Abstract
Die vorliegende Erfindung betrifft Mittel zur topischen Verabreichung einer Verbindung der Formel (I) :(CH3)2CH-(CH2)3-CH(CH3)-(CH2)3-CH(CH3)-(CH2)3-C(A)(CH3)-C(B)2-C(O)-R, in der R Wasserstoff, OR1, N(OH)R1 oder NR2R3 darstellt, R1, R2 und R3 unabhängig voneinander Wasserstoff, C1 -C22-Alkyl, C1-C22-Alkenyl, C7-C12-Arylalkyl (insbesondere Benzyl, Phenethyl und Phenylpropyl), Retinyl, Tocopheryl, Ascorbyl oder einen von einer Aminosäure oder Peptid herstammenden Rest darstellt und A und B Wasserstoffatome darstellen oder A und ein Rest B eine Doppelbildung bilden und der andere Rest B ein Wasserstoffatom darstellt oder der Rest A ein Wasserstoffatom darstellt und die Reste B zusammen ein Sauerstoffatom bilden oder einer der Reste B eine Hydroxylgruppe darstellt und der andere Rest B und der Rest A Wasserstoffatome darstellen, enthaltend eine Verbindung der Formel (I) und einen pharmazeutisch und/oder kosmetisch verträglichen Träger, mit der Maßgabe, daß das Mittel kein Retinoid enthält.
Description
Topisches Mittel enthaltend Phytansäure oder ein Derivat davon
Die vorliegende Erfindung betrifft Mittel zur topischen Verabreichung, die Phytansäure oder ein Derivat davon enthalten. Die Mittel sind insbesondere geeignet zur Behandlung von Cellulite und/oder Unterhaut-Fettpolstern aber auch zur Behandlung von Hautalterung oder einer gestörten oder dysfunktionellen epidermalen Barriere.
Orangenhaut oder Cellulite ist ein verbreitetes ästhetisches Problem, an dem viele Frauen leiden. Cellulite, oder auch lokale Lipodystrophie entwickelt sich zunächst durch Veränderungen in der Lymph- und Blutzirkulation, was dann wiederum strukturelle Umgestaltungen im subkutanen Fettgewebe und der umhüllenden Collagenmatrix auslöst. Durch diese Vorgänge werden die Fettzellen von den Ernährungs- und den Entsorgungsbahnen abgeschnitten und schwellen zu Millimeter-kleinen Knötchen an, die von festem Collagengewebe umgeben werden. Diese Knötchen verbinden sich später zu größeren Aggregaten von bis zu 20 mm Durchmesser und drängen in die Dermis. Durch die umhüllenden Collagenfasern, die weiterhin in der Subkutis verankert bleiben, kommt es zu Einschnürungen, die als Ursache für das ästhetische Problem angesehen werden. In der späten Phase kann es überdies noch zu besonderem Schmerzgefühl kommen, da die Knoten die Nervenenden durch Druck reizen.
Die Cellulite wird in der Regel als kosmetisches Problem betrachtet, allerdings kann Cellulite auch als Erkrankung angesehen werden, die einer Therapie bedarf. Dies beruht darauf, daß insbesondere im Bereich der Lymphgefäße stark verfettete subkutane Gewebe auch Schmerzzustände hervorrufen können, die einer arzneilichen Behandlung bedürfen. Auch kann Cellulite daran leidende Personen stark psychisch belasten und eine entsprechende Behandlung erfordern.
Grundsätzlich werden bei der Vorbeugung und Behandlung von Cellulite im Stand der Technik zwei Verfahren unterschieden. Zum einen werden mechanische Behandlungen, wie z.B. Massagen angewendet und zum anderen bestimmte Formulierungen, die auf die
Haut aufgetragen werden. Solche, die auf die Haut aufgetragen werden, kann man weiterhin in drei Gruppen gliedern. Zu der ersten Gruppe gehören Mittel mit Wirkstoffen, die geeignet sind, eine Restrukturierung des Proteingeflechts zu fördern. Dazu gehören z.B. Retinoide (A. Kligman et al., Topical retinol improves cellulite, J. Dermatol. Treat. 10, 119- 126, 1999 und J. Invest. Dermatol. 96, 975 1991 , Topical all-trans retinoic acid stimulates Collagen synthesis). Diese Mittel sind jedoch ungeeignet, die Größe der Fettpolster positiv zu beeinflussen, und Retinoide sind für viele Patienten bei topischer Anwendung nicht gut verträglich.
