position geschoben und die anderen Einzelschieber werden in die zweite Position geschoben, sodass die Strahlengänge durch die Öffnungen der anderen Einzelschieber verlaufen und nur das gewählte Filterpaar durchlaufen. Die Einzelschieber bieten zudem den Vorteil, dass mehrere Filterpaare gleichzeitig in die beiden Strahlengänge gebracht werden können. Somit können beispielsweise ein Hochpassfilter und ein Tiefpassfilter gemeinsam verwendet werden.
Kurze Beschreibung der Zeichnungen
Ausführungsbeispiele der Erfindung sind in den Zeichnungen dargestellt und in der nachfolgenden Beschreibung näher erläutert.
Figuren 1 a und b zeigen jeweils eine schematische Darstellung einer ersten Ausführungsform des erfindungsgemäßen optischen Systems mit einem Filterrad.
Figur 2 zeigt eine schematische Darstellung einer zweiten Ausführungsform des erfindungsgemäßen optischen Systems mit einem gemeinsamen Filterschieber.
Figur 3 zeigt eine schematische Darstellung einer dritten Ausführungsform des erfindungsgemäßen optischen Systems mit mehreren Einzelschiebern.
Ausführungsbeispiele der Erfindung In den Figuren 1 bis 3 sind mehrere optische Filter 10a, 10b, 11a, 11b, 12a, 12b, 13a, 13b dargestellt. Die mit „a“ gekennzeichneten Filter 10a, 11a, 12a, 13a bilden zusammen mit den entsprechenden mit „b“ gekennzeichneten Filtern 10b, 11b, 12b, 13b mit gleicher Nummer Filterpaare 10a & 10b, 11a & 11b, 12a & 12b, 13a & 13b. Die Filter desselben Filterpaars 10a & 10b, 11a & 11b, 12a & 12b, 13a & 13b sind komplementär ausgebildet. Die mit „a“ gekennzeichneten Filter 5a, 6a, 7a, 8a, 9a, welche einer Seite der Filterpaare angehören werden fortan als „erste Filter“ bezeichnet. Entsprechend werden die mit „b“ gekennzeichneten Filter 5b, 6b, 7b, 8b, 9b, welche der anderen Seite der Filterpaare angehören fortan als „zweite Filter“ bezeichnet. Die hier dargestellten optischen System sind Fluoreszenzmesssysteme und dienen zur der Fluoreszenzspektroskopie. Hierfür sind die ersten Filter 10a, 11a, 12a, 13a als Bandpassfilter ausgebildet sein, die nur das Anregungslicht passieren lassen, und die zweiten Filter 10b, 11b, 12b, 13b können als Hochpass- oder Tiefpassfilter ausgebildet sein, die das Anregungslicht sperren und nur das Fluoreszenzlicht passieren lassen.
In den Figuren 1 a und b ist jeweils eine erste Ausführungsform des optischen Systems zur Fluoreszenzspektroskopie dargestellt. Eine breitbandige Lichtquelle 1, beispielsweise eine weiße LED, eine phosphorbasierte Lichtquelle oder eine Gasentladungslampe, sendet ein Anregungslicht aus. Das Anregungslicht verläuft entlang eines ersten Strahlengangs 2 zu einem Spiegel 3 und wird von diesem in Richtung einer Probe 4 umgelenkt. Die Probe 4 wird in einer nicht dargestellten Probenaufnahme gehalten. Die Probe 4 wird durch das Anregungslicht angeregt und gibt ein Fluoreszenzlicht entlang eines zweiten Strahlengangs 5 ab. Das Fluoreszenzlicht verläuft entlang des zweiten Strahlengangs 5 zu einem Detektor 6 und wird von diesem empfangen. Der Detektor 6 ist eingerichtet, zumindest ausgewählte Nachweisbänder zu detektieren. Vorzugsweise wird hier ein Halbleiterdetektor, insbesondere eine Photodiode verwendet. In Figur 1 a lenkt der Spiegel 3 das Anregungslicht seitlich in die Probe 4, sodass das Anregungslicht entlang des ersten Strahlengangs 2 von einer anderen Seite eingestrahlt wird als das Fluoreszenzlicht entlang des zweiten Strahlengangs 5 abgegeben wird. In Figur 1 b lenkt der Spiegel 3 das Anregungslicht entlang des ersten Strahlengangs 2 über die gleiche Seite wie das Fluoreszenzlicht entlang des zweiten Strahlengangs 5 abgegeben wird in die Probe 4. In beiden Fällen kreuzen sich der erste Strahlengang 2 und der zweite Strahlengang 5 nur in der Probe 4. Das optische System kann verschiedene optische Elemente aufweisen, die das Licht formen und lenken. Die genaue Ausgestaltung hängt von der Abstrahlcharakteristik der Lichtquelle 1, der aktiven Fläche des Detektors 6 sowie des generellen Proben- und Gerätekonzepts ab. Als Beispiele können Linsen, Spiegel, Umlenkprismen, Hohlspiegel, Wellenleiter, holografische oder diffraktive optische Elemente eingesetzt werden. Im ersten Strahlengang 2 ist eine erste Apertur 7 vorgesehen und im zweiten Strahlengang 5 ist eine zweite Apertur 8 vorgesehen. Das Licht verläuft jeweils kulminiert durch die Aperturen 7, 8.
