Die vorliegende Erfindung betrifft die Verwendung von 3ß-Hydroxy-chlormadinonacetat (17a-acetoxy-chloropregna-4,6-dien-3ß-ol-20-on) und/oder 3a-Hydroxy-chlormadinonacetat (17a-acetoxy-chloropregna-4,6-dien-3a-ol-20-on) zur Herstellung eines Arzneimittels zur Behandlung von Haut, vorzugsweise menschlicher Haut mittels topischer Anwendung, sowie eine pharmazeutische oder kosmetische Zusammensetzung enthaltend 3ß-Hydroxy-chlormadinonacetat und/oder 3a-Hydroxy-chlormadinonacetat.

3-HYDROXy-CHLORMADINONACBTAT ZUR TOPISCHEN BEHANDLUNG VON ANDROGEN-ABHANGIGEN
HAUTERKRANKUNGΞN
Die vorliegende Erfindung betrifft die Verwendung von 3ß-Hydroxy-chlormadinona- cetat (17α-acetoxy-chloropregna-4,6-dien-3ß-ol-20-on) und/oder 3α-Hydroxy-chlor- madinonacetat (17α-acetoxy-chloropregna-4,6-dien-3α-ol-20-on) zur Herstellung eines Arzneimittels zur Behandlung von Haut, vorzugsweise menschlicher Haut mittels topischer Anwendung, sowie eine pharmazeutische oder kosmetische Zusammensetzung enthaltend 3ß-Hydroxy-chlormadinonacetat und/oder 3α- Hydroxy-chlormadinonacetat.
In Mitteleuropa wird die Zahl an hormonell bedingten Hauterkrankungen wie Akne, Alopezie, Seborrhöe und/oder Hirsutismus auf 35 bzw. 50% geschätzt. Akne, Alopezie, Seborrhöe und Hirsutismus sind meistens Androgen-abhängige Erkrankungen.
Akne, insbesondere die „gewöhnliche" Akne, tritt hauptsächlich durch einen verstärkten Androgeneinfluss während der Pubertät auf. Alopezie wird unter anderem durch eine genetisch bedingte Überempfindlichkeit gegenüber dem Hormon Dihydrotestosteron (DHT), einem androgenen Hormon, ausgelöst. Seborrhöe ist eine krankhaft veränderte Absonderung der Talgdrüsen und bei Hirsutismus findet ein übermäßiger Haarwuchs nach männlichem Verteilungsmuster bei Frauen statt.
Durch hormonale Behandlung, insbesondere durch Gestagene mit antiandrogener Wirkung, kann der Krankheitsverlauf dieser Androgen-abhängigen Krankheiten günstig beeinflusst werden. Derartige antiandrogene Gestagene werden üblicherweise oral verabreicht.
In WO 86/01402, WO 91/06284 und EP 0 765 663 werden die Gestagene Chlorma- dinonacetat (CMA), Cyproteronacetat (CPA) bzw. Megestrolacetat (MGA) zur Her-
Stellung eines Arzneimittels zur topischen Anwendung Androgen-abhängiger Erkrankungen beschrieben.
Nachteilig bei diesen topisch angewendeten, antiandrogen wirksamen Arzneimitteln ist aber, dass ihre topische Anwendung auch gleichzeitig zu einer hohen systemischen Belastung mit dem jeweiligen Wirkstoff führt.
Die Aufgabe der vorliegenden Erfindung war es daher, eine pharmazeutische oder kosmetische Zusammensetzung zur topischen Behandlung und/oder Prophylaxe von Androgen-abhängigen Erkrankungen zur Verfügung zu stellen, bei deren Anwendung die systemische Belastung durch den entsprechenden Wirkstoff praktisch vernachlässigbar ist. Diese Zusammensetzungen sollen daher sowohl zur topischen Behandlung der Haut und der Haare von weiblichen als auch männlichen Patienten geeignet sein.
Diese Aufgabe wird durch die Verwendung von 3ß-Hydroxy-chlormadinonacetat (17α-acetoxy-chloropregna-4,6-dien-3ß-ol-20-on) und/oder 3α-Hydroxy-chlormadino- nacetat (17α-acetoxy-chloropregna-4,6-dien-3α-ol-20-on) zur Herstellung eines Arzneimittels zur topischen Behandlung von Haut, vorzugsweise menschlicher Haut, und gegebenenfalls auch Haaren gelöst.
Die antiandrogenen Wirkstoffe 3ß-Hydroxy-chlormadinonacetat bzw. 3α-Hydroxy- chlormadinonacetat zeichnen sich überraschenderweise durch eine weit geringere Hautdurchdringung verglichen mit CMA aus, wodurch bei einer effektiven Behandlung von Androgen-abhängigen Erkrankungen durch topische Anwendung eine systemische Wirkstoffbelastung praktisch verhindert wird.
Ein weiterer Gegenstand der vorliegenden Erfindung ist auch eine pharmazeutische Zusammensetzung zur Behandlung der Haut mittels topischer Anwendung umfassend 3ß-Hydroxy-chlormadinonacetat und/oder 3α-Hydroxy-chlormadinonacetat in einer Menge von 0,01 bis 20 Gew.-%, vorzugsweise 0,01 bis 10 Gew.-%, bezogen auf das Gesamtgewicht der erfindungsgemäßen, pharmazeutischen Zusammensetzung, und wenigstens ein pharmazeutisch verträgliches Vehikel, vorzugsweise in üblichen Mengen.
Der Begriff "pharmazeutisch verträgliche Vehikel" im Sinne der Erfindung bedeutet übliche pharmazeutische Hilfs- und Zusatzstoffe zur Formulierung von pharmazeutischen Zusammensetzungen, wie pharmazeutisch verträgliche Puffer, Konservierungsmittel, Reduktionsmittel, Antioxidantien, UV-Absorber, Stabilisatoren, Penetrationspromotoren, Emulgatoren, Geliermittel, Verdickungsmittel, Tenside, Duftstoffe, Aromazusätze und Farbstoffe. Derartige Hilfs- und Zusatzstoffe sind im Allgemeinen vorzugsweise pharmazeutisch unbedenklich.
Geeignete Puffer, die für die erfindungsgemäße, pharmazeutische Zusammensetzung verwendet werden können, sind dem Fachmann bekannt. So kann als Puffer Bernsteinsäure, Zitronensäure, Milchsäure, Phosphorsäure, Trinatriumphosphat, Di- natriumhydrogenphosphat, Natriumdihydrogenphosphat, Natriumcarbonat, sowie Kombinationen aus Milchsäure und Natriumhydroxid eingesetzt werden. Mit dem Puffer wird vorzugsweise der pH-Wert der Zusammensetzung auf 5,5-7,5 eingestellt.
