Abstract
Die vorliegende Erfindung betrifft eine Oralgel-Zusammensetzung zum Auftrag auf das Zahnfleisch und zum Verbleib auf diesem für mindestens 3 Minuten sowie ihre Verwendung. Um ein Produkt für die Oralpflege und insbesondere zur wirksamen Behandlung (d.h. zur Reduzierung und Verhinderung) von Parodontitis/Gingivitis und Zahnfleischrückgang zur Verfügung zu stellen, wird im Rahmen der Erfindung vorgeschlagen, dass die Oralgel-Zusammensetzung mindestens 0,00001 und weniger als 0,5 Gew.-% Coffein und einen Gelbildner enthält. Es wurde unerwartet festgestellt, dass durch die erfindungsgemäße Oralgel-Zusammensetzung eine Aktivierung des Zellstoffwechsels und somit eine wirksame Behandlung und/oder Prävention von Parodontitis/Gingivitis sowie von Zahnfleischrückgang möglich ist.
Description
BESCHREIBUNG
Oralgel-Zusammensetzung und ihre Verwendung
Die vorliegende Erfindung betrifft eine Oralgel-Zusammensetzung zum Auftrag auf das Zahnfleisch und zum Verbleib auf diesem für mindestens 3 Minuten sowie ihre Verwendung.
Schon allein wegen der weltweit immer kohlenhydratreicheren Ernährung gewinnt die Zahn- und Zahnfleischpflege immer mehr an Bedeutung. So wird neben ästhetischen Aspekten zunehmend besonderer Wert auf die Vorsorge gelegt, wobei hier vor allem die Verminderung oder gar Vermeidung von Plaque, Karies, und/oder Halitosis (Mundgeruch) sowie ein gesundes Zahnfleisch im Mittelpunkt stehen.
Das Zahnfleisch zeichnet sich unter anderem dadurch aus, dass es die Zähne zervikal umgibt. Dabei umschließt das Zahnfleisch den Zahnhals, wodurch die Eintrittsstelle des Zahnes in den Kieferknochen der Mundhöhle abgedichtet wird. Das Zahnfleisch dient somit unter anderem dem Schutz und als Halterung des Zahnes.
Die verschiedenen Teile eines natürlichen Zahnes sind Zahnkrone, Zahnhals und Zahnwurzel, wobei diese aus mehreren Schichten aufgebaut sind. Von diesen Schichten sieht man normalerweise nur den außenliegenden Zahnschmelz (Enamelum), der das Dentin und weitere Schichten umschließt. Um beispielsweise Nahrung ohne Schaden an den Zähnen zerbeißen oder zermahlen zu können, ist der Zahnschmelz sehr hart. Er besteht zu etwa 97 Gew.% aus Hydroxylapatit, welcher die Summenformel Ca5(P04)3(0H) aufweist. Das Dentin gilt auch als Zahnhartsubstanz und besteht zu etwa zwei Dritteln ebenfalls aus Hydroxylapatit. Dentin enthält neben Hydroxylapatit aber auch noch Proteine und Wasser und ist aus diesem Grund nicht so hart wie der Zahnschmelz.
Häufige Zahn- sowie Zahnfleischerkrankungen wie Karies und Parodontitis/Gingivitis können auf der Bildung von bakteriellen Mikrofilmen basieren und/ oder auf bakteriellen Entzündungen beruhen. Weiterhin ist das Problem des
Zahnfleischrückgangs weitverbreitet und tritt insbesondere mit zunehmendem Lebensalter sehr häufig auf.
Die DE 29806937 U1 beschreibt eine Zahncreme, der eine oder mehrere anregende Substanzen zugefügt sind, die beispielsweise Coffein oder Nicotin sein können.
Die DE 201 19966 U1 beschreibt ebenfalls eine mit belebenden Stoffen angereicherte Zahnpasta, wobei Coffein und/oder Teein oder andere stimulierende Stoffe in der Zahnpastenrezeptur enthalten sind.
Eine coffeinhaltige Zahncreme ist auch aus der JP 2010/275261 A1 bekannt.
Auch aus der Veröffentlichung „Neurophysiological effect of flavor and caffeine added to toothpaste“, Sangyo Eiseigaku Zasshi, Vol. 52, Issue 4, Pages 172-181, 2010:1478424 CAN 156:675063 ist es bekannt, Zahnpasta zur Reduzierung von Müdigkeit unter anderem mit Coffein anzureichern.
Aus der WO 2021/013283 A1 sind Zusammensetzungen für Mundhygienemittel, insbesondere Zahnpasten, bekannt, die Coffein und lysinhaltige Bitterblocker enthalten. Da Mundhygienemittel in der Regel nicht geschluckt, sondern ausgespuckt werden, muss das Coffein, damit es wirksam ist, in einer ausreichend hohen Konzentration vorliegen. Die Bitterblocker dienen dazu, auch bei diesen sehr hohen Coffeinkonzentrationen noch ein angenehmes Geschmackserlebnis zu erzielen.
