Abstract

Es wird ein neues Verfahren zur Herstellung der als Zwischenprodukt für die Synthese von Röntgenkontrastmitteln verwendeten Triiodtrimesinsäure beschrieben.

Description

Verfahren zur Herstellung von Triiodtrimesinsäure
Triiodtrimesinsäure (Formel I )ist ein wichtiges Zwischenprodukt für die Herstellung von nichtionischen triiodierten Röntgenkontrastmitteln. So wird beispielsweise in der DE 3001292 (Schering) die Herstellung von Trisamiden der Triiodtrimesinsäure beschrieben. Derartige Verbindungen sind aufgrund ihrer günstigen pharmakologischen Eigenschaften besonders interessant. Die wirtschaftliche Nutzung dieser Trisamide scheiterte daran, daß bis jetzt noch kein kommerziell nutzbarer Zugang zur eigentlichen Ausgangsverbindung, der Triiodtrimesinsäure, gefunden wurde. Ein Kontrastmittel mit besonders guten Eigenschaften, das losimid , das bereits in Phase III der klinischen Prüfungen war, scheiterte in der weiteren Entwicklung an den zu hohen Herstellungskosten der Triiodtrimesinsäure.
So wird in DE 2831496 die Herstellung der Triiodtrimesinsäure, ausgehend von Nitroisophthalsäure, beschrieben:
Bei diesem Verfahren wird Nitroisophthalsäure zur Aminoverbindung hydriert und anschließend mit Chlorjod iodiert. Die Einführung der fehlenden Carboxyl-Gruppe gelingt über eine Sandmeyer-Reaktion (HN02/ KCN / CuCN). Dieser Reaktionsschritt erwies sich besonders bei der Ansatzvergrößerung als sehr kritisch, da zum einen Blausäure entsteht und außerdem die Kupferionen im Uberschuss eingesetzt werden müssen. Die Beseitigung von Kupferabfällen aus den Reaktionswässern ist im Großmaßstab als besonders kritisch anzusehen. Ein kritischer Schritt ist auch die vollständige Verseifung des Nitrils zur Carbonsäure. Hierbei wird intermediär das Amid durchlaufen, das sich als sehr schwer hydrolysierbar erwies.
In einer weiteren Patentschrift (NYCOMED: US 5,882,628) werden Zwischenstufen beschrieben, die als Ausgangsprodukte für die Synthese von Triiodtrimesinsäure in Betracht kämen:
Triiodmesitylen wird unter acetylierenden Bedingungen in einer
Oxidationsreaktion mit Kaliumpermanganat / Acetanhydrid / Essigsäure/ Schwefelsäure in das
Triacetat überführt (Ausbeute: 35 %). Das Triacetat wird isoliert und mit
Kaliumcarbonat in Methanol zum Trisalkohol verseift (Ausbeute: 94 %). Der
Trisalkohol wird dann durch Swem-Oxidation in Dimethylsulfoxid als
Lösungsmittel in 67 % Ausbeute zum Trisaldehyd umgesetzt.
Die weitere Oxidation zur Tricarbonsäure ist in diesem Patent nicht beschrieben worden, und auch in der Literatur wird über diesen Schritt nichts berichtet.
Es bestand daher der Bedarf nach einer neuen Synthese, welche Triiodtrimesinsäure in hoher Gesamtausbeute unter günstigen Umwelt- und Sicherheitsaspekten liefert und das auch die Fähigkeit zur Ansatzvergrößerung besitzt. Diese Anforderungen sind überraschenderweise mit der neuen, hier vorliegende zweistufigen Synthese gegeben. Das folgende Syntheseschema zeigt den von Triiodmesitylen ausgehenden neuen Syntheseweg: (ll) (I)
Hierbei wird ausgehend von Triiodmesitylen mit Kalium- oder Natriumpermanganat in einer Mischung aus Essigsäureanhydrid, Essigsäure und Schwefelsäure in einem Volumenverhältnis 15-30 : 10-20 : 1 ,25-3,5, bevorzugt 20 :15 :2,5 umgesetzt. Die Schwefelsäure wird in der Konzentration 70 % - 100 %, bevorzugt 95 - 100 % eingesetzt. Die Umsetzung erfolgt bei Temperaturen von 10 bis 120 °C, bevorzugt 20 - 100 °C, besonders bevorzugt 40-80°C. Die Reaktϊonsdauer beträgt 12 - 36 Stunden, bevorzugt 15 - 25 Stunden.