Als Stabilisatoren bzw. Konservierungsmittel können Antioxidantien, Reduktionsmittel und/oder UV-Absorber eingesetzt werden. Insbesondere bevorzugt wird für die erfindungsgemäße, pharmazeutische Zusammensetzung wenigstens ein Reduktionsmittel ausgewählt aus der Gruppe umfassend Sulfite, wie Natriumsulfit, Kaliumsulfit, Ammoniumsulfit, Natriumhydrogensulfit, Kaliumhydrogensulfit, Natriumbisulfit, Calciumsulfit, Calciumhydrogensulfit, Kaliumbisulfit, Natriumdisulfit, Ammoniumhydrogensulfit, Natriummetabisulfit, Kaliummetabisulfit; Mercaptocarbonsäuren, wie 2-Mercaptopropionsäure, 3-Mercaptopropionsäure, Mercapto-Bernsteinsäure, Thioglycolsäure, Ammoniumthioglycolat, Natrium- thioglycolat, L-Cystein, Dimercapto-Adipinsäure; Mercapto-amine, wie L- Cysteinethylester, L-Cysteinmethylester, N-acetyl-L-cystein, Cysteamin; Mercapto- amide, wie Thioglycolamid, N-hydroxyethyl-mercapto-acetamid, N-methyl-mercapto- acetamid, 2-Mercapto-propionamid; Hydroxide, wie Guanidinhydroxid, Natriumhydroxid; Alkohole und Diole, wie Resorcinol, Thioglycerin, Glycerol-monothioglycolat, Glycol-thioglycolat; Di-thio-Verbindungen, wie Dihydroliponsäure, Natrium-dihydroli- ponsäure, Dithiothreitol, 1 ,3-dithiopropanol; Lithiumchlorid, Tris(hydroxymethyl)phosphin, Thioglycol-hydrazid, 2-Mercapto-ethansulfonsäure, Homocystein-thiolacton, Polythiolpolymere, Salze von Hydrogensulphids, Amine in
alkalischer Lösung, Salze von Hydrogencyanid, Borohydrid, Dithionit, Estersalze von Sulphoxylaten, Ameisensäure, Oxalsäure, Diazolidinyl-Harnstoff, lodopropynylbutyl- carbamat, Chloromethylisotiazolinon, Methylisothiazolinon, Butylparaben, Ethylpara- ben, Methylparaben, Propylparaben, Isobutylparaben und Phenoxyethanol eingesetzt. Die Stabilisatoren werden vorzugsweisein einer Menge von 0,001 bis 2 Gew.-%, bezogen auf das Gesamtgewicht der erfindungsgemäßen, pharmazeutischen Zusammensetzung eingesetzt.
Vorzugsweise wird als Antioxidans-Komponente für die erfindungsgemäße, pharmazeutische Zusammensetzung wenigstens ein Antioxidans ausgewählt aus der Gruppe umfassend Ascorbinsäure (Vitamin C), Natrium-L-Ascorbat, Calcium-L- Ascorbat, Ascorbylpalmitat, Butylhydroxyanisol, Butylhydroxytoluol, Calcium-Di- natrium-EDTA, Propylgallat, Octylgallat, Dodecylgallat (Laurylgallat), Isoascorbin- säure, Natriumisoascorbat, Lecithin, Milchsäure, Polyphosphat, Schwefeldioxid, Selen, Tocopherol (Vitamin E), α-Tocopherol, γ-Tocopherol, δ-Tocopherol, Zinn-Il- Chlorid, Zitronensäure, Natriumeitrat und Kaliumeitrat verwendet. Das Antioxidans wird vorzugsweise in einer Menge von 0,001 bis 2 Gew.-%, bezogen auf das Gesamtgewicht der erfindungsgemäßen, pharmazeutischen Zusammensetzung eingesetzt.
Für die erfindungsgemäße, pharmazeutische Zusammensetzung können bevorzugt als UV-Absorber folgende Verbindungen eingesetzt werden:
(A) p-Amino-benzoesäure (PABA)-Typ UV-Absorber, wie p-Amino-benzoesäure,
PABA-Monoglycerinester, N.N-Dipropoxy-PABA-ethylester, N,N-Diethoxy PABA-ethylester, N,N-Dimethyl PABA-ethylester, N,N-Dimethyl PABA-butyl- ester, N,N-Dimethyl PABA-amylester und N,N-Dimethyl PABA-octlylester.
(B) Anthranilsäure-Typ UV-Absorber, wie Homomethyl-N-Acetyl-anthranilsäure.
(C) Salicylsäure-Typ UV-Absorber: Amyl-salicylsäure, Menthyl-salicylsäure, Ho- momenthyl-salicylsäure, Octyl-salicylsäure, Phenyl-salicylsäure, Benzyl- sali- cylsäure und/oder p-lsopropanol-phenyl-salicylsäure.
(D) Zimtsäure-Typ UV-Absorber, wie Octyl-Zimtsäure, Ethyl-4-isopropyl-zimtsäure,
Methyl-2,5-diisopropyl-zimtsäure, Ethyl-2,4-diisopropyl-zimtsäure, Methyl-2,4- diisopropyl-zimtsäure, Propyl-p-methoxy-zimtsäure, Isopropyl-p-methoxy-zimt-
säure, Isoamyl-p-methoxy-zimtsäure, Octyl-p-methoxy-zimtsäure (oder 2- ethylhexyl-p-methoxy-zimtsäure), 2-Ethoxyethyl-p-methoxy-zimtsäure, Cyclo- hexyl-p-methoxy-zimtsäure, Ethyl-α-cyano-ß-phenyl-zimtsäure, 2-Ethylhexyl-α- cyano-ß-phenyl-zimtsäure und/oder Glycerin-mono-2-ethylhexanoyl dipara- methoxy-zimtsäure.
(E) Benzophenon-Typ UV-Absorber, wie 2,4-Dihydroxy-benzophenon, 2,2'-Di- hydroxy-4-methoxy-benzophenon, 2,2'-Dihydroxy-4,4'-dimethoxy-benzophe- non, 2,2',4,4'-Tetrahydroxy-benzophenon, 2-Hydroxy-4-methoxy-benzophe- non, 2-Hydroxy-4-methoxy-4'-methyl-benzophenon, 2-Hydroxy-4-methoxy- benzophenon -5-Sulfonsäuresalze, 4-Phenyl-benzophenon, 2-Ethylhexyl-4'- phenyl-benzophenon-2-carboxylat, 2-Hydroxy-4-n-octoxy-benzophenon und/oder 4-Hydroxy-3-carboxy-benzophenon.
(F) Andere UV-Absorber, wie 3-(4'-Methylbenzyliden)-D,L-kampfer, 3-Benzyliden-
D.L-kampfer, Urocansäure, Ethyl-urocansäure, 2-Phenyl-5-methylbenzoxazol, 2,2'-Hydroxy-5-methylphenyl-benzotriazol, 2-(2'-Hydroxy-5'-t-octylphe- nyl)benzotriazol, 2-(21-Hydroxy-5'-methylphenyl)benzotriazole, Dibenzalazin, Dianisoylmethan, 4-Methoxy-41-t-butyl-dibenzoylmethan, 5-(3,3-Dimethyl-2- norbornyliden)-3-pentan-2-on, Kalziumlake, Bariumoxid, Aluminiumoxid, Eisenoxid und/oder Titandioxid.