Im ersten Ausführungsbeispiel gemäß Figur 1 ist vorgesehen, die Filter 10a, 10b, 11a, 11b, 12a, 12b, 13a, 13b auf einem Filterrad 9 anzuordnen, um die Aperturen 7, 8 abzudecken. Das Filterrad 9 ist eine Kreisscheibe und um ein Drehzentrum 2 in dessen Mittelpunkt drehbar gelagert. Um das Filterrad 9 zu drehen, weist das optische System eine hier nicht dargestellte mechanische oder elektromechanische Vorrichtung auf, mit der die Drehung extern gesteuert werden kann. Wie nachfolgend im Detail erläutert, werden die Filter 10a, 10b, lla, 11b, 12a, 12b, 13a, 13b mit den Aperturen 7, 8 ausgerichtet und decken diese ab. Dabei deckt nur ein Filterpaar 10a & 10b, 11a & 11b, 12a & 12b, 13a & 13b gleichzeitig die beiden Aperturen 7, 8 ab. Die Filterpaare 10a & 10b, 11a & llb, 12a & 12b, 13a & 13b sind in diesem Ausführungsbeispiel entlang eines Quadrats, dessen Mittelpunkt im Drehzentrum des Filterrads 9 liegt, angeordnet. Die ersten Filter 10a, 11a, 12a, 13a sind in den Ecken des Quadrats Q angeordnet. Durch die Ecken des Quadrats Q verläuft ein nicht dargestellter erster konzentrischer Kreis mit dem Drehzentrum des Filterrads 9 als Mittelpunkt, sodass die ersten Filter 10a, 11a, 12a, 13a auf dem ersten Kreis angeordnet sind. Die zweiten Filter 10b, 11b, 12b, 13b des zugehörigen Filterpaars 10a & 10b, 11a & 11b, 12a & 12b, 13a & 13b sind in den Mittelpunkten der gegenüberliegenden Seiten des Quadrats angeordnet. Es wird ausgehend von der Ecke stets dieselbe gegenüberliegende Seite, z. B. die linke gegenüberliegende Seite gewählt. Durch diese Mittelpunkte verläuft ein ebenfalls nicht dargestellter zweiter konzentrischer Kreis, sodass die zweiten 10b, 11b, 12b, 13b auf dem zweiten Kreis angeordnet sind. Der Radius des zweiten Kreises ist kleiner als der Radius des ersten Kreises, dementsprechend ist der Abstand vom Drehzentrum des Filterrads 9 zu den ersten Filtern 10a, 11a, 12a, 13a größer ist als der Abstand vom Drehzentrum zu den zweiten Filtern 10b, 11b, 12b, 13b. Die zweite Apertur 8 ist so angeordnet, dass sie auf eine Ecke (in Figur 1 z. B. die rechte Ecke) des Quadrats fällt, und die erste Apertur 7 ist so angeordnet, dass sie auf den Mittelpunkt der gegenüberliegenden Seite des Quadrats fällt. Die Aperturen 7, 8 können auch andersherum angeordnet sein. Da das Drehzentrum des Filterrads 9 und der Mittelpunkt des Quadrats zusammenfallen, werden bei der Drehung des Filterrads 9 aufgrund der Symmetrie des Quadrats die ersten Filter 10a, 11a, 12a, 13a in den Ecken des Quadrats die erste Apertur 7 durchlaufen und entsprechend die zugehörigen zweiten Filter 10b, 11b, 12b, 13b desselben Filterpaars 10a & 10b, 11a & 11b, 12a & 12b, 13a & 13b auf den Seiten des Quadrats gleichzeitig die zweite Apertur 8 durchlaufen. Die Filter 10a, 11a, 12a, 13a und 10b, 11b, 12b, 13b sind jeweils im gleichen Abstand zueinander angeordnet, sodass die Drehung für den Wechsel des Filterpaars 10a & 10b, 11a & 11b, 12a & 12b, 13a & 13b jeweils um den gleichen Winkel stattfindet. Die Ecken des Quadrats und die Mittelpunkte der Seiten des Quadrats weisen unterschiedliche Abstände zum Drehzentrum des Filterrads 9 auf und liegen zudem nicht auf einer gemeinsamen Geraden mit dem Drehzentrum.