Dann wird abdestilliert. Dieses kann direkt aus dem Ansatz erfolgen, wobei gegebenenfalls der Druck reduziert wird. Man erhält dabei ein Gemisch aus Essigsäure und Acetanhydrid. Man kann aber auch erst Wasser zugeben um das überschüssige Essigsäureanhydrid zu zerstören und destilliert dann reine Essigsäure ab. Die zurückgewonnene Essigsäure kann wiederverwendet werden. Dann neutralisiert man im Rückstand durch Zugabe einer anorganischen Base wie Natronlauge, Kalilauge oder Calciumhydroxid in fester Form oder, bevorzugt, als wäßrige Lösung die Schwefelsäure und setzt das Einengen fort. Nach beendetem Eindampfen wird bei Temperaturen zwischen 60 - 100 °C Wasser zugesetzt, anschließend ausgerührt und das ausgefallene 1 ,3,5-Triiod- 2,4-diacetoxymethylen-6-methylbenzol (II) abfiltriert, welches mit Wasser und Methanol gewaschen und anschließend getrocknet wird.
Im nächsten Reaktionsschritt wird das 1 ,3,5-Triiod-2,4-diacetoxymethylen-6- methylbenzol (II) in Wasser bei Temperaturen von 60 - 100 °C, bevorzugt 80 - 100 °C (Rückfluß) vorgelegt und mit einer wäßrigen Lösung einer anorganischen Base, bevorzugt Natronlauge, Kalilauge ,CaIciumhydroxid, Natriumcarbonat oder Kaliumcarbonat, besonders bevorzugt aber NaOH oder KOH versetzt. Die Zutropfdauer der Base beträgt 30 Minuten bis 10 Stunden, bevorzugt 30 Minuten bis 3 Stunden. Anschließend wird 1 bis 24 Stunden, bevorzugt 3 - 12, besonders bevorzugt 4 - 8 Stunden bei Temperaturen zwischen 60 und 100 °C , besonders bevorzugt 80 - 100 °C, gerührt. Anschließend wird mit einer anorganischen Säure (pH = 6-7), bevorzugt Clorwasserstoffsäure, Bromwasserstoffsäure oder Schwefelsäure, besonders bevorzugt aber konzentrierte Chlorwasserstoffsäure neutralisiert, zur heißen Reaktionslösung Mangnesiumsulfat zugegeben sowie eine wäßrige Kalium- bzw. Natriumpermanganat-Lösung zugetropft und anschließend wird 1 bis 24 Stunden, bevorzugt 1 - 12, besonders bevorzugt 2 - 4 Stunden bei Temperaturen zwischen 60 und 100 °C , besonders bevorzugt 80 - 100 °C, gerührt. Die anschließende Aufarbeitung kann auf unterschiedliche Weise erfolgen:
1. Der Uberschuss an Oxidationsmittel kann durch Zugabe eines Reduktionsmittels in fester oder gelöster Form (wäßrige Lösung) zerstört werden. Hierzu eignen sich insbesondere anorganische Sulfite oder Hydrogensulfite, wie Natriumsulfit/hydrogensulfit oder Kaliumsulfit hydrogensulfit, besonders bevorzugt Natriumsulfit oder niedere Alkohole wie Methanol, Ethanol, Isopropanol, Glycol. Dieses geschieht bei Temperaturen zwischen 10 bis 100 °C bevorzugt 20 - 80 °C. Anschließend wird durch Zugabe von Schwefelsäure auf einen pH Wert von 0,1 - 3, bevorzugt 0,5 - 1 gestellt und das Produkt durch Extraktion mit einem organischen Lösungsmittel wie Ethylacetat, Propylacetat, Methyl -butylether (MTB), Tetrahydrofuran ( THF), n-Butanol, Methyl-THF, Dichlormethan, Toluol extrahiert. Bevorzugt sind Ethylacetat und MTB. Die organischen Phasen können gegebenenfalls mit Wasser, Sole oder angesäuertem Wasser gewaschen werden und zur Trockene eingedampft werden. Als besonders günstig hat sich erwiesen, direkt auf das zur Kristallisation verwendete Lösungsmittel umzudestillieren . Dabei wird während der Destillation durch kontinuierliche Zugabe eines zweiten Lösungsmittels das erste durch das zweite Lösungsmittel ersetzt. Als Lösungsmittel zur Kristallisation haben sich besonders Cyclohexan und n-Heptan, sowie deren Mischungen mit Ethylacetat, Toluol und MTB bewährt. In manchen Fällen kann auch auf eine wie oben beschriebene Extraktion mit einem organischen Lösungsmittel verzichtet werden. Hierzu wird weitgehend unter reduziertem Druck zur Trockene eingeengt und anschließend Wasser durch Zugabe eines azeotropbildenden Lösungsmittels wie Methanol, Ethanol, Isopropanol, Dichlormethan, MTB, Ethylacetat, Butylacetat, Butanol, Toluol oder THF weitgehend entfernt . Die Restwassermenge wird durch Karl- Fischer-(KF) Titration bestimmt.. Dann wird der zurückbleibende Rückstand mit einem organischen Lösungsmittel, wie Methanol, Ethanol, Isopropanol, Dichlormethan, MTB, Ethylacetat, Buthylacetat, Butanol, Toluol oder THF bei Temperaturen zwischen 20 - 100 °C ausgerührt und von den Salzen abfiltriert. Das Filtrat kann zur Trockene eingedampft werden oder aber, wie oben beschrieben, auf ein zur Kristallisation geeignetes Lösungsmittel umdestilliert werden.