Der UV-Absorber wird vorzugsweise in einer Menge von 1 bis 10 Gew.-%, bezogen auf das Gesamtgewicht der erfindungsgemäßen, pharmazeutischen Zusammensetzung, eingesetzt.
Die erfindungsgemäße, pharmazeutische Zusammensetzung kann gegebenenfalls noch weitere, pharmazeutisch verträgliche Zusatzstoffe, wie beispielsweise Penetrationspromotoren, Lösungsmittel wie Wasser und/oder Alkohole, oberflächenaktive Stoffe, Geliermittel, Weichmacher, Emulgatoren, Stabilisatoren, hydrophobe oder hydrophile Polymeren, Duftstoffe, Filmbildner und/oder Verdickungsmittel enthalten, soweit solche Zusatzstoffe nicht die Hautdurchdringung beeinflussen.
Geeignete Penetrationspromotoren im Sinne der vorliegenden Erfindung umfassen vorzugsweise Penetrationspromotoren ausgewählt aus der Gruppe umfassend Säureamide und Amine. Besonders bevorzugt wird als Penetrationspromotor
Harnstoff eingesetzt. Der Penetrationspromotor wird vorzugsweise in einer Menge von 0,5 bis 10 Gew.-%, bezogen auf das Gesamtgewicht der erfindungsgemäßen, pharmazeutischen Zusammensetzung eingesetzt.
Emolliens können bei der Zubereitung der erfindungsgemäßen, pharmazeutischen Zusammensetzung in solchen Mengen eingesetzt werden, dass eine Austrocknung der Haut verhindert bzw. vermindert wird. Als Emolliens können vorzugsweise wenigstens eine Verbindung ausgewählt aus der Gruppe umfassend Öle, Wachse, Silikonöle, Triglyceridester, Azetoglyceridester, ethoxylierte Glyceride, Alkylester, Alkenylester, Fettsäuren, Fettalkohole, Fettalkoholester, Lanolin, Lanolin-Derivate, mehrfach hydrierte Alkohole, deren Ether, mehrfach hydrierte Alkoholester, Wachsester, Bienenwachs-Derivate, pflanzliche Wachse, Phospholipide, Sterole und Amide. Die Emolliens werden vorzugsweise in einer Menge von 1 bis 25 Gew.-%, bezogen auf das Gesamtgewicht der erfindungsgemäßen Zusammensetzung eingesetzt.
Bevorzugte Emulsions-Stabilisatoren in der erfindungsgemäßen Zusammensetzung umfassen nichtionische, anionische, kationische und amphiphile Tenside, die vorzugsweise ausgewählt sind aus der Gruppe umfassend Polyethylenglycol-, Polyethylenglycerolderivate, Tweens, Tritons, Spans, Polyglycerine, Polyalkylglyceride, Alkylsulfonate, Arylsulfonate, Alkylphosphate, Arylphosphate, Alkyl-Betain-Derivate und Phosphatidylglycerol. Die Emulsionsstabilisatoren werden vorzugsweise in einer Menge von 0,1 bis 5 Gew.-%, bezogen auf das Gesamtgewicht der erfindungsgemäßen Zusammensetzung, eingesetzt.
Emulgatoren werden vorzugsweise der Zusammensetzung in solchen Mengen beigegeben, dass sie ein gleichmäßiges Vermischen der Komponenten der erfindungsgemäßen, pharmazeutischen Zusammensetzung ermöglichen. Übliche Emulgatoren sind vorzugsweise anionische, kationische und/oder nichtionische Emulgatoren. Beispiele für derartige Emulgatoren umfassen vorzugsweise Kaliumstearat, Natriumstearat, Ammoniumstearat, Triethanolaminstearat, Glycerin monostearat, Natriumlauryl-5-sulfat, Natriumacetylsulfat, N-(stearoyl-colamino formylmethyl)-pyridin-chlorid, N-soja-N-ethyl morpholin ethosulfat, Alkyl-dimethyl- benzyl ammoniumchlorid, Diisobutylphenoxythioxyethyl-dimethyl-benzy-
ammoniumchlorid, Acetyl-pyridiumchlorid, Monostearat, Polyethylenoxidstearat, Polyethylenoxidsorbitan-monostearat, Sorbitan, Propylenglycol-monostearat und/oder ethoxyliertes Lanolin. Die Emulgatorkomponente wird vorzugsweise in einer Menge von 0,1 bis 10 Gew.-%, bezogen auf das Gesamtgewicht der erfindungsgemäßen, pharmazeutischen Zusammensetzung, eingesetzt.
Weiterhin können auch oberflächenaktive Stoffe (Tenside), die nichtionische, kationische, anionische und/oder ampholytische Eigenschaften besitzen, miteingesetzt werden.
Bevorzugt werden nichtionische oberflächenaktive Stoffe für die Herstellung der erfindungsgemäßen, pharmazeutischen Zusammensetzung eingesetzt, wie vorzugsweise Sorbitanester, wie Sorbitan-monolaurat, Sorbitan-monooleat, Sorbitan- monoisostearat; Polyethylenoxid-sorbitanester, wie Polyethylenoxid -sorbitan- monoisostearat, Polyethylenoxid -sorbitan-monolaurat, Polyethylenoxid -sorbitan- monooleat; Glycerinester, wie Glycerin-monoisostearat, Glycerin-monomyristat; Polyethylenoxidglycerinether, wie Polyethylenoxidglycerin monoisostearat, Polyethylenoxidglycerin-monomyristat; Polyglycerinfettsäureester, wie Diglycerin- monostearat, Decaglycerin-decaisostearat, Diglycerin-diisostearat; Glycerinfettsäureester, wie Glycerin-monocaprat, Glycerin-monolaurat, Glycerin-monomyristat, Glycerin-monopalmitat, Glycerin-monooleat, Glycerin-monostearat, Glycerin-monolinoleat, Glycerin-monoisostearat, Glycerin-monodilinoleat, Glycerin- monodicaprat; Polyethylenoxidglycerinfettsäureester, wie Polyethylenoxidglycerin- monomyristat, Polyethylenoxidglycerin-monooleat, Polyethylenoxidglycerin-mo- nostearat; Polyethylenoxid-verzweigte Alkylether, wie Polyethylenoxid-octyl- dodecylalkohol, Polyethylenoxid-2-decyl tetradecylalkohol; Polyethylenoxidalkylether, wie Polyethylenoxid-oleyl-alkoholether, Polyethylenoxid-acetylalkoholether; Polyethylenoxid-hydrogenierte Castoröl-fettsäureester, wie Polyethylenoxid-hyd- rogeniertes Castoröl, Polyethylenoxid-dihydrocholesterolether, Polyethylenoxid- hydrogeniertes Castoröl isostearat und/oder Polyethylenoxidalkylarylether, wie Polyethylenoxid-octylphenolether umfassen.