Eine zweite Gruppe von Formulierungen enthält Wirkstoffe, die eine verbesserte Durchblutung hervorrufen. Bekannt sind hier vor allem Formulierungen mit Koffein. Wirksam sind diese Mittel vor allem in der Frühphase von Cellulite, wenn im subkutanen Fettgewebe noch eine hinlängliche Durchdringung von Blutgefäßen existiert. Im Zuge des Verlaufs nimmt jedoch die Anzahl der Blutgefäße im angeschwollenen Fettgewebe drastisch ab, und derartige Mittel verlieren damit an Wirksamkeit.
Eine dritte Gruppe von Methoden versucht, den Fettstoffwechsel vorteilhaft zu beeinflussen. Das Gleichgewicht zwischen Lipolyse und Lipogenese bestimmt die Größe der Fettpolster und ist damit ein wesentlicher Faktor, der, im Falle eines Ungleichgewichts zugunsten der Lipogenese, die Entstehung von Cellulite begünstigt. So beschreibt die WO 03/009826 den Einsatz von Steroiden zur Wiederherstellung dieses Gleichgewichts. Die Verwendung von Steroiden kann aber beträchtliche Nebenwirkungen hervorrufen.
Eine Reihe von Mitteln, die gegen Hautalterung oder bei einer gestörten Barrierefunktion der Epidermis (einer gestörten oder verletzten Hautbarriere) verwendbar sind, sind bekannt, beispielsweise aus der WO 01/43704 oder der WO 98/32444.
Die WO 01/43704 offenbart eine Reihe von Verbindungen, die topisch auf die Haut aufgebracht werden können und die die Biosynthese und/oder Bioaktivität von endogenen Chemikalien fördern. Insbesondere sollen die Verbindungen die Kommunikation zwischen Keratinozyten, Fibroblasten und anderen Zelltypen der Haut durch Aktivierung der Genexpression, die die zellulare Aktivität verbessert, vermitteln. Die WO 01/43704 nennt eine Reihe von Verbindungen, die eine derartige Aktivität aufweisen, darunter auch Phytol und Derivate davon. Die Druckschrift betrifft weder Phytansäure noch die Behandlung von Cellulite und Unterhaut-Fettpolstern.
Die WO 98/32444 betrifft ein Verfahren zur Behandlung der Epidermis bei einem Patienten, bei dem die Barrierefunktion der Epidermis gestört ist und schlägt als Wirkstoff einen Aktivator für bestimmte Rezeptoren vor. Weder Phytansäure noch Derivate davon werden in dieser Druckschrift offenbart, und auch die Behandlung von Cellulite und/oder Unterhaut- Fettpolstern ist nicht Gegenstand der WO-Schrift.
Die WO 01/64177 beschreibt die Verwendung von Flavonen oder Isoflavonen zur Behandlung von Cellulite.
Die DE 199 40 415 beschreibt, daß natürliche Fettsäuren des Isoprenid- und Acetogenintyps mit Methyl- und Ethylverzweigungen und synthetische verzweigtkettige Fettsäuren als Diätetika und Zusatzstoffe zu Nahrungs- und Genußmitteln den Fettabbau bei Menschen fördern können. Die Druckschrift betrifft ausschließlich die systemische Aufnahme der Wirkstoffe, und die dort beschriebene Erfindung beruht darauf, daß die Wirkstoffe mit Bindungsstellen, die weit entfernt vom Behandlungsziel Haut liegen, interagieren. Eine topische Anwendung wird in dieser Druckschrift nicht beschrieben. Arzneimittel, die eine systemische Aufnahme benötigen, sind in der Regel völlig ungeeignet wenn man Hautzustände topisch behandeln will. Zum einen kommt es bei der systemischen Aufnahme zur bevorzugten Interaktion des Aktivstoffes mit Bindungsstellen, die weit entfernt vom Behandlungsziel liegen und ungezielte Interaktionen eröffnen zudem noch unerwünschte Seiteneffekte; zum anderen metabolisieren Wirkstoffe in der Haut zudem noch völlig anders, da in der Haut andere Enzymsysteme aktiv sind als z. B. in der Leber.