Es können aber andere Geometrien für die Filter des Filterrads, wie z. B. Fünfecke oder generell n-Ecke vorgesehen sein.
In den Figuren 2 und 3 ist eine zweite bzw. dritte Ausführungsform des optischen Systems zur Fluoreszenzspektroskopie dargestellt. Die optischen Systeme gemäß der zweiten Ausführungsform und gemäß der dritten Ausführungsform unterscheiden sich von der ersten Ausführungsform nur durch den Filterträger. Für den übrigen Aufbau wird auf obenstehende Beschreibung verwiesen. Dabei kann das optische System hinsichtlich des Spiegels 3 und dem Verlauf des ersten Strahlengangs 2 wie in Figur la oder wie in Figur 1 b dargestellt ausgebildet sein.
Im zweiten Ausführungsbeispiel gemäß Figur 2 ist vorgesehen, die Filter 10a, 10b, 11a, 11b, 12a, 12b, 13a, 13b auf einem Filterschieber 14 anzuordnen, um die Aperturen 7, 8 abzudecken. Der Filterschieber 14 kann entlang der Verschieberichtung 14 verschoben werden. Die Filter 10a, 11a, 12a, 13a einer Seite des Filterpaars 10a & 10b, 11a & 11b, 12a & 12b, 13a & 13b sind in einer Reihe 16 hintereinander in Verschieberichtung 15 angeordnet. Ebenso sind die Filter 10b, 11b, 12b, 13b der Seite des Filterpaars 10a & 10b, 11a & 11b, 12a & 12b, 13a & 13b in einer Reihe 17 hintereinander in Verschieberichtung 15 angeordnet, und zwar in der gleichen Reihenfolge wie der zugehörige Filter des Filterpaars 10a & 10b, 11a & 11b, 12a & 12b, 13a & 13b in der anderen Reihe 16. Der Abstand zwischen den Reihen 16, 17 ist konstant und die Abstände zwischen den Filtern 10a, 11a, 12a, 13a und 10b, 11b, 12b, 13b in den jeweiligen Reihen 16 und 17 sind gleich. Die Aperturen 7, 8 sind über den beiden Reihen 16, 17 angeordnet und weisen den gleichen Abstand zueinander wie die Reihen 16, 17 auf. Dabei ist die erste Apertur 7 über der Reihe 16 der Filter 10a, 11a, 12a, 13a der einen Seite der Filterpaare 10a & 10b, 11a & 11b, 12a & 12b, 13a & 13b angeordnet und die zweite Apertur 8 ist über der Reihe 17 der Filter 10b, 11b, 12b, 13b der anderen Seite der Filterpaare 10a & 10b, 11a & 11b, 12a & 12b, 13a & 13b angeordnet Durch Verschieben des Filterschiebers 14 entlang der Verschieberichtung 15 bedeckt ein Filterpaar 10a & 10b, 11a & 11b, 12a & 12b, 13a & 13b gleichzeitig beide Aperturen 7, 8.