Schließlich wird auch von Johnson & Johnson eine coffeinhaltige Mundspülung unter der Bezeichnung „LISTERINE GREEN TEA“ angeboten.
Die Verweildauer von Zahnpasta bzw. Mundspülung/Mundwasser im Mundraum beschränkt sich jedoch auf ca. 30 Sekunden bis maximal 3 Minuten, so dass während der Dauer der Zahnpflege nur wenig Coffein mit dem Zahnfleisch interagieren kann. Anschließend werden diese in der Regel ausgespuckt und in der Regel auch mit Wasser ausgespült. Dies führt dazu, dass die effektive Konzentration
an Coffein in der Mundhöhle sehr schnell abnimmt. Derartige Mundhygienemittel dienen der Pflege, der Befeuchtung und dem Abtöten von Bakterien, haben jedoch keine langfristige positive Wirkung auf die Zellen des Zahnfleisches.
Aus der DE 102019 119589 A1 ist ein Mundhygienemittel enthaltend Coffein und einen Bitterblocker bekannt, wobei der Bitterblocker aus 8 % L-Lysin Hydrochlorid und 92 % Maltodextrin besteht und die Konzentration von Coffein pro Portion des Mundhygienemittels zwischen 5 und 200 mg liegt.
Die US 2013/0129641 A1 beschreibt Zusammensetzungen zur Kariesprophylaxe mit einer Calciumquelle und einer Phosphatquelle, welche einen pH-Wert von 6,0 bis 8,5 hat. Diese Zusammensetzungen enthalten Theobromin.
Der vorliegenden Erfindung liegt die Aufgabe zugrunde, ein Produkt für die Oralpflege und insbesondere zur wirksamen Behandlung (d.h. zur Reduzierung und Verhinderung) von Parodontitis/Gingivitis und Zahnfleischrückgang zur Verfügung zu stellen.
Weiterhin soll bei der Verwendung des Oralgels das ökologische Gleichgewicht im Mundraum nicht signifikant gestört und/oder eine Zahnverfärbung oder Geschmacksstörung riskiert werden.
Diese Aufgaben werden erfindungsgemäß dadurch gelöst, dass die Oralgel- Zusammensetzung a) mindestens 0,00001 und weniger als 0,5 Gew.-% Coffein, und b) einen Gelbildner enthält.
Coffein ist ein Methylxanthin, welches sich in seiner Wirkung überraschenderweise hinsichtlich der Zahnfleischregeneration als anderen Methylxanthinen, wie Theobromin oder Theophyllin, überlegen herausgestellt hat. Insbesondere das subjektive Anwendungsempfinden von coffeinbasiertem Zahngel, was den Geschmack, die Wirkung auf das Zahnfleisch, die Verringerung von
Zahnfleischbluten hat sich in einer Anwendungsstudie mit 50 Teilnehmern als signifikant besser erwiesen als das von theobromin- oder theophyllinbasiertem Zahngel. Zudem wurden in dieser Anwendungsstudie Coffein-Konzentrationen von weniger als 0,5 Gew.-% überraschenderweise besser bewertet als höhere Coffein- Konzentrationen (von mehr als 0,5 %).
Anders als ein Zahngel wird das erfindungsgemäße Oralgel nicht auf die Zähne, sondern auf das Zahnfleisch aufgetragen. Es könnte daher auch als Zahnfleischgel bezeichnet werden, da es primär der Pflege und Behandlung des Zahnfleischs dient.
Im Gegensatz zu den coffeinhaltigen Mundpflegeprodukten gemäß dem Stand der Technik ist die Verweilzeit des erfindungsgemäßen Oralgels im Mundraum um ein Vielfaches länger, da es nicht ausgespült wird. Dadurch wird eine längere Interaktion des Coffeins mit dem Zahnfleisch erzielt. Darüber hinaus wird das erfindungsgemäße Oralgel nicht ausgespuckt, sondern über den Speichel langsam aufgelöst, so dass sich eine vorteilhaftere Freisetzungskinetik ergibt als bei den Mundpflegeprodukten gemäß dem Stand der Technik. Es handelt sich somit um ein sogenanntes „leave- on“-Produkt. Zudem enthält das erfindungsgemäße Oralgel keine Putzkörper, was gerade bei akut gereiztem Zahnfleisch eine schonende Behandlung ermöglicht.
Eine kleinere Menge Coffein als bei den nur kurzzeitig einwirkenden Mundpflegeprodukten ist daher vorteilhafter. Es hat sich im Rahmen der Erfindung überraschenderweise herausgestellt, dass es vorteilhaft ist, wenn kleinere Coffein- Mengen (< 0,5 Gew.-%) über einen längeren Zeitraum zur Verfügung stehen, weil sich hierdurch ein „Depoteffekt“ ergibt. Nachteilig bei höheren Coffein- Konzentrationen ist, dass zellschädigende Wirkungen auftreten können.