Die WO 01/66080 offenbart, daß Phytol die Wirkung von Retinoiden zur Erzielung einer Verbesserung es Hautzustands unterstützen kann. Insbesondere sollen die durch Phytol unterstützen Retinoide den Auswirkungen von Alterungsvorgängen in der Epidermis und Dermis entgegenwirken. Die Druckschrift geht davon aus, daß Phytol nach dem Auftrag auf die Haut in Phytansäure umgewandelt wird. In der Tat kann diese Umsetzung bei oraler Aufnahme in der Leber zu einem gewissen Grad stattfinden, neuere Untersuchungen haben aber ergeben, daß diese Umsetzung bei topischer Verabreichung auf die Haut nicht eintritt. Phytol ist daher bei topischer Verabreichung auf die Haut entgegen der Offenbarung der WO 01/66080 kein Vorläuferarzneimittel (Prodrug) der Phytansäure.
Phytansäure ist bekannt als potentieller RXR-Agonist wie dies in der WO 01/66080 beschrieben ist. Die Bindungsaffinität ist jedoch 200 fach schwächer im Vergleich zur Retinsäure, und kosmetische Effekte wie z.B. die von Retinsäure bekannte Anti-Falten- Wirkung können deshalb von Phytansäure, allein aus der sehr schwachen RXR-Bindung, nicht erwartet werden. Im Kampf um die RXR-Bindungsstelle würde Phytansäure auch gegenüber dem natürlich vorhandenen Ligand Retinsäure chancenlos ausgehen.
Die WO 01/66080 betrifft auch nicht die Behandlung von Cellulite und offenbart auch keine eigene, von Retinoiden unabhängige Wirksamkeit des Phytols bei topischer Verwendung. Bei der Behandlung von Hauterkrankungen mit Retinoiden können aber unerwünschte Nebenwirkungen auftreten.
Es besteht daher ein Bedarf an neuen Mitteln, die sowohl kosmetisch aber auch pharmazeutisch verwendet werden können und die nach topischer Anwendung insbesondere zur Behandlung von Cellulite und/oder Unterhaut-Fettpolstern wirksam sind. Die Mittel sollen ebenfalls die Alterung der Haut verlangsamen oder vorzugsweise rückgängig machen, das heißt insbesondere Hautfalten und Fältchen glätten, Altersflecken reduzieren und die mechanischen Eigenschaften der Haut wie Glattheit, Textur, Elastizität verbessern und den Hautton und die Farbgleichmäßigkeit verbessern. Die erfindungsgemäßen Mittel sollen ebenfalls bevorzugt eine angegriffene oder beschädigte Hautbarriere möglichst schnell reparieren und dadurch die Hautfeuchte verbessern, also insbesondere auch trockene Haut behandeln oder das Auftreten von trockener Haut verhindern.
Es ist Aufgabe der Erfindung, ein derartiges Mittel zur Verfügung zu stellen, das darüber hinaus noch möglichst arm an Nebenwirkungen ist und die Nachteile der bekannten Mittel des Standes der Technik nicht aufweist.
Die Lösung dieser Aufgabe beruht auf dem überraschenden Befund, daß Phytansäure und Derivate davon der Formel

In einer besonderen Ausführungsform ist einer der Reste R2 und R3 ein Wasserstoffatom und der zweite Rest wie vorstehend definiert.
In einer weiteren bevorzugten Ausführungsform ist einer der Reste B eine Hydroxylgruppe.
In einer weiteren bevorzugten Ausführungsform bilden beide Reste B ein Sauerstoffatom.
Die Arylalkylreste sind über die Alkyleinheit gebunden.
In einer besonders bevorzugten Ausführungsform betrifft die Erfindung die Verwendung von Verbindungen der Formel

In einer besonderen Ausführungsform der Verbindungen der Formel I und Il ist R Wasserstoff, OR1, NHR1 oder N(OH)R1 und R1 stellt Wasserstoff, C1-C22-AIkYl, C1-C22- Alkenyl, Benzyl, Phenethyl, Phenpropyl, Retinyl, Tocopheryl, Ascorbyl oder einen von einer Aminosäure oder einem Peptid herstammenden Rest dar, und A und B sind entweder beide Wasserstoffatome oder bilden zusammen eine Doppelbindung.