Eingesetzte anionische oberflächenaktive Stoffe für die erfindungsgemäße, pharmazeutische Zusammensetzung umfassen vorzugsweise Salze, wie Diethanolaminsalz,
Triethanolaminsalz, Aminosäurensalz, Natriumsalz, Kaliumsalz; höherer Fettsäuren, wie Ölsäure, Stearinsäure, Isostearinsäure, Palmitinsäure, Myristinsäure, Ethercar- bonsäure-alkaline Salze, N-acylaminosäurensalze und höhere Alkylsulfonsäuresalze. Der oberflächenaktive Stoff wird vorzugsweise in einer Menge von 0,1 bis 10 Gew.- %, bezogen auf das Gesamtgewicht der erfindungsgemäßen, pharmazeutischen Zusammensetzung, eingesetzt.
Für die erfindungsgemäßen Zusammensetzungen geeignete Gelbildner umfassen vorzugsweise natürliche oder synthetische Polymere. Natürliche Polymere sind vorzugsweise ausgewählt aus der Gruppe umfassend Agar-Agar, Alginsäure, Alginat, amidiertes Pektin, Propylenglycolalginat, Carbomer, Carrageenan, Casein, Dammar- Gummi, Dextrine, Furcellaran, Gelatine, Guargummi, Guarkernmehl, Gellan, Ghatti- Gummi, Gummi arabicum, Gummi aus Fichtensaft, Johannisbrotkernmehl, Karaya- gummi, Keratin, Konjakmehl, L-HPC, Locust Bean Gum, Mastix, Pektin, Schellack, (ggf. modifizierte) Stärke, Tarakernmehl, Traganth, Xanthangummi und deren Derivate. Bevorzugte synthetische Polymere, die als Geliermittel für die erfindungsgemäße Zusammensetzung eingesetzt werden können sind ausgewählt aus der Gruppe umfassend Acrylsäurepolymere, Carbomer, Polyacrylamide und Alkylenoxid- polymere. Die Gelbildner werden vorzugsweise in einer Menge von 0,1 bis 5 Gew.- %, bezogen auf das Gesamtgewicht der erfindungsgemäßen Zusammensetzung eingesetzt.
Verdickungsmittel, die vorzugsweise in den erfindungsgemäßen Zusammensetzungen enthalten sein können, umfassen beispielsweise Candelilla, Carnauba- und mikrokristalline Wachse, Carbomer und Polyethylenoxid-Verdicker. Das Verdickungsmittel wird vorzugsweise in einer Menge von 0,5 bis 2 Gew.-%, bezogen auf das Gesamtgewicht der erfindungsgemäßen, pharmazeutischen Zusammensetzung eingesetzt.
Die erfindungsgemäße, pharmazeutische Zusammensetzung kann vorzugsweise wenigstens einen Farbstoff und/oder einen Duftstoff enthalten.
In einer bevorzugten Ausführungsform der erfindungsgemäßen, pharmazeutischen Zusammensetzung liegt als Duftstoff bzw. Aromazusatz wenigstens eine Verbindung
ausgewählt aus der Gruppe umfassend ungesättigte oder gesättigte, acyclische, monocyclische oder bicyclische Monoterpene und Sesquiterpene vor, die jeweils mit 1 , 2, 3 oder 4 gleichen oder verschiedenen Substituenten ausgewählt aus der Gruppe umfassend eine -OH, Oxo (=0 bzw. -O-), -O-C1-5-Alkyl, -O-Phenyl, -C(=O)- OH, -C(=O)-O-C1-5-Alkyl, -C(=O)-O-Phenyl, -O-C(=O)-C1-5-Alkyl, -O-C(=O)-Phenyl- Gruppe und -CN subsituiert sein können, wobei der Alkyl- und/oder Phenyl-Rest mit 1 , 2 oder 3 Substituenten ausgewählt aus der Gruppe umfassend OH, Oxo (=0) und -O-C-ι-5-Alkyl substituiert sein kann;
Phenyl- oder Naphthyl-Verbindungen, die jeweils mit 1 , 2, 3, 4 oder 5 gleichen oder verschiedenen Substituenten ausgewählt aus der Gruppe umfassend eine -C(=O)-H, -C(=O)-C1-5-Alkyl, -C(=O)-Phenyl, -OH, -C1-5-Alkyl, -C2-5-Alkenyl, -O-C1-5-Alkyl, -O- C(=O)-Ci-5-Alkyl, -O-C(=O)-C2-6-Alkylen, -O-C(=O)-Phenyl, -O-C(=O)-OH, Phenyl, -C(=O)-OH, -C(=O)-O-C1-5-Alkyl, -C(=O)-O-C2-8-Alkenyl, -C(=O)-O-Phenyl, -C(=O)-O- C3-8-Cycloalkyl, -CH=C(C1.5-Alkyl)(C(=O)-H)l -NO2 und -NH2-Gruppe substituiert sein können, wobei die Substituenten jeweils über eine lineare oder verzweigte Ci-7- Alkylen- oder C2-5-Alkenylen-Brücke gebunden sein können, die jeweils mit 1 , 2 oder 3 gleichen oder verschiedenen Substituenten ausgewählt aus der Gruppe umfassend F, Cl und Br substituiert sein kann;
Phenyl- oder Naphthyl-Verbindungen, die mit 1 oder 2 ungesättigten oder gesättigten 5- bis 7-gliedrigen aliphatischen Ringen kondensiert sein können, wobei die Ringe jeweils 1 , 2 oder 3 Heteroatom(e) ausgewählt aus der Gruppe bestehend aus Sauerstoff, Stickstoff und Schwefel als Ringglied(er) aufweisen können und mit 1 , 2, 3, 4 oder 5 gleichen oder verschiedenen Substituenten ausgewählt aus der Gruppe umfassend eine Ci-5-Alkyl, -OH und -O-Ci-5-Alkyl-Gruppe substituiert sein können;
und
ungesättigte oder gesättigte, unsubstituierte, lineare, verzweigte oder cyclische Ester, Alkohole und Aldehyde, die 5 bis 20, bevorzugt 5 bis 16, Kohlenstoffatome und ggf. 1 , 2 oder 3 Heteroatom(e) ausgewählt aus der Gruppe bestehend aus Sauerstoff und Schwefel als Ketten- bzw. Ringglied(er)er aufweisen, vor.