Bei dem erfindungsgemäßen Oralgel beträgt die Verweildauer auf dem Zahnfleisch zwischen 3 Minuten und 72 Stunden, vorzugsweise zwischen 5 Minuten und 48 Stunden, insbesondere zwischen 10 min und 24 Stunden. Derart lange Verweilzeiten werden mit keinem der bekannten Mundpflegeprodukte erreicht.
Es wurde unerwartet festgestellt, dass durch die erfindungsgemäße Oralgel-
Zusammensetzung eine Aktivierung des Zellstoffwechsels und somit eine wirksame Behandlung und/oder Prävention von Parodontitis/Gingivitis sowie von Zahnfleischrückgang möglich ist. Die erfindungsgemäße Oralgel-Zusammensetzung kann im Rahmen der vorliegenden Erfindung sowohl kosmetische als auch medizinische Pflegeprodukte umfassen.
Des Weiteren wurde festgestellt, dass durch die Verwendung der erfindungsgemäßen Oralgel-Zusammensetzung das Anhaften des Coffeins direkt an dem Zahnfleisch bzw. dem Zahnfleischsaum unterstützt wird, so dass das Coffein über einen längeren Zeitraum in direktem Kontakt hiermit steht, wodurch eine hohe Absorptionsrate erreicht wird.
Hierbei wird das Coffein je nach Anwendergruppe unterschiedlich dosiert. Es soll eine Tagesdosis von 0,3 mg Coffein/kg Körpergewicht (EFSA NDA Panel (EFSA Panel on Dietetic Products, Nutrition and Allergies, 2015, Scientific opinion on the safety of caffeine, EFSA Journal 2015; 13 (5); 4102, 120 pp. Doi:10.2903/j.efsa.2015.4102) nicht überschritten werden, die sowohl über eine einmalige Anwendung als auch über mehrere Anwendungen im Laufe eines Tages (wobei 1 bis 3 Anwendungen pro Tag bevorzugt werden) aufgenommen werden kann.
Bevorzugt enthält die Oralgel-Zusammensetzung Coffein in einer Menge von 0,0001 bis 0,25 Gewichtsprozent, bevorzugt von 0,001 bis 0,1 Gewichtsprozent und besonders bevorzugt von 0,01 bis 0,05, insbesondere 0,025 Gewichtsprozent enthält, jeweils bezogen auf das Gesamtgewicht der Oralgel-Zusammensetzung.
Erfindungsgemäß ist die Zusammensetzung ein Oralgel, d.h. ein im Mund(bereich) anzuwendendes Gel. Das Gel wird auf das Zahnfleisch aufgetragen. Dieses Aufträgen kann mit Hilfe der Finger oder auch mit einer dazu geeigneten Vorrichtung wie z.B. einem Wattestäbchen, einem Pinsel oder einer Zahnbürste erfolgen. Alternativ kann das Gel auch in eine Beißschiene gefüllt werden, welche dann über einen längeren Zeitraum wie beispielsweise über Nacht getragen werden kann. Durch das vorstehend beschriebene Aufbringen des Gels auf das Zahnfleisch wird
ein langfristiger Kontakt zwischen Gel und Zahnfleisch erreicht und damit eine Intensivanwendung gewährleistet.
Im Rahmen der vorliegenden Erfindung wird unter einem Gel ein disperses System verstanden, welches aus mindestens zwei Komponenten besteht. Die feste Komponente wird als Gelbildner bezeichnet und kann dabei ein dreidimensionales Netzwerk ausbilden. In den Zwischenräumen der festen Komponente wird die flüssige Komponente eingelagert und damit immobilisiert. Die beiden Komponenten durchdringen sich also.
In einer bevorzugten Ausführungsform ist die flüssige Komponente Wasser.
Folglich umfasst die erfindungsgemäße Oralgel-Zusammensetzung als essentiellen Bestandteil (b) einen Gelbildner.
Gelbildner können synthetische oder natürliche Polymermaterialien sein. Die natürlichen Polymermaterialien sind bevorzugt Polymermaterialien auf pflanzlicher Basis.
In einer bevorzugten Ausführungsform umfasst der Gelbildner Methylcellulose, Ethylcellulose, Hydroxyethylcellulose, Hydroxypropylcellulose, Hydroxypropylmethylcellulose, Carboxymethylcellulose,
Natriumcarboxymethylcellulose, Agaraose, Agar, Scelrotium Gum, Xanthan Gum, Guar Gum, Carrageenan, Polyvinylpyrrolidon sowie Mischungen davon, besonders bevorzugt Hydroxyethylcellulose, Hydroxypropylcellulose,
Hydroxypropylmethylcellulose, Carboxymethylcellulose, Agaraose, Agar, Scelrotium Gum, Guar Gum, Carrageenan, Polyvinylpyrrolidon sowie Mischungen davon.