Bei den erfindungsgemäßen Mitteln handelt es sich einerseits um kosmetische Mittel, bei denen die Phytansäure bzw. das Derivat davon mit kosmetisch verträglichen Zusatzstoffen formuliert wird, als auch um pharmazeutische Mittel, bei denen die Phytansäure bzw. das Derivat davon mit pharmazeutisch verträglichen Zusatzstoffen formuliert wird. Sofern im Rahmen dieser Anmeldung nichts anderes dargelegt wird, sind offenbarte Zusatzstoffe sowohl kosmetisch verträgliche Zusatzstoffe als auch pharmazeutisch verträgliche Zusatzstoffe.
Erfindungsgemäß wird ebenfalls die Verwendung von Phytansäure und Derivaten davon wie vorstehend definiert zur Herstellung von topischen kosmetischen Mitteln und zur Herstellung von topischen Arzneimitteln zur Vorbeugung und/oder Behandlung von Cellulite, Unterhaut-Fettpolstern, Hautalterung, insbesondere Glättung von Hautfalten und Fältchen, Reduktion von Altersflecken, Verbesserung der mechanischen Eigenschaften der Haut wie z.B. Glattheit, Textur, Elastizität, Verbesserung des Hauttons und der Farbgleichmäßigkeit sowie zur Behandlung bzw. Reparatur einer geschädigten oder verletzten Hautbarriere zur Verfügung gestellt.
Erfindungsgemäß wird ebenfalls die kosmetische Verwendung von Phytansäure bzw. einem Derivat davon wie vorstehend definiert zur Behandlung von Cellulite, Unterhaut- Fettpolstern, Hautalterung, insbesondere Glättung von Hautfalten und Fältchen, Reduktion von Altersflecken, Verbesserung der mechanischen Eigenschaften der Haut wie z.B. Glattheit, Textur, Elastizität, Verbesserung des Hauttons und der Farbgleichmäßigkeit sowie zur Behandlung bzw. Reparatur einer geschädigten oder verletzten Hautbarriere zur Verfügung gestellt.
Die Verwendung von Phytansäure bzw. einem Derivat davon wie vorstehend definiert ist beispielsweise zur Behandlung und aktiven Prävention trockener Haut und zur Stärkung der Barrierefunktion der Haut sowie zur Behandlung, Pflege und Prophylaxe von sensibler Haut
und/oder zur Behandlung und Prophylaxe der Symptome einer negativen Veränderung der physiologischen Homöostase der gesunden Haut, insbesondere defizitärer, sensitiver oder hypoaktiver Hautzustände oder defizitärer, sensitiver oder hypoaktiver Zustände von Hautanhangsgebilden, entzündlicher Hautzustände sowie des atopischen Ekzems, der polymorphen Lichtdermatose, Psoriasis, Vitiligo, empfindlicher, juckender oder gereizter Haut, Veränderungen der normalen Lipidperoxidation, einer Veränderung des Ceramid-, Lipid- und Energiestoffwechsels der gesunden Haut, einer Veränderung des physiologischen transepidermalen Wasserverlustes, einer Verminderung der Hauthydratation und Abnahme des Feuchtigkeitsgehaltes der Haut, Veränderung des Natural Moisturizing Factor-Gehalts, Verminderung der Zell-Zell-Kommunikation, Mangelerscheinungen der intrazellulären DNS-Synthese, DNS-Schädigungen und Verminderung von endogenen DNS-Reparaturmechanismen, Aktivierung von Metalloproteinasen und/oder anderer Proteasen bzw. Inhibierung der entsprechenden endogenen DNS-Reparaturmechanismen und Abweichungen von den normalen post- translationalen Modifikationen von Bindegewebsbestandteilen.
Phytansäure reguliert die Sebumproduktion der Haut und beugt einer zu starken Produktion von Sebum vor. Im Bereich der Kopfhaut kommt es zu einer geringeren Rückfettung des Haares nach dem Waschen. Phytansäure-haltige Haarpflegemittel eignen sich deshalb gut für stark fettendes Haar oder für Kurzhaarfrisuren, bei denen man eine Rückfettung sehr schnell wahrnehmen kann. Vorteilhaft wirkt sich die Regulierung der Hautlipide auch auf der Kopfhaut aus, denn sie wirkt einer Schuppenbildung entgegen. Schuppenbildung wird insbesondere durch trockene Haut gefördert. Erfindungsgemäß kann Phytansäure deshalb auch in der Behandlung und Prophylaxe von Kopfhautschuppen eingesetzt werden.