Besonders bevorzugt enthält die erfindungsgemäße, pharmazeutische Zusammensetzung als Duftstoff bzw. Aromazusatz wenigstens eine natürliche oder naturidentische Verbindung ausgewählt aus der Gruppe umfassend Anethol, Benzaldehyd, Benzylacetat, Benzylalkohol, Benzylformiat, iso-Bornylacetat, Campher, Neral, Citronellal, Citronellol, Citronellylacetat, para-Cymen, Decanal, Dihydrolinalool, Di- hydromyrcenol, Dimethylphenylcarbinol, Eukalyptol, Geraniol, Geranylacetat, Gera- nylnitril, cis-3-Hexenylacetat, Hydroxycitronellal, Limonen, Linalool, Linalooloxid, Linalylacetat, Linalylpropionat, Methylanthranilat, alpha-Methylionon, Methyl nonyla- cetaldehyd, Methylphenylcarbinylacetat, Menthon, iso-Menthon, Myrcen, Myrcenyla- cetat, Myrcenol, Nerol, Nerylacetat, Nonylacetat, Phenylethylalkohol, alpha-Pinen, beta-Pinen, gamma-Terpinen, alpha-Terpinol, beta-Terpinol, Terpinylacetat, para- tert-Butylcyclohexylacetat, alpha-Amylzimtaldehyd, Amylsalicylat, Caryophyllen, Cedren, Zimtalkohol, Dimethylbenzylcarbinylacetat, Ethylvanillin, Eugenol, iso-Euge- nol, Tricyclodecenylacetat, Piperonal, 3-cis-Hexenylsalicylat, Hexylsalicylat, Lilial, gamma-Methylionon, Nerolidol, Patschulialkohol, Phenylhexanol, beta-Selinen, Trichlormethylphenylcarbinylacetat, Triethylcitrat, Vanillin, Dimethoxybenzaldehyd, Benzophenon, Ethylenbrassylat, Galaxolid, Hexylzimtaldehyd, Lyral, Methylcedrylon, Methyl-beta-naphthylketon, Moschusketon, Phenylethylphenylacetat, Ambrettolid, Cyclohexylsalicylat, delta-Nonalacton, delta-Undecalacton, Dodecalacton, Ethylun- decylenat, Exaltolid, gamma-Undecalacton, Hexadecanolid, Myristicin und Moschus- xylol.
Weitere Duftstoffe bzw. Aromazusätze, welche die erfindungsgemäße, pharmazeutische Zusammensetzung umfassen kann, sind in David J. Rowe, Chemistry and Technology of Flavors and Fragrances, Taylor & Francis Group, 2004; Michael Edwards, Fragrances of the World 2005, Crescent House Pub, 2004 und David Pybus, The Chemistry of Fragrances, Royal Society of Chemistry, 1999 offenbart. Die entsprechenden Offenbarungen gelten hiermit als Teil der vorliegenden Offenbarung.
Als Duftstoff für die erfindungsgemäße, pharmazeutische Zusammensetzung kann auch wenigstens eine natürlich vorkommende Mischung von Duftstoffen bzw. Aromazusätzen verwendet werden. Insbesondere eignet sich wenigstens ein Duftstoffbzw. Aromazusatz-Mischung ausgewählt aus der Gruppe umfassend Rosmarinöl, Sandelholzöl, Veilchenöl, Zitronengrasöl, Lavendelblütenöl, Eukalyptusöl, Pfeffer-
minzöl, Kamillenöl, Nelkenblätteröl, Zimtöl, Thymianöl, Teebaumöl, Cajeputöl, Niaou- liöl, Manukaöl, Zitrusöl, Latschenkieferöl, Jasminöl, Geraniumöl, Kümmelöl, Fichten- nadelöl, Bergamotteöl, Terpentinöl, Linalolöl, Blutorangenöl, Cypressenöl, Edeltan- nenöl, Fenchelöl, Grapefruitöl, Ingweröl, Kiefernadelöl, Lavandinöl, Lemongrassöl, Limetteöl, Mandarinenöl, Melissenöl, Myrrhenöl, Patchouliöl, Rosenholzöl und Thu- jaöl genannt.
Der Duftstoff bzw. Aromazusatz wird vorzugsweise in einer Menge von 0,001 bis 2 Gew.-%, bezogen auf das Gesamtgewicht der erfindungsgemäßen, pharmazeutischen Zusammensetzung, eingesetzt.
Bevorzugt eingesetzte Farbstoffe für die erfindungsgemäße, pharmazeutische Zusammensetzung umfassen:
A) anorganische und organische Pigmente, wie beispielsweise Titandioxid,
Zirkonoxid, Ceroxid, Zinkoxid, Eisenoxid, Preussisch-Blau, Kohleschwärze, Calciumlake und Aluminiumlake.
B) fettlösliche Farbstoffe, wie beispielsweise Sudanrot, DC Red 17, DC Green 6,
Beta-Caroten, Sojaöl, Sudanbraun, DC Yellow 11 , DC Violet 2, DC Orange 5 und Quinoline Yellow.
C) wasserlösliche Farbstoffe, wie beispielsweise Eisensulfate, (Rhodamine), Methylenblau und natürliche Farbstoffe.
Der Farbstoff wird vorzugsweise in einer Menge von 0,001 bis 2 Gew.-%, bezogen auf das Gesamtgewicht der erfindungsgemäßen, pharmazeutischen Zusammensetzung, eingesetzt.
Die erfindungsgemäße, pharmazeutische Zusammensetzung zeichnet sich vorzugsweise dadurch aus, dass der/die Wirkstoff(e) 3ß-Hydroxy-chlormadinonacetat und/oder 3α-Hydroxy-chlormadinonacetat vorzugsweise in einer hydrophilen Komponente molekular dispers verteilt vorliegt(en).
Als hydrophile Komponente kommen vorzugsweise Zucker oder zuckerähnliche Substanzen, wie Saccharose, Lactose, Mannit, Zuckeralkohol, Harnstoff oder andere stickstoffhaltigen Verbindungen zur Anwendung.
Alternativ kann die erfindungsgemäße, pharmazeutische Zusammensetzung vorzugsweise den Wirkstoff 3ß-Hydroxy-chlormadinonacetat und/oder 3α-Hydroxy- chlormadinonacetat in einer rückfettenden Grundlage, ggf. in Kombination mit einer hydrophilen Komponente molekular dispers verteilt enthalten.
Durch eine derartige Zubereitung auf Basis einer rückfettenden Grundlage wird zusätzlich der Anregung zur Überproduktion von Fett durch die Haut entgegengewirkt, indem kontrolliert eine gewisse, zur Rekonstitution der Haut notwendige Menge Fett angeboten wird. Übliche rückfettende Zubereitungen sind dem Fachmann bekannt und umfassen beispielsweise Lipogele oder Öl-in-Wasser Emulsionen.
Die erfindungsgemäße, pharmazeutische Zusammensetzung kann als Flüssigkeit, Schaum, Creme, Gel, Paste, Balsam, Spray, Salbe, Lotion, Spülung („Conditioner"), Tonikum, Tinktur, Milch, Mus, Pulver zum Auflösen, Emulsion (Öl-in-Wasser, Was- ser-in-ÖI), Serum, Öl, Shampoo, Suspension wie Liposome oder Nanosome, oder als Dispersion formuliert sein.