Ganz besonders besteht der Gelbildner aus einem natürlichen Ausgangsmaterial, bevorzugt ausgewählt aus der Gruppe der Cellulosen und Xanthane.
Als Cellulosen kommen hierbei insbesondere Hydroxyethylcellulose, Hydroxypropylcellulose, Hydroxypropylmethylcellulose, Carboxymethylcellulose, am
meisten bevorzugt Hydroxyethylcellulose in Frage.
In einer weiteren bevorzugten Ausführungsform enthält die erfindungsgemäße Oralgel-Zusammensetzung Gelbildner in einer Menge von 0,01 bis 4,0 Gewichtsprozent, besonders bevorzugt von 1 ,5 bis 3,0 Gewichtsprozent und insbesondere von etwa 2,0 Gewichtsprozent, jeweils bezogen auf das Gesamtgewicht der Oralgel-Zusammensetzung.
In einer bevorzugten Ausführungsform umfasst die erfindungsgemäße Oralgel- Zusammensetzung weiterhin (c) einen oder mehrere pH-Regulatoren und/oder (d) einen oder mehrere pharmazeutische oder kosmetische Hilfsstoffe. pH-Regulatoren (c) sind Substanzen, die dazu dienen, bei einer Zusammensetzung einen bestimmten pH-Wert oder einen bestimmten pH-Wert-Bereich einzustellen.
Beispiele für pH-Regulatoren, sind Essigsäure, Acetaten, Milchsäure, Lactaten, Äpfelsäure, Malate, Fumarsäure, Citronensäure, Citrate, Weinsäure, Tartrate, Orthophosphaten, Di-, Tri und Polyphosphate, Salzsäure, Chloriden, Schwefelsäure, Sulfate, Hydroxiden, Oxide, Adipinsäure, Adipate, Gluconsäure, Gluconate, Phosphorsäure, Calciumcarbonat oder ein Hydrat davon. Ein bevorzugtes Beispiel für einen pH-Regulator, der zugegeben werden kann, wenn ein niedrigerer pH-Wert gewünscht wird, ist Phosphorsäure (H3PO4).
Die erfindungsgemäße Oralgel-Zusammensetzung weist vorzugsweise keinen sauren pH-Wert von unter 5,5 auf. Bei einer zu sauren Zusammensetzung würde nämlich das Risiko einer Demineralisation der Zahnhartsubstanz (Erosion) bestehen.
Vorzugsweise weist die erfindungsgemäße Oralgel-Zusammensetzung einen pH- Wert im Bereich von 5,0 bis 9,0 auf, mehr bevorzugt einen pH-Wert im Bereich von 6,5 bis 8,0, insbesondere einen pH-Wert von etwa 7,0. Es hat sich herausgestellt, dass die Remineralisation der Zahnhartsubstanz durch einen pH-Wert in dem genannten Bereich begünstigt ist. Vorzugsweise umfasst der pH-Regulator (c) Calciumcarbonat oder ein Hydrat davon. Alternativ umfasst der pH-Regulator eine
Mischung aus (c) Calciumcarbonat (oder ein Hydrat davon) und Natriumhydroxid.
In einer weiteren bevorzugten Ausführungsform enthält die erfindungsgemäße Oralgel-Zusammensetzung einen pH-Regulator (c) in einer Menge von 0,05 bis 3,0 Gewichtsprozent, mehr bevorzugt von 0,1 bis 2,5 Gewichtsprozent, insbesondere bevorzugt 0,2 bis 1,5 Gewichtsprozent, jeweils bezogen auf das Gesamtgewicht der Oralgel-Zusammensetzung.
In einer bevorzugten Ausführungsform kann die erfindungsgemäße Oralzusammensetzung einen oder mehrere pharmazeutische oder kosmetische Hilfsstoffe enthalten. Diese pharmazeutischen oder kosmetischen Hilfsstoffe werden beispielsweise in Toothpastes, Monographs in Oral Science, Vol. 23, Ist edition, 2013 beschrieben.
Vorzugsweise umfassen die pharmazeutischen oder kosmetischen Hilfsstoffe antimikrobielle Substanzen, Aromastoffe und Zuckeralkohole.