In der dritten Ausführungsform gemäß Figur 3 sind mehrere Einzelschieber 20, 21, 22, 23 vorgesehen, die gemeinsam als ein Filterträger fungieren. Die Einzelschieber 20, 21, 22, 23 sind in Richtung des ersten Strahlengangs 2 und in Richtung des zweiten Strahlengangs 5 hintereinander angeordnet. Ein erster Einzelschieber 20 weist ein erstes Filterpaar 10a, 10b auf, ein zweiter Einzelschieber 21 weist ein zweites Filterpaar 11a, 11b auf, ein dritter Einzelschieber 22 weist ein drittes Filterpaar 12a, 12b auf und ein vierter Einzelschieber 23 weist ein viertes Filterpaar 13a, 13b auf. Jeder Einzelschieber kann zwischen einer ersten Position, in der die beiden Filter des entsprechenden Filterpaars 10a & 10b, 11a & 11b, 12a & 12b, 13a & 13b in den Strahlengängen 2, 5 platziert sind, und einer zweiten Position, in der die beiden Öffnungen 24, 25 in den beiden Strahlengängen 2, 5 platziert sind, entlang einer Verschieberichtung werden. Die Einzelschieber 20, 21, 22, 23 können unabhängig voneinander verschoben werden. Um die Einzelschieber 20, 21, 22, 23 zu verschieben, weist das optische System eine hier nicht dargestellte mechanische oder elektromechanische Vorrichtung auf, mit der die Verschiebung extern gesteuert werden kann. Die Filter jedes Filterpaare 10a & 10b, 11a & 11b, 12a & 12b, 13a & 13b weisen jeweils den gleichen Abstand zueinander auf. Jeder Einzelschieber 20, 21, 22, 23 weist zudem eine Öffnung 24, die in Verschieberichtung 26 neben einem Filter 10a, 11a, 12a, 13a einer Seite der Filterpaare 10a & 10b, 11a & 11b, 12a & 12b, 13a & 13b angeordnet ist, und eine Öffnung 25, die in Verschieberichtung 26 neben einem Filter 10b, 11b, 12b, 13b der anderen Seite der Filterpaare 10a & 10b, 11a & 11b, 12a & 12b, 13a & 13b angeordnet ist, auf. Die Öffnungen 24, 25 weisen zueinander den gleichen Abstand wie die Filter jedes Filterpaare 10a & 10b, 11a & 11b, 12a & 12b, 13a & 13b auf und sind von den jeweiligen Filtern 10a, 10b, 11a, 11b, 12a, 12b, 13a, 13b gleich weit in dieselbe Richtung versetzt. Die Öffnungen 24 sind so ausgebildet, dass die der erste Strahlengang 2 ungehindert durch diese verläuft, und die Öffnung 25 ist so ausgebildet, dass der zweite Strahlengang 5 ungehindert durch diese verläuft, wenn der zugehörige Einzelschieber 20, 21, 22,
23 in der zweiten Position die Öffnungen 24, 25 in die Strahlengänge 2, 5 verschiebt. Die Öffnungen 24, 25 und die Filter (10a, 10b, 11a, 11b, 12a, 12b, 13a, 13b) dienen hierbei als Apertur. In Figur 3 ist beispielsweise der dritte Einzelschieber 22 in der ersten Position und die anderen Einzelschieber 20, 21, 23 sind in der zweiten Position. Das Anregungslicht durchläuft also entlang des ersten Strahlengangs 2 nacheinander die Öffnung 24 des ersten Einzelschiebers 20, die Öffnung 24 des zweiten Einzelschieber 21, den Filter 12a des dritten Einzelschiebers 22 und die Öffnung 24 des vierten Einzelschiebers 23. Gleichermaßen durchläuft das Fluoreszenzlicht entlang des zweiten Strahlengangs 5 die Öffnung 25 des vierten Einzelschiebers 23, den Filter 12b des dritten Einzelschiebers 22, die Öffnung 25 des zweiten Einzelschiebers 21 und die Öffnung 25 des ersten Einzelschiebers 20.

Claims

Ansprüche
1. Optisches System aufweisend eine Lichtquelle (1), einen Detektor (6) und eine Probenaufnahme, wobei ein erster Strahlengang (2) von der Lichtquelle (1) zur Probenaufnahme verläuft und ein zweiter Strahlengang (5), der vom ersten Strahlengang (2) getrennt ist, von der Probenaufnahme zum Detektor (6) verläuft, gekennzeichnet durch einen Filterträger (9, 14, 20, 21, 22, 23), der mehrere Filter (10a, 10b, 11a, 11b, 12a, 12b, 13a, 13b) aufweist, wobei jeweils zwei Filter (10a, 10b, 11a, 11b, 12a, 12b, 13a, 13b) ein Filterpaar bilden und wobei der Filterträger (9, 14, 20, 21, 22, 23) so im optischen System angeordnet ist, dass der erste Strahlengang (2) durch einen Filter (10a, 11a, 12a, 13a) eines Filterpaars verläuft und der zweite Strahlengang (5) durch den anderen Filter (10b, 11b, 12b, 13b) desselben Filterpaars verläuft.