Die Herstellung entsprechender, die erfindungsgemäße pharmazeutische Zusammensetzung enthaltender Formulierungen und die dabei üblicherweise zum Einsatz kommenden Hilfs- und Zusatzstoffe sind dem Fachmann bekannt. Dazu wird z.B. auf Bauer et al., "Lehrbuch der pharmazeutischen Technologie" 6. Auflage, WVG Stuttgart, 1999 verwiesen, deren einschlägige Offenbarungen hiermit als Offenbarung der vorliegenden Anmeldung eingeführt werden.
Besonders bevorzugt zeichnet sich die erfindungsgemäße, pharmazeutische Zusammensetzung dadurch aus, dass sie als Vehikel mindestens einen Puffer, mindestens ein Konservierungsmittel, mindestens einen Aromazusatz und/oder mindestens einen Hilfsstoff enthält und vorzugsweise als Creme, Salbe, Lotion, Gel, Tinktur, auftragbare Maske, streichfähige Masse, Gesichtswasser, Haarbalsam, Haarconditioner, Haargel, Haarkur, Haarwasser, Haarcreme oder Haarshampoo formuliert ist.
Ein weiterer Gegenstand der vorliegenden Erfindung betrifft eine kosmetische Zusammensetzung zur Pflege der Haut und gegebenenfalls auch der Haare durch topi-
sehe bzw. äußere Anwendung umfassend 0,01 bis 5 Gew.-%, vorzugsweise 0,01 bis 2 Gew.-%, von Sß-Hydroxy-chlormadinonacetat und/oder 3α-Hydroxy- chlormadinonacetat und wenigstens ein kosmetisches Vehikel, vorzugsweise in üblichen Mengen, wobei der Prozent-Mengenanteil von 3ß-Hydroxy- chlormadinonacetat und/oder 3α-Hydroxy-chlormadinonacetat jeweils auf das Gesamtgewicht der erfindungsgemäßen, kosmetischen Zusammensetzung bezogen ist.
Als kosmetische Vehikel sind vorzugsweise die üblichen Hilfsstoffe und/oder Zusatzstoffe für kosmetische Zusammensetzungen geeignet. Vorzugsweise können die vorstehend aufgeführten Hilfs- und Zusatzstoffe zum Einsatz kommen, die für die Formulierung von pharmazeutischen Zusammensetzungen verwendet werden. Diese Zusatz- bzw. Hilfsstoffe sind ebenso vorzugsweise physiologisch verträglich. Die Mengen der jeweiligen Komponenten sind vorzugsweise so gewählt, dass die erfindungsgemäße, kosmetische Zusammensetzung konform mit der EU Kosmetikrichtlinie 76/768/EEC bzw. der EU-Richtlinie 95/17/EC ist.
Wie bereits ausgeführt, wurde überraschend gefunden, dass sich die antiandrogenen Wirkstoffe 3ß -Hydroxy-chlormadinonacetat und 3α-Hydroxy-chlormadinonacetat durch eine 6-fach geringere Hautdurchdringung und einer 0,7-fache höheren antiandrogenen Wirksamkeit, verglichen mit CMA, auszeichnen. Durch diese vorteilhaften Eigenschaften wird bei einer topischen Behandlung von Androgenabhängigen Erkrankungen eine systemische Wirkstoffbelastung praktisch verhindert.
Ein weiterer Gegenstand der vorliegenden Erfindung ist daher die Verwendung von 3ß-Hydroxy-chlormadinonacetat und/oder 3α-Hydroxy-chlormadinonacetat zur Herstellung eines Arzneimittels zur topischen Behandlung von Haut, vorzugsweise menschlicher Haut und gegebenenfalls der Haare, vorzugsweise zur lokalen topischen Behandlung von Androgen-abhängigen Erkrankungen, ganz besonders bevorzugt zur Prophylaxe und/oder Behandlung von Androgen-abhängigen Erkrankungen wie Akne, Alopezie, Seborrhöe und/oder Hirsutismus.
Akne ist hauptsächlich eine Erkrankung des Talgdrüsenfollikels, die zunächst nichtentzündliche Komedonen hervorbringt, im späteren Verlauf aber auch eine
Reihe entzündlicher Effloreszenzen (u.a. Papeln, Pusteln und Knoten) entstehen lassen kann. Bei einigen Akneformen können Terminal- und Vellushaarfollikel betroffen sein.
Akne wird nach Ursache, Ausprägung bzw. Schweregrad und Lebensalter systematisiert.
Akne kann aber auch exogen („von außen") verursacht werden.
Die Acne inversa ist eine häufig schwere Entzündung der Talgdrüsen und Terminalhaarfollikel, vorzugsweise in intertriginösen Arealen wie z.B. Achselhöhle, Leistenregion und Gesäßfalte.
Nach Ausprägung bzw. Schweregrad unterscheidet man die Acne comedonica von der Acne papulopustulosa und der Acne conglobata. Das Hautbild der Acne comedonica ist von Komedonen und höchstens wenigen, sehr leichten Entzündungen (Papeln) geprägt. Narbenbildung ist nicht zu befürchten. Bei der Acne papulopustulosa dominieren Papeln und Pusteln, Narbenbildung in Folge der Entzündungsprozesse ist möglich. Die Acne conglobata bezeichnet das zusätzliche Auftreten von Fistelkomedonen und Knoten, in Folge droht erhebliche Vernarbung. Akne kann sowohl in akuter als auch in chronischer Form auftreten.
Alopezie / Alopecia bezeichnet ganz allgemein eine sichtbare Lichtung des Kopfhaars, d.h. ein Zustand mit abnorm "schütterem" Haupthaar oder mit haarlosen Hautbezirken im Sinne einer Glatze.
Die häufigst auftretende Form der Alopezie umfasst den androgenetischen Haarausfall (Alopecia androgenetica).
Seborrhöe ist ein medizinischer Begriff für die krankhaft veränderte Absonderung der Talgdrüsen. Er wird mit Mangel an Biotin (Vitamin B7) sowie Vitamin B6 (Pyridoxin) in Verbindung gebracht. Der Begriff wird manchmal auch für das Seborrhoische Ekzem benutzt. Patienten mit Seborrhöe leiden oft auch unter zu trockener Haut und Verhornungen. Seborrhoe oleosa bewirkt eine ölige Beschaffenheit der Haut, Se- borrhoe sicca manifestiert sich in kleieförmigen, fettigen Schuppungen. Die Seborr-
höe fördert unter anderem das Auftreten von Rosacea, Acne vulgaris und Acne necroticans.