Antimikrobielle Substanzen sind Substanzen, welche Mikroorganismen, wie Bakterien, abtöten oder deren Vermehrung stark reduzieren können. Es gibt neben antimikrobiellen Substanzen mit einer unspezifischen Abwehr von Bakterien und Pilzen ebenfalls solche, die beispielsweise nur gegen spezielle Bakterien wirken. Durch den Einsatz von antimikrobiellen Substanzen kann beispielsweise auch Mundgeruch bekämpft werden. Vorzugsweise können antimikrobielle Substanzen in einer Menge von 0,01 bis 1 ,0 Gew.%, bevorzugt 0,05 bis 0,5 Gew.% in der erfindungsgemäßen Oralgel-Zusammensetzung enthalten sein. Beispiele für die in der Oralpflege eingesetzten antimikrobiellen Substanzen sind Zinkverbindungen wie Zinkchlorid und Zinkcitrat sowie Chlorhexidin, Triclosan, Cetylpyridiniumchlorid und Zinnchlorid. Besonders bevorzugt enthält die erfindungsgemäße Oralgel- Zusammensetzung kein Chlorhexidin.
Aromastoffe können der erfindungsgemäßen Oralzusammensetzung den gewünschten Geschmack geben. Zudem können Aromastoffe speichelanregend sein, wobei die Feuchtigkeit des Speichels einen positiven Einfluss auf die
Remineralisierung des Zahnes haben kann. Ein Beispiel für einen speichelanregenden Aromastoff ist Pellitorin, insbesondere trans-Pellitorin. Weitere Beispiele für Aromastoffe sind die kommerziell erhältlichen Aromastoffe „Aktiv Fresh PO 128844“ und „Aroma Citromin P0125337“ sowie Menthol.
In einer bevorzugten Ausführungsform umfasst der pharmazeutische oder kosmetische Hilfsstoff (d) Zuckeralkohole, insbesondere Xylit oder Sorbitol. Xylit kann die Anzahl von Kariesbakterien minimieren und deren Wachstum hemmen. Weiterhin kann Xylit den Speichelfluss stimulieren. Durch die erhöhte Menge an Speichel fällt eine erhöhte Menge an Phosphat an. Dieses Phosphat kann zusammen mit dem Calcium(ionen) aus der erfindungsgemäßen Oralgel-Zusammensetzung zu Hydroxylapatit reagieren. Die erfindungsgemäße Oralgel-Zusammensetzung kann Xylit in einer Menge von 0,5 bis 4 Gew.%, vorzugsweise 0,7 bis 2,5 Gew.%, insbesondere etwa 1,0 Gew.% bezogen auf das Gesamtgewicht der Oralzusammensetzung enthalten. Neben oder statt Xylit kann die erfindungsgemäße Zusammensetzung auch Sorbitol enthalten.
In einer weiteren bevorzugten Ausführungsform umfasst das erfindungsgemäße Oralgel einen geeigneten Bitterblocker (e). Dieser Bitterblocker besteht zwischen 0,0001 bis 15 Gew.%, in einer Ausführungsform zwischen 2 und 12 Gew.%, in einer weiteren Ausführungsform zwischen 3 und 10 Gew.%, in einerweiteren Ausführungsform zu 1, 2, 3, 4, 5, 6, 7, 8 oder 9 Gew.% aus L-Lysin, in einerweiteren Ausführungsform in den gleichen Mengen aus L-Lysin Hydrochlorid, und einem Bitterblockerträgerstoff. Hierbei wird so viel Bitterblockerträgerstoff hinzugefügt, dass ein Gesamtprozentwert von 100 Gew.% für den Bitterstoffblocker selbst erreicht wird, also z.B. 85 Gew.% Bitterblockerträgerstoff bei 5 Gew.% Bitterblocker.
Alternativ ist es auch möglich, dass das erfindungsgemäße Oralgel frei ist von Bitterblockern, was insbesondere bei geringeren Coffeinkonzentrationen der Fall sein wird. Bitterblocker können das Geschmacksempfinden insgesamt negativ beeinflussen.
Herkömmliche Zahnpflegezusammensetzungen wie beispielsweise Oralgele
enthalten häufig Fluoridverbindungen wie beispielsweise Natriumfluorid, Aminfluoride oder Zinnfluorid. In einer besonders bevorzugten Ausführungsform der vorliegenden Erfindung enthält die erfindungsgemäße Oralgel-Zusammensetzung allerdings gerade keine Fluoridverbindung und ist somit im wesentlichen bzw. sogar vollständig fluoridfrei.
Weiterhin enthalten bekannte Zahnpflegezusammensetzungen wie beispielsweise Tooth Mouse von GC tierische Bestandteile wie Casein-Phosphopeptid- Komponenten. In einer besonders bevorzugten Ausführungsform enthält die erfindungsgemäße Oralgel-Zusammensetzung keine tierischen Bestandteile und ist somit vegan.