2. In manchen Fällen hat es sich als vorteilhaft erwiesen, kein Redukionsmittel zuzugeben, sondern durch Zugabe von Schwefelsäure den pH Wert auf 0,1 - 3, bevorzugt aber 0,1 - 1 zu stellen und dann wie oben beschrieben unter reduziertem Druck zur Trockene einzudampfen und den Rückstand mit einem organischen Lösungsmittel auszurühren (analoge Vorgehensweise wie unter 1 -)
Das hier beschriebene Syntheseverfahren zeichnet sich durch einige wichtige Vorteile gegenüber dem Verfahren des Stands der Technik aus: Als alleiniges Oxidationsmittel wird das billige und ungiftige Kalium - bzw. Natriumpermanganat verwendet. Die entstehenden Manganabfälle können in einem Recycling-Verfahren durch Aufoxidation wiederverwendbar gemacht werden. Das Verfahren ist robust und ohne weiteres im Multi-Tonnen-Maßstab durchführbar. Es zeichnet sich durch umweltfreundliche Lösungsmittel aus. Das Ver-. fahren ist billig in der Durchführung, besteht nur aus zwei Stufen und liefert eine hohe Gesamtausbeute von 80 - 85 % der Theorie Dieses Verfahren stellt somit einen wertvollen Beitrag zur Herstellung einer Zwischenstufe der für die medizinische Diagnostik wichtigen triiodierten Trisamide dar.
Die nachfolgenden Beispiele dienen zur Beschreibung des neuen Verfahrens: Herstellung von Trijodtrimesitylen
Erfolgte nach Synthesis 6, 486 (1980); WO 96/09282, J. Med.Chem. (2000) 43 (10), 1940 oder nach Synlett (2002), (4), 598
Herstellung von 1 ,3,5-Triiod-2.4-diacetoxymethylen-6-methylbenzol (II)
400 g (803 mmol) Triiodmesitylen werden in 0,9 I Acetanhydrid, 1 ,4 I Essigsäure und 121 ml konz. Schwefelsäure suspendiert und bei 40 °C werden 160 g (1 ,01 mol) Kaliumpermanganat portionsweise (über 2 Stunden) hinzugegeben und 18 Stunden bei 40 °C gerührt. Nach beendeter Reaktion gibt man unter Rühren vor- sichtig 152 ml Wasser zu und rührt 2 Stunden bei Raumtemperatur nach. Dann gibt man 363.2 ml 50 %ige Natronlauge zu (exakt die Menge um die Schwefelsäure zu neutralisieren!). Anschließend wird unter reduziertem Druck weitgehend abdestilliert (eingeengt). Man tropft unter Rühren 4 I Wasser zu und rührt anschließend eine Stunde bei 10 °C. Herstellung von Triiodtrimesinsäure (I) (Variante A) 250 g (407 mmol) 1 ,3,5-Triiod-2,4-diacetoxymethylen-6-methylbenzol werden in 2,5 I Wasser suspendiert und bei 90 °C vorsichtig mit 200 ml 50 %ige Natronlauge versetzt und 5 h bei 90 °C gerührt. Anschließend wird mit konz. Chlorwasserstoffsäure neutralisiert (pH Wert = 6-7), mit 367 g (1 ,488 mol) Magnesiumsulfat versetzt, eine Lösung aus 411 g (2,604 mol)
Kaliumpermanganat in 4 I Wasser zugetropft und 2 h unter Rückfluß gerührt. Man kühlt auf Raumtemperatur (RT) ab, gibt 50 g Natriumsulfit zu und rührt eine Stunde bei Raumtemperatur. Dann wird mit 50 % Schwefelsäure auf einen pH Wert von 1 gestellt. Man gibt 2 I Ethylacetat zu und rührt gut durch. Man trennt die organische Phase ab und extrahiert die Wasserphase 2 mal mit 0,5 I Ethylacetat nach. Die organischen Phasen werden vereinigt, einmal mit 2,5 I Wasser nachgewaschen und anschließend auf Cyclohexan umdestilliert. Beim Abkühlen (0 °C) kristallisiert das Produkt aus. Ausbeute: 206 g (86 % der Theorie)