Nach der Internationalen statistischen Klassifikation der Krankheiten und verwandter Gesundheitsprobleme (ICD - international Classification of Diseases) der Weltgesundheitsorganisation (WHO) wird Seborrhöe den Kategorien „Seborrhoisches Ekzem (Seborrhoische Dermatitis)" L21 und „Seborrhoische Keratose" L82 zugeteilt, wobei die Klasse L21 weiter in die Unterklassen L21.0 (Seborrhoea capitis, seborrhoischer Milchschorf), L21.1 (Seborrhoisches Ekzem der Kinder), L21.8 (Sonstiges seborrhoisches Ekzem), L21.9 (Seborrhoisches Ekzem, nicht näher bezeichnet) unterteilt ist und die Klasse L82 die Krankheiten Dermatosis papulosa nigra und das Leser- Tre/af-Syndrom umfasst.
Hirsutismus (ICD-Klasse L68.0) ist übermäßiger Haarwuchs nach männlichem Verteilungsmuster bei Frauen, der entweder durch die vermehrte Bildung männlicher Sexualhormone (Testosteron) oder aber durch psychogene Faktoren wie sozialer Stress oder ein Angstsyndrom ausgelöst werden kann.
Besonders bevorzugt ist die Verwendung von 3ß-Hydroxy-chlormadinonacetat und/oder 3α-Hydroxy-chlormadinonacetat zur Herstellung eines Arzneimittels zur Prophylaxe und/oder Behandlung von Seborrhöe, Akne, Alopezie, und/oder Hirsutismus, wobei eine derartige Prophylaxe und/oder Behandlung vorzugsweise eine lokale, topische Behandlung der Haut und/oder der Haare und/oder zur Förderung des Haarwuchses sowohl bei weiblichen wie auch bei männlichen Patienten umfasst. Besonders bevorzugt ist die vorzugsweise lokale, topische Anwendung des Arzneimittels zur Behandlung von Seborrhöe, Acne vulgaris, androgenetische Alopezie, und/oder Hirsutismus.
Das erfindungsgemäße Arzneimittel enthält vorzugsweise 0,01 bis 20, bevorzugter 0,01 bis 10 und ganz besonders bevorzugt 0,01 bis 5 Gew.-% an 3ß -Hydroxy- chlormadinonacetat und/oder 3α-Hydroxy-chlormadinonacetat, bezogen jeweils auf das Gesamtgewicht des erfindungsgemäßen Arzneimittels.
Ein weiterer Gegenstand der vorliegenden Erfindung ist die Verwendung einer - wie vorstehend beschriebenen - kosmetischen Zusammensetzung zur Pflege der Haut und auch gegebenenfalls der Haare durch topische Anwendung, vorzugsweise von Menschen, vorzugsweise zur Vorbeugung von und/oder Anwendung bei Seborrhöe und/oder Akne, vorzugsweise Acne vulgaris, androgenetischer Alopezie und/oder zur Förderung des Haarwuchses sowohl bei weiblichen wie auch bei männlichen Patienten.
Für eine topische Behandlung wird die erfindungsgemäße, pharmazeutische oder kosmetische Zusammensetzung vorzugsweise mindestens einmal täglich, besonders bevorzugt 3-mal täglich und ganz besonders bevorzugt 2-mal täglich auf die menschliche Haut und/oder auf die Haare aufgetragen.
Bevorzugt wird die erfindungsgemäße, pharmazeutische oder kosmetische Zusammensetzung auf die Kopfhaut, Gesichtshaut, die Haut des Oberkörpers, wie beispielsweise der Haut der Rückenpartie und/oder der Brustpartie und/oder auf die Haare aufgetragen.
Die nachfolgenden Beispiele dienen zur näheren Erläuterung der Erfindung, sind jedoch nicht einschränkend hinsichtlich ihres Umfangs auszulegen:
Beispiel 1 : Eine pharmazeutisch wirksame Lösung aus:
3ß-Hydroxy-chlormadinonacetat 2,0 g
Isopropylmyristat 10 g
EDTA (Ethylendiamintetraessigsäure) 0,025 g
Ethanol 96% 35,0 g
Transcutol (Diethylenglykolmonoethylether) 2,0 g

wird durch Vermischen und Auflösen der Komponenten hergestellt.
Beispiel 2: Eine kosmetische Lösung aus:
3ß-Hydroxy-chlormadinonacetat 1 ,0 g

wird durch Vermischen und Auflösen der Komponenten und anschließender Einstellung des pH-Wertes auf 5,5 mit Hilfe einer 10%-igen NaOH-Lösung hergestellt.
Beispiel 3: Bestimmung der Permeation von Chlormadinonacetat (CMA) und 3ß- Hydroxy-chlormadinonacetat durch humane Bauchhaut
Bei Bauchreduktionsoperationen angefallene humane Bauchhautstücke wurden in eine Petrischale mit Präparierlösung (Na+: 144 mmol/l, K+: 5,9 mmol/l, Ca2+: 3,7 mmol/l, Cl": 157 mmol/l) gegeben. Die Hautstücke wurden von der Coriumseite aus, unter Feuchthaltung mit der Präparierlösung, in mehreren Schritten bis zu einer Schichtdicke von ca. 300 μm abgetragen. Man erhielt dabei eine mechanisch stabile Hautpräparation, bestehend aus Epidermis mit ca. 150-200 μm Cutis. Die Schichtdicke der Hautpräparate wurde durch Ausmessen mit einer Bügelmessschraube der Firma MITUTOYO ermittelt. Die Fläche der verwendeten Hautpräparate betrug ca. 19,7 mm2.
Die Bestimmung der Permeation von CMA und 3ß-Hydroxy-chlormadinonacetat durch humane Bauchhaut erfolgte in einer Durchflusszelle, die schematisch in Figur 1 dargestellt ist. Hierbei wurde eine Rezeptorflüssigkeit (1 ) an der Unterseite des Hautpräparats (2) mit einer Peristaltikpumpe bei einer Geschwindigkeit von 270 μl pro Stunde vorbeigepumpt. Das Hautpräparat wurde zwischen dem Oberteil (3) und dem Unterteil (4) der Permeabilitätszelle (5) eingespannt. Von oben wurde jeweils Vehikel mit 3ß-Hydroxy-chlormadinonacetat (5%-ige Suspension in wässriger 0,05%- iger Polysorbat 80 Lösung) oder Chlormadinonacetat (5%-ige Suspension in wässriger 0,05%-iger Polysorbat 80 Lösung) auf die Haut aufgebracht, das an-
schließend durch das Hautpräparat (2) in die Rezeptorflüssigkeit (1 ) diffundiert. Das Hautpräparat wurde durch eine stationären Donatorflüssigkeit (9), die sich in einem Reservoir (7) befand befeuchtet. Der Innendurchmesser der Rezeptorflüssigkeits- Zuleitung (6) betrug 0,6 mm, das Rezeptorflüssigkeitsvolumen unter der Haut betrug ca. 80 μl. Die Rezeptorflüssigkeit (1 ) wurde in festgelegten Zeitintervallen mit Hilfe eines Fraktionssammlers (8) gesammelt und anschließend mittels HPLC analysiert. Durch diese Experimente erhielt man eine quantitative Aussage über die diffundierte Stoffmenge in Abhängigkeit von der Zeit.