In einer bevorzugten Ausführungsform enthält die erfindungsgemäße Oralgel- Zusammensetzung
- mindestens 0,00001 und weniger als 0,5 Gew.%, vorzugsweise 0,0001 bis 0,01 Gew.%, mehr bevorzugt 0,0005 bis 0,001, besonders bevorzugt 0,005 Gew.% Coffein,
- 5 bis 25 Gew.%, vorzugsweise 10 bis 20 Gew.%, mehr bevorzugt 12 bis 18 Gew., insbesondere etwa 15 Gew.% Ca5(P04)3(0H),
- 0,5 bis 5 Gew.%, vorzugsweise 1 bis 4 Gew.%, mehr bevorzugt 1 ,5 bis 3 Gew.%, insbesondere etwas 2,0 Gew.% 0,15 bis 0,5 Gew.% ein oder mehrere Calciumsalze ausgewählt aus Calciumchlorid, Calciumbromid, Calciumnitrat, Calciumacetat, Calciumgluconat, Calciumlactat, Calciumtartrat sowie deren Flydrate und Mischungen davon
- 1 bis 4 Gew.%, vorzugsweise 1 ,5 bis 3 Gew.%, insbesondere etwa 2 Gew.%, Gelbildner - 0,05 bis 3,0 Gew.%, vorzugsweise 0,1 bis 2,5 Gew.%, insbesondere 0,2 bis 1,5 Gew.% pH-Regulator

Bevorzugt ist die Oralgel-Zusammensetzung frei von Fluoriden und/oder Zinnsalzen
und/oder Chlorhexidin und/oder Cetylpyridiniumchlorid und/oder Triclosan.
Zinnsalze wie Zinnfluorid und Zinnchlorid führen zu Verfärbungen der Zähne. Chlorhexidin führt ebenfalls zu Verfärbungen der Zähne und Geschmacksbeeinträchtigungen. Cetylpyridiniumchlorid kann auch zu Verfärbungen der Zähne führen. Bei Triclosan besteht die Gefahr einer Resistenzbildung.
Weiterhin betrifft die vorliegende Erfindung die Verwendung der erfindungsgemäßen Oralgel-Zusammensetzung zur Behandlung von und/ oder bei der Prävention bei Parodontitis/Gingivitis bzw. Zahnfleischrückgang.
In diesem Zusammenhang beträgt die Verweildauer der Oralgel-Zusammensetzung auf dem Zahnfleisch vorzugsweise zwischen 3 Minuten und 72 Stunden, vorzugsweise zwischen 5 Minuten und 48 Stunden, insbesondere zwischen 10 min und 24 Stunden.
Nachstehend wird die Erfindung an Beispielen erläutert.

Das Oralgel ist ein weißes, homogenes Gel mit einem zitronig, minzig, süßen Geschmack und weist einen gemittelten pH-Wert von 7,7 auf.
Studie 1 : Wirkung von Coffein auf die biologischen Funktionen humaner epidermaler HaCaT-Keratinozyten (Viabilitätsstudie)
Um die Wirkung von Coffein auf die biologischen Funktionen humaner epidermaler HaCaT-Keratinozyten zu untersuchen, wurden Zellversuche an humanen epidermalen HaCaT-Keratinozyten durchgeführt, wobei folgende Untersuchungen durchgeführt wurden:
1. P13-A biologische Versuchsreihe 1 mit zwei unabhängigen Untersuchungen
2. P13-B biologische Versuchsreihe 2 mit zwei unabhängigen Untersuchungen.
Die anfängliche Zellzahl betrug 5.000 Zellen/well in 100 pl Kulturmedium.
Das Kulturmedium war HaCaT Medium (DMEM mit Zusatz von 10 % FBS, 1 % Penicillin-Streptomycin, 0,5 % Fungizone).
Die Behandlungsgruppen waren folgende:
1. Kontrollgruppe (CTRL)
2. 0,00005 % Coffein
3. 0,0001 % Coffein
4. 0,0005 % Coffein
5. 0,001 % Coffein
6. 0,005 % Coffein
Die Versuchsdauer betrug 24, 48 und 72 Stunden.
Die Untersuchung erfolgte in Form einer kolorimetrischen MTT-Untersuchung.

Die Studie ergab, dass keine der getesteten Konzentrationen auch bei einer Versuchsdauer von 72 Stunden eine Cytotoxizität zeigte und dass bestimmte Konzentrationen von Coffein die Zahl von lebensfähigen Zellen erhöhte.
Studie 2: Wirkung von Coffein auf die biologischen Funktionen humaner epidermaler FlaCaT-Keratinozyten (Zellproliferationsstudie)
Um die Wirkung von Coffein auf die Zellproliferation humaner epidermaler FlaCaT- Keratinozyten zu untersuchen, wurden humane FlaCaT-Keratinozyten einer CyQUANT Viabilitätsuntersuchung unterzogen, wobei folgende Versuchsreihen durchgeführt wurden:
1. P22 Experiment 1 (biologische Versuchsreihe 1)
2. P24 Experiment 2 (biologische Versuchsreihe 2).
Die anfängliche Zellzahl betrug 5.000 Zellen/well in 1.500 pl Kulturmedium.