Die Erfindung betrifft daher auch die Verwendung von Phytansäure bzw. einem Derivat davon wie vorstehend definiert als Haarpflegemittel, permanent oder auswaschbar, wie z.B. Haarkuren oder Shampoos, insbesondere die Behandlung und/oder Prophylaxe von fettigen Haaren und/oder Schuppenbildung.
Erfindungsgemäß wird die Phytansäure bzw. das Derivat davon nicht gemeinsam mit einem Retinoid eingesetzt, und die Phytansäure dient nicht zur Verstärkung eines weiteren Inhaltsstoffes, wie eines Retinoids. Die erfindungsgemäßen Mittel enthalten bevorzugt kein Retinoid. Erfindungsgemäß ebenfalls bevorzugt wird Phytansäure bzw. das Derivat davon
als alleiniger Wirkstoff zur Behandlung von Cellulite und/oder Unterhaut-Fettpolstern eingesetzt.

Soweit im Rahmen dieser Beschreibung von einem "Mittel" gesprochen wird, ohne daß eine genauere Spezifizierung erfolgt, wird hierunter sowohl ein kosmetisches Mittel als auch ein Arzneimittel verstanden. Zur Abgrenzung zwischen kosmetischen Mitteln und Arzneimitteln kann z.B. auf Römpp, Chemielexikon, 10te Auflage und die darin zitierte Literatur verwiesen werden.
Zur Definition des Begriffs "Retinoid" wird auf die WO 01/66080 verwiesen. Der Begriff "Retinoid" ist in der vorliegenden Anmeldung definiert wie in der WO 01/66080.
Der Begriff "Phytansäure", wie er im Rahmen dieser Beschreibung verwendet wird, bezieht sich auf die 3,7,11 ,15-Tetramethylhexadecansäure. Die Säure kommt natürlich in zwei Formen vor, in der 3R,7R,11 R-Form und in der 3S,7R,11 R-Form. Erfindungsgemäß bezieht sich der Begriff "Phytansäure" auf jede der natürlich vorkommenden Formen einzeln oder im Gemisch, sowie auf andere Formen der Phytansäure und auf Gemische einer oder mehrerer nicht natürlich vorkommender Formen der Phytansäure, gegebenenfalls im Gemisch mit einer oder beiden der natürlich vorkommenden Formen. Besonders bevorzugt wird erfindungsgemäß Phytansäure in einer natürlich vorkommenden Form oder im Gemisch aus beiden natürlich vorkommenden Formen verwendet. Phytansäure ist eine bekannte Verbindung und kommerziell erhältlich. Alle Epimere der Phytansäure sind umfaßt.
Neben der Verwendung von Phytansäure ist es erfindungsgemäß ebenfalls bevorzugt, ein Derivat der Phytansäure wie vorstehend definiert zu verwenden, insbesondere ein Derivat, das auf oder in der Haut oder vor oder während der Verabreichung vollständig oder teilweise in Phytansäure umgewandelt wird. Besonders bevorzugt handelt es sich bei den erfindungsgemäß eingesetzten Derivaten der Phytansäure um Ester der Phytansäure, insbesondere um Alkylester, z.B. CrCi0-Alkylester der Phytansäure. Als besonders bevorzugt können CrC6-Alkylester, insbesondere der Methylester, Ethylester, iso- Propylester, n-Propylester, n-Butylester und tert-Butylester genannt werden. Die Ester der Phytansäure können auf bekannte Art und Weise nach Standardmethoden aus der Phytansäure gewonnen werden. Geeignete Verfahren zur Herstellung der bevorzugten Ester der Phytansäure sind in den Beispielen beschrieben.