2. Optisches System nach Anspruch 1, dadurch gekennzeichnet, dass das optische System ein Fluoreszenzmesssystem ist, wobei ein Anregungslicht entlang des ersten Strahlengangs (2) durch den einen Filter (10a, 11a, 12a, 13a) des Filterpaars verläuft und ein Fluoreszenzlicht getrennt vom Anregungslicht entlang des zweiten Strahlengangs (5) durch den anderen Filter (10b, 11b, 12b, 13b) des Filterpaars verläuft.
3. Optisches System nach einem der Ansprüche 1 oder 2, dadurch gekennzeichnet, dass die Filter (10a, 10b, 11a, 11b, 12a, 12b, 13a, 13b) eines Filterpaars unterschiedliche Frequenzbereiche sperren.
4. Optisches System nach einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass der Filterträger ein Filterrad (9) ist, wobei das Filterrad (9) um ein Drehzentrum drehbar gelagert ist.
5. Optisches System nach Anspruch 4, dadurch gekennzeichnet, dass die Filter (10a, 10b, 11a, 11b, 12a, 12b, 13a, 13b) eines Filterpaars unterschiedliche Abstände zum Drehzentrum aufweisen und/oder die Filter (10a, 10b, 11a, 11b, 12a, 12b, 13a, 13b) eines Filterpaars und das Drehzentrum nicht auf einer gemeinsamen Geraden liegen. Optisches System nach Anspruch 4, dadurch gekennzeichnet, dass die Filter (10a, 10b, 11a, 11b, 12a, 12b, 13a, 13b) entlang zweier konzentrischer Kreise um das Drehzentrum angeordnet sind. Optisches System nach einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass der Filterträger ein Filterschieber (14) ist, wobei der Filterschieber (14) entlang einer Geraden verschiebbar ist, wobei die Filter (10a, 10b, 11a, 11b, 12a, 12b, 13a, 13b) jeder Seite des Filterpaars in Reihen in Richtung der Geraden angeordnet sind. Optisches System nach einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass der Filterträger mehrere in Richtung der Strahlengänge hintereinander angeordnete Einzelschieber (20, 21, 22, 23) aufweist, wobei in jedem Einzelschieber (20, 21, 22, 23) ein Filterpaar (10a, 10b, 11a, 11b, 12a, 12b, 13a, 13b) und zwei Öffnungen (24, 25) vorgesehen sind und die Einzelschieber (20, 21, 22, 23) jeweils in eine erste Position, in der die Filter (10a, 10b, 11a, 11b, 12a, 12b, 13a, 13b) des Filterpaars in den Strahlengängen (2, 5) angeordnet sind, und in eine zweite Position, in der beiden Öffnungen (24, 25) in den Strahlengängen (2, 5) angeordnet sind, verschiebbar sind. Verwendung eines Filterträgers (9, 14, 20, 21, 22, 23) in einem optischen System mit einer Lichtquelle (1), einem Detektor (6) und einer Probenaufnahme, wobei ein erster Strahlengang (2) des optischen Systems von der Lichtquelle (1) zur Probenaufnahme verläuft und ein zweiter Strahlengang (5) des optischen Systems, der vom ersten Strahlengang (2) getrennt ist, von der Probenaufnahme zum Detektor (6) verläuft, dadurch gekennzeichnet, dass der Filterträger (9, 14, 20, 21, 22, 23) mehrere Filter (10a, 10b, 11a, 11b, 12a, 12b, 13a, 13b) aufweist, wobei jeweils zwei Filter (10a, 10b, 11a, 11b, 12a, 12b, 13a, 13b) ein Filterpaar bilden und wobei der Filterträger (9, 14, 20, 21, 22, 23) so im optischen System angeordnet ist, dass der erste Strahlengang (2) durch einen Filter (10a, 11a, 12a, 13a) eines Filterpaars verläuft und der zweite Strahlengang (5) durch den anderen Filter (10b, 11b, 12b, 13b) desselben Filterpaars verläuft.