Das Kulturmedium war FlaCaT Medium (DMEM mit Zusatz von 10 % FBS, 1 % Penicillin-Streptomycin, 0,5 % Fungizone).
Die Behandlungsgruppen waren folgende:
1. Kontrollgruppe (CTRL)
2. 0,00005 % Coffein
3. 0,0005 % Coffein
4. 0,005 % Coffein
Die Versuchsdauer betrug 24, 48 bzw. 72 Stunden.
Die Untersuchung erfolgte mittels fluorimetrischer CyQUANT Untersuchung, wobei der DNA-Inhalt gemessen wurde, der die Zellproliferation widerspiegelt.

Die Studie zeigt, dass Coffein die Zellproliferation zu fördern scheinen; die besten proliferationssteigernden Effekte wurden nach 72 Stunden festgestellt.
Studie 3: Wirkung von Coffein auf die biologischen Funktionen humaner epidermaler FlaCaT-Keratinocyten (Zell-Proliferations-Studie)
Um die Wirkung von Coffein auf die biologischen Funktionen humaner epidermaler FlaCaT-Keratinocyten zu untersuchen, wurden humane FlaCaT-Keratinocyten Q- PCR und ELISA unterzogen, wobei folgende Versuchsreihen durchgeführt wurden:
1. Für Q-PCR: Bestimmung von VEGF, HGF und EGF mRNA-Expression in Zelllysaten
2. Für ELISA: Bestimmung von VEGF, HGF und EGF Proteinsynthese/-sekretion in das Kulturmedium
Die anfängliche Zellzahl betrug
1. für Q-PCR: 140.000 Zellen/well in 1.500 pl Kulturmedium
2. für ELISA: 140.000 Zellen/well in 1.500mI Kulturmedium.
Das Kulturmedium war HaCaT Medium (DMEM mit Zusatz von 10 % FBS, 1 % Penicillin-Streptomycin, 0,5 % Fungizone).
Die Behandlungsgruppen waren folgende:
1. Kontrollgruppe (CTRL)
2. 0,00005 % Coffein
3. 0,0005 % Coffein
4. 0,005 % Coffein
Die Versuchsdauer betrug für
1. Q-PCR 24 Stunden (einfache Behandlung zum Zeitpunkt 0 h)
2. ELISA 48 Stunden (doppelte Behandlung zum Zeitpunkt 0 h und 24 h ohne Medienwechsel).
Die Untersuchung erfolgte
1. für Q-PCR durch Bestimmung von VEGF mRNA-Expression in Zelllysaten
2. für ELISA durch Bestimmung von VEGF-Proteinsynthese/sekretion in dem Kulturmedium.

Die Studie ergab,
- Q-PCR (24 Stunden Behandlungszeit) - Experimente 1 und 2: Alle Coffein- Konzentrationen scheinen die mRNA-Expression von VEGF im Vergleich zu der Kontrollgruppe zu erhöhen.
- ELISA (48 Stunden Behandlungszeit) - Experimente 1 und 2: 0,0005 %
Coffein erhöhen die Produktion/Sekretion von VEGF-Protein im Vergleich zur Kontrollgruppe in beiden Experimenten. Bei Experiment 2 zeigte auch 0,005 %
Coffein Wirkung.
Zusammengenommen legen diese Daten nahe, dass bestimmte Coffein- Konzentrationen die Synthese und Produktion/Sekretion von VEGF in kultivierten humanen epidermalen HaCaT-Keratinocyten erhöhen.
Die Expression von HGF-spezifischer mRNA, FIGF-spezifischem Protein, TNFa- spezifischer mRNA, TNFa-spezifischem Protein und TGFßl -spezifischem Protein lag jeweils in beiden Experimenten unter der Detektionsgrenze.
Die Experimente bezüglich GM-CSF zeigen
- Q-PCR-Experiment (24 h Behandlungszeit) - Experimente 1 und 2: Alle getesteten Coffein-Konzentrationen steigerten offenbar die mRNA-Expression von VEGF im Vergleich zur Kontrollgruppe.
- ELISA (48 h Behandlungszeit) - Experimente 1 und 2: 0,0005 % Coffein steigerten die Produktion/Sekretion von VEGF-Protein im Vergleich zur Kontrollgruppe in beiden Experimenten. In Experiment 2 zeigten Oj , 005 % Coffein ebenfalls Wirkung.
Zusammengenommen legen diese Daten deutlich nahe, dass bestimmte Konzentrationen von Coffein die Synthese/Produktion/Sekretion von VEGF in kultivierten humanen epidermalen HaCaT-Keratinozyten steigern.