Erfindungsgemäß ebenfalls bevorzugt sind Verbindungen der Formel (I) wie vorstehend definiert, bei denen der Rest R1 ein CrC-io-Alkenylrest ist, insbesondere ein Ci-C6-
Alkenylrest. Der Alkenylrest hat bevorzugt weniger als drei Doppelbindungen, insbesondere eine oder zwei Doppelbindungen. Besonders bevorzugt sind auch Verbindungen der Formel (I) wie vorstehend definiert, bei denen der Rest R1 einen Ascorbylrest darstellt. Der Rest R1 kann auch ein Aminosäure- oder ein Peptidrest sein. Eine derartige Verbindung ist eine typische Proform für Phytansäure, da sie ein Substrat für Proteasen oder Amidasen darstellt, die die Verbindung in Phytansäure umwandeln. Sofern der Rest R ein Wasserstoffatom darstellt, handelt es sich bei der Verbindung der Formel (I) um Phytal, und erfindungsgemäß sind alle Epimere des Phytals ebenfalls umfaßt. Sofern die Reste A und B zusammen eine Doppelbindung bilden, handelt es sich um Phytensäurederivate, und auch von diesen Verbindungen sind alle Epimere und sowohl die E-Form als auch die Z-Form umfaßt. Bevorzugt stellen A und B aber Wasserstoffatome dar, und falls A und B gemeinsam eine Doppelbindung bilden, ist der Rest R1 bevorzugt eine Hydroxylgruppe, so daß die Verbindung dann Phytensäure selbst darstellt. Die Verbindungen können auf bekannte Art und Weise durch Anwendung chemischer Standardmethoden, z.B. ausgehend von der Phytansäure, hergestellt werden.
Verbindungen der Formel Il wie vorstehend definiert, in denen der Rest R OR1 darstellt und R1 ein n-Propyl oder ein C4-C22-Alkylrest ist, sind ebenfalls neu und die Erfindung betrifft auch diese neuen Verbindungen als solche.
Die Derivate der Phytansäure sind entweder selbst gegen Cellulite und/oder Unterhaut- Fettpolster aktiv oder werden vor, bei oder nach der topischen Anwendung in die aktive Phytansäure umgewandelt.
Die erfindungsgemäßen Mittel sind insbesondere zur kosmetischen oder arzneilichen Behandlung von Cellulite oder Unterhaut-Fettpolstern geeignet. Darüber hinaus sind sie auch ausgesprochen wirksam bei der Vorbeugung und Behandlung von Hautalterung, insbesondere bei der Glättung von Hautfalten und Fältchen, bei der Reduktion von Altersflecken, bei der Verbesserung von mechanischen Eigenschaften der Haut wie Glattheit, Textur und Elastizität und darüber hinaus verbessern sie den Hautton und die Farbgleichmäßigkeit. Die erfindungsgemäßen Mitteln eignen sich auch besonders vorteilhaft zur kosmetischen oder arzneilichen Behandlung einer gestörten oder verletzten Hautbarriere (epidermale Barriere) und dadurch hervorgerufene Zustände, wie sie beispielsweise in der WO 98/32444 genannt sind, insbesondere Fluid- oder Elektrolytanomalien, Hypothermie und Infektionen durch die Haut bei Frühgeborenen, die
jünger als 33 Wochen sind, Entzündung der Schleimhäute, wie z.B. Lippenentzündung, aufgesprungene Lippen, nasale Irritationen und Vulvovaginitis, ekzematöse Dermatitis, wie z.B. atopische oder seborrhoische Dermatitis, allergische Dermatitis oder nicht-allergische Kontaktdermatitis, rissiges Ekzema, photoallergische Dermatitis, phototoxische Dermatitis, Phytophotodermatitis, Strahlendermatitis und Stauungsdermatitis, Geschwüre und oberflächliche Hautdefekte, ausgelöst durch Trauma, Verbrennungen, bullöse Erkrankungen oder Haut- oder Schleimhautischämie, mehrere Formen der Ichthyose, Epidermolysis bullosa, Schuppenflechte, hypertrophe Narben und Wulstnarben, dermale Veränderungen der intrinsischen Alterung und Photoalterung, Blasenbildung durch Reibung hervorgerufen durch mechanisches Scheren der Haut und dermale Atrophie aufgrund der topischen Verwendung von Corticosteroiden.
Im Bezug auf die zu behandelnden Erkrankungen wird vollinhaltlich auf die WO 98/32444 Bezug genommen.
Die erfindungsgemäßen Mittel enthalten Phytansäure bzw. ein Derivat der Phytansäure und geeignete kosmetische und/oder pharmazeutisch verträgliche Zusatzstoffe.