Herstellung von Triiodtrimesinsäure (I) (Variante B)
250 g (407 mmol) 1 ,3,5-Triiod-2,4-diacetoxymethylen-6-methylbenzol werden in 2,5 I Wasser suspendiert und bei 90 °C vorsichtig mit 200 ml 50 %ige Natronlauge versetzt und 5 h bei 90 °C gerührt. Anschließend wird mit konz. Die organischen Phasen werden vereinigt, einmal mit 2,5 I Wasser nachgewaschen. Die Essigesterlösung wird andestilliert um azeotrop Wasser zu entfernen (Wassergehalt nach Karl-Fischer-Titration < 0,2 %). Eine so hergestellte Lösung kann zur weiteren Umsetzung eingesetzt werden (z.B. Herstellung des Säurechlorids mit SOCI2). Durch Eindampfen eines Aliquots wurde die Ausbeute bestimmt.

Herstellung von Triiodtrimesinsäure (I) (Variante C)
250 g (407 mmol) 1 ,3,5-Triiod-2,4-diacetoxymethylen-6-methylbenzol werden in 2,5 I Wasser suspendiert und bei 90 °C vorsichtig mit 200 ml 50 %ige Natronlauge versetzt und 5 h bei 90 °C gerührt. Bei einem Wassergehalt < 1 % (KF) gibt man weitere 10 I Isopropanol zu und rührt eine Stunde bei 40 °C. Man filtriert vom Salzbrei ab und spült 2 mal mit 3 I Isopropanol nach. Das Filtrat wird im Vakuum zur Trockne eingedampft.

Herstellung von Triiodtrimesinsäure (I) (Variante D)
250 g (407 mmol) 1 ,3,5-Triiod-2,4-diacetoxymethylen-6-methylbenzol werden in 2,5 I Wasser suspendiert und bei 90 °C vorsichtig mit 200 ml 50 %ige Natronlauge versetzt und 5 h bei 90 °C gerührt. Anschließend wird mit konz. Das Filtrat wird im Vakuum zur Trockne eingedampft.

Das Filtrat wird im Vakuum zur Trockne eingedampft.

Claims

Patentansprüche
1. Verfahren zur Herstellung von Triiodtrimesinsäure, dadurch gekennzeichnet, dass man in einem OxidationsverfahrenTriiodmesitylen mit Kalium- oder Natriumpermanganat in einer Mischung aus Essigsäureanhydrid, Essigsäure und Schwefelsäure bei Temperaturen von 10 bis 120° C innerhalb von 12 bis 36 Stunden umsetzt, anschließend das so erhaltene Zwischenprodukt 1 ,3,5- Triiod-2,4-diacetoxymethylen-6-methylbenzol isoliert, dieses dann mit Base 1 ,5 bis 34 Stunden bei Temperaturen zwischen 60 und 100 °C behandelt, danach die Reaktionslösung neutralisiert und dann mit einer wässrigen Kalium- oder Natriumpermaganat-Lösung bei Temperaturen von 60 bis 100 °C innerhalb von 1 bis 24 Stunden behandelt.
2. Verfahren nach Anspruch 1 , dadurch gekennzeichnet, dass die Umsetzung zum Zwischenprodukt bei 40 bis 80° C durchgeführt wird.
3. Verfahren nach Anspruch 1 , dadurch gekennzeichnet, dass die Umsetzung zum Zwischenprodukt innerhalb von 15 bis 25 Stunden durchgeführt wird.
4. Verfahren nach Anspruch 1 , dadurch gekennzeichnet, dass die basische Behandlung des Zwischenproduktes bei 80 bis 100° C durchgeführt wird.
5. Verfahren nach Anspruch 1 , dadurch gekennzeichnet, dass die basische Behandlung des Zwischenproduktes innerhalb von 3 bis 12 Stunden durchgeführt wird.
6. Verfahren nach Anspruch 1 , dadurch gekennzeichnet, dass die Oxidation des Zwischenproduktes bei 80 bis 100° C durchgeführt wird.
7. Verfahren nach Anspruch 1 , dadurch gekennzeichnet, dass die Oxidation des Zwischenproduktes innerhalb von 2 bis 4 Stunden durchgeführt wird.