Zusammengenommen legen diese Daten - trotz der widersprüchlichen Ergebnisse in
den beiden Q-PCR-Experimenten - nahe, dass Coffein höchstwahrscheinlich nicht die Synthese/Produktion/Sekretion von TGFß2 in kultivierten humanen epidermalen HaCaT-Keratinozyten verändert. Die Experimente bezüglich TGFßl zeigten

Claims
1. Oralgel-Zusammensetzung zum Auftrag auf das Zahnfleisch und zum Verbleib auf diesem für mindestens 3 Minuten, umfassend
(a) mindestens 0,00001 und weniger als 0,5 Gew.-% Coffein und
(b) einen Gelbildner.
2. Oralgel-Zusammensetzung nach Anspruch 1, dadurch gekennzeichnet, in einer Menge von 0,0001 bis 0,25 Gewichtsprozent, bevorzugt von 0,001 bis 0,1 Gewichtsprozent und besonders bevorzugt von 0,01 bis 0,05, insbesondere 0,025 Gewichtsprozent enthält, jeweils bezogen auf das Gesamtgewicht der Oralgel-Zusammensetzung.
3. Oralgel-Zusammensetzung nach Anspruch 1 oder Anspruch 2, dadurch gekennzeichnet, dass der Gelbildner ausgewählt ist aus der Gruppe enthaltend Cellulose und Cellulosederivate wie Methylcellulose, Ethylcellulose, Hydroxyethylcellulose, Hydroxypropylcellulose, Hydroxypropylmethylcellulose, Carboxymethylcellulose, Natriumcarboxymethylcelullose, Agaraose, Agar, Pektine, Sclerotium Gum, Xanthan Gum, Guar Gum, Carrageenan,
Alginsäure, Polyvinylalkohol, Polyvinylpyrrolidon sowie Mischungen hiervon.
4. Oralgel-Zusammensetzung nach einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass der Gelbildner aus einem natürlichen Ausgangsmaterial besteht, bevorzugt ausgewählt aus der Gruppe der Cellulosen und Xanthane.

Oralgel-Zusammensetzung nach einem der Ansprüche 1 bis 4, dadurch gekennzeichnet, dass die Oralgel-Zusammensetzung Gelbildner in einer Menge von 0,01 bis 4,0 Gewichtsprozent, besonders bevorzugt von 1 ,5 bis 3,0 Gewichtsprozent und insbesondere von etwa 2,0 Gewichtsprozent enthält, jeweils bezogen auf das Gesamtgewicht der Oralgel-Zusammensetzung.
6. Oralgel-Zusammensetzung nach einem der Ansprüche 1 bis 5, dadurch
gekennzeichnet, dass die Oralgel-Zusammensetzung weiterhin (c) einen oder mehrere pH-Regulatoren und/oder (d) einen oder mehrere pharmazeutische oder kosmetische Hilfsstoffe umfasst.
7. Oralgel-Zusammensetzung nach Anspruch 6, dadurch gekennzeichnet, dass der pH-Regulator (d) ausgewählt ist aus der Gruppe bestehend aus Essigsäure, Acetaten, Milchsäure, Lactaten, Äpfelsäure, Malaten,
Fumarsäure, Citronensäure, Citraten, Weinsäure, Tartraten, Orthophosphaten, Di-, Tri- und Polyphosphaten, Salzsäure, Chloriden, Schwefelsäure, Sulfaten, Hydroxiden, Oxiden, Adipinsäure, Adipaten, Gluconsäure, Gluconaten, Calciumcarbonat oder ein Hydrat davon umfasst.
8. Oralgel-Zusammensetzung nach Anspruch 6, dadurch gekennzeichnet, dass der pharmazeutische oder kosmetische Hilfsstoff (d) einen oder mehrere aus antimikrobiellen Substanzen, Aromastoffen und Zuckeralkoholen umfasst.
9. Oralgel-Zusammensetzung gemäß einem der Ansprüche 1 bis 8, dadurch gekennzeichnet, dass die Oralgel-Zusammensetzung einen pH-Wert im Bereich von 5,5 bis 8,5, mehr bevorzugt einen pH-Wert im Bereich von 7,0 bis 8,0, insbesondere einen pH-Wert von etwa 7,5 aufweist.
10. Oralgel-Zusammensetzung gemäß einem der Ansprüche 1 bis 9, dadurch gekennzeichnet, dass die Oralgel-Zusammensetzung frei ist von Fluoriden und/oder Zinnsalzen und/oder Chlorhexidin und/oder Cetylpyridiniumchlorid und/oder Triclosan.
11.Verwendung der Oralgel-Zusammensetzung gemäß einem der Ansprüche 1 bis 10 zur Behandlung und/oder bei der Prävention von Parodontitis/Gingivitis bzw. Zahnfleischrückgang.
12. Verwendung der Oralgel-Zusammensetzung gemäß Anspruch 11 , dadurch gekennzeichnet, daß die Verweildauer der Oralgel-Zusammensetzung zwischen 3 Minuten und 72 Stunden, vorzugsweise zwischen 5 Minuten und
48 Stunden, insbesondere zwischen 10 min und 24 Stunden beträgt.