Als Rezeptorflüssigkeit wurde ein wässriger, isotonischer Tris-Puffer (20 mmol/l) mit 20% Ethanol verwendet und mit NaCI auf 310 mosmol (0,9 Gew.%) eingestellt. Der pH-Wert der Rezeptorflüssigkeit betrug 7,35. Aus einem Pflaster, das den zu untersuchenden Wirkstoff enthielt, wurde mittels einer Lochstanze ein der Fläche des Hautpräparates entsprechendes Stück ausgestanzt und auf das Hautpräparat aufgeklebt. Die Versuchsdauer betrug 21 h und das Probenintervall betrug 3 h. Die Experimente wurden bei einer Versuchstemperatur von 34°C durchgeführt. Es wurden jeweils 3 unabhängige Messreihen mit CMA und 3ß-Hydroxy-chlormadinonacetat gemacht.
Die mittlere abgegebene Dosis (Mittelwert von 3 unabhängig durchgeführten Experimenten) durch humane Bauchhaut nach 24 Stunden war jeweils 4,4 μg für CMA und 0,7 μg für den Metaboliten 3ß-Hydroxy-chlormadinonacetat. Die Hautpermeationsfähigkeit von 3ß-Hydroxy-chlormadinonacetat ist somit weit geringer als die von CMA.

Claims
1. Verwendung von Sß-Hydroxy-chlormadinonacetat (17α-acetoxy-chloropregna- 4,6-dien-3ß-ol-20-on) und/oder Sα-Hydroxy-chlormadinonacetat (17α-acetoxy- chloropregna-4,6-dien-3α-ol-20-on) zur Herstellung eines Arzneimittels zur topischen Behandlung von Haut, vorzugsweise menschlicher Haut und gegebenenfalls der Haare.
2. Verwendung nach Anspruch 1 zur Herstellung eines Arzneimittels enthaltend 0,01 bis 20 Gew.-% an 3ß-Hydroxy-chlormadinonacetat und/oder 3α-Hydroxy-chlorma- dinonacetat.
3. Verwendung nach Anspruch 1 oder 2 zur topischen Behandlung von Androgenabhängigen Erkrankungen.
4. Verwendung nach einem der Ansprüche 1 bis 3 zur Prophylaxe und/oder Behandlung von Seborrhöe, Akne, vorzugsweise Acne vulgaris, Hirsutismus und/oder Alopezie, vorzugsweise androgenetischer Alopezie und/oder zur Förderung des Haarwuchses sowohl bei weiblichen wie auch bei männlichen Patienten.
5. Pharmazeutische Zusammensetzung zur Behandlung der Haut mittels topischer Anwendung umfassend 0,01 bis 20 Gew.-% von 3ß-Hydroxy-chlormadinonacetat und/oder 3α-Hydroxy-chlormadinonacetat und wenigstens ein pharmazeutisch verträgliches Vehikel.
6. Kosmetische Zusammensetzung zur Pflege der Haut durch topische Anwendung umfassend 0,01 bis 5 Gew.-% von 3ß-Hydroxy-chlormadinonacetat und/oder 3α- Hydroxy-chlormadinonacetat und wenigstens ein kosmetisches Vehikel.
7. Pharmazeutische oder kosmetische Zusammensetzung nach Anspruch 5 oder 6, dadurch gekennzeichnet, dass sie als Vehikel mindestens einen Puffer, mindestens ein Konservierungsmittel, mindestens einen Aromazusatz und/oder mindestens einen Hilfsstoff enthält.
8. Pharmazeutische oder kosmetische Zusammensetzung nach einem der Ansprüche 5 bis 7, dadurch gekennzeichnet, dass die pharmazeutische oder kosmetische Zusammensetzung als Creme, Salbe, Lotion, Gel, Tinktur, auftragbare Maske, streichfähige Masse, Gesichtswasser, Haarbalsam, Haarconditioner, Haargel, Haarkur, Haarwasser, Haarcreme oder Haarshampoo formuliert ist.
9. Pharmazeutische oder kosmetische Zusammensetzung nach Anspruch 7 oder 8, dadurch gekennzeichnet, dass das Konservierungsmittel wenigstens eine Verbindung ausgewählt aus der Gruppe umfassend Antioxidantien, Reduktionsmittel und UV-Absorber ist.
10. Pharmazeutische oder kosmetische Zusammensetzung nach einem der Ansprüche 7 bis 9, dadurch gekennzeichnet, dass der/die Wirkstoff(e) 3ß-Hydroxy- chlormadinonacetat und/oder 3α-Hydroxy-chlormadinonacetat in einer hydrophilen Komponente molekular dispers verteilt vorliegt(en).
11. Pharmazeutische oder kosmetische Zusammensetzung nach einem der Ansprüche 7 bis 10, dadurch gekennzeichnet, dass darin der Wirkstoff 3ß-Hydroxy- chlormadinonacetat und/oder 3α-Hydroxy-chlormadinonacetat in einer rückfettenden Grundlage, ggf. in Kombination mit einer hydrophilen Komponente molekular dispers verteilt, vorliegt.
12. Pharmazeutische oder kosmetische Zusammensetzung nach einem der Ansprüche 7 bis 11 , dadurch gekennzeichnet, dass die Zusammensetzung einen Penetrationspromotor, vorzugsweise eine Verbindung ausgewählt aus der Gruppe umfassend Säureamide und/oder Amine, enthält.
13. Pharmazeutische oder kosmetische Zusammensetzung nach einem der Ansprüche 7 bis 12, dadurch gekennzeichnet, dass die Zusammensetzung wenigstens einen Farbstoff und/oder einen Duftstoff enthält.
14. Pharmazeutische Zusammensetzung nach einem der Ansprüche 7 bis 13, geeignet zu einer mindestens einmal täglichen, topischen Behandlung der menschlichen Haut, vorzugsweise der Kopfhaut, Gesichtshaut, des Oberkörpers, der Haut der Rückenpartie und/oder Brustpartie, und gegebenenfalls der Haare.
15. Kosmetische Zusammensetzung nach einem der Ansprüche 7 bis 13, geeignet zu einer mindestens einmal täglichen Pflege der menschlichen Haut, vorzugsweise der Kopfhaut, Gesichtshaut, des Oberkörpers, der Haut der Rückenpartie und/oder Brustpartie, und gegebenenfalls der Haare.
16. Verwendung einer kosmetischen Zusammensetzung nach einem der Ansprüche 7-13, zur Vorbeugung von und/oder Anwendung bei Seborrhöe und/oder Akne, vorzugsweise Acne vulgaris, androgenetischer Alopezie und/oder zur Förderung des Haarwuchses sowohl bei weiblichen wie auch bei männlichen Patienten.