Besonders bevorzugt ist der Wirkstoff, das heißt die Phytansäure oder das Derivat der Phytansäure, in den erfindungsgemäßen Mitteln in einer Menge von 0,0001 Gew.-% bis etwa 50 Gew.-%, bezogen auf das Gesamtgewicht der Zusammensetzung, vorhanden. Stärker bevorzugt ist die Phytansäure bzw. das Derivat der Phytansäure in einer Menge von 0,01 Gew.-% bis etwa 20 Gew.-%, noch stärker bevorzugt in einer Menge von etwa 0,1 Gew.-% bis etwa 15 Gew.-%, z.B. 1 bis etwa 5%, bezogen auf das Gesamtgewicht der Zusammensetzung, vorhanden.
Die erfindungsgemäßen Mittel umfassen ein oder mehrere kosmetisch verträgliche bzw. pharmazeutische verträgliche Träger und/oder Zusatz- bzw. Wirkstoffe, wie sie üblicherweise in derartigen Zubereitungen verwendet werden. Hier können beispielhaft Fette, Öle, Wachse, Silikone, Emulgatoren, Alkohole, Polyole, Verdickungsmittel, anfeuchtende und/oder feuchthaltende Substanzen, Tenside, Weichmacher, Schaumbremsen, anionische, kationische, nicht-ionische oder amphotere Polymere, Alkanisierungs- oder Acidifizierungsmittel, Enthärter, Adsorbentien, Lichtschutzmittel, Elektrolyte, Sequestrierungsmittel, organische Lösungsmittel, Konservierungsmittel,
Bakterizide, Antioxidantien, Vitamine, Duftstoffe, Aromen, Süßungsmittel, Farbstoffe und Pigmente genannt werden.
Geeignete Zusammensetzungen sind z.B. flüssige oder feste ÖI-in-Wasser-Emulsionen, Wasser-in-ÖI-Emuisionen, Mehrfachemulsionen, Mikroemulsionen, PIT-Emulsionen, Pickering-Emulsionen, Hydrogele, alkoholische Gele, Lipogele, ein- oder mehrphasige Lösungen, Schäume, Salben, Pflaster, Suspensionen, Puder, Cremes oder andere übliche Mittel. Die erfindungsgemäßen Mittel können auch in wasserfreier Form, wie beispielsweise als Öl oder Balsam, formuliert sein, z.B. mit pflanzlichen oder tierischen Ölen, Mineralölen, synthetischen Ölen oder Mischungen davon als Trägerstoffe.
Geeignete Formulierungen zur Behandlung von Cellulite sind in der WO 01/64177 für die Wirkstoffe Flavone und Isoflavone beschrieben. Die dort beschriebenen topischen Formulierungen zur Behandlung von Cellulite eignen sich prinzipiell auch zur Formulierung von Phytansäure und der Derivate davon, wobei der Wirkstoff bzw. das Pflanzenextrakt aus den Formulierungen der WO 01/64177 durch Phytansäure bzw. ein Derivat davon ersetzt wird. Insoweit wird auf die Offenbarung der WO 01/64177 Bezug genommen.
Die erfindungsgemäßen Formulierungen enthalten bevorzugt einen oder mehrere übliche Fettstoffe, z.B. pflanzliche Öle, flüssige Paraffinöle, Isoparaffinöle, synthetische Kohlenwasserstoffe, Di-n-alkylester, Fettsäuren, Fettalkohole, Esteröle, Hydroxycarbonsäurealkylester, Dicarbonsäureester, Dioiester, symmetrische, unsymmetrische oder cyclische Ester oder Kohlensäure mit Fettalkoholen, Mono-, Di- und Trifettsäureester von gesättigten und/oder ungesättigten linearen und/oder verzweigten Fettsäuren mit Glycerin, Wachse und Siliconverbindungen. Geeignete Beispiele derartiger Fettstoffe sind in der WO 01/64177 offenbart, auf die insoweit Bezug genommen wird.
Die Fettstoffe sind in der Regel in einer Menge von 0,1 bis 50 Gew.-%, bevorzugt von 0,1 bis 20 Gew.-%, insbesondere von 0,1 bis 15 Gew.-% in dem erfindungsgemäßen Mittel vorhanden (jeweils bezogen auf das gesamte Mittel).
Die erfindungsgemäßen Mittel können, wie die Mittel der WO 01/64177, eine oder mehrere oberflächenaktive Substanzen als Emulgatoren oder Dispergiermittel enthalten. Geeignete Beispiele derartiger Emulgatoren bzw. Dispergiermittel sind in der WO 01/64177 genannt, auf die insoweit Bezug genommen wird.
Die Emuigatoren können in den erfindungsgemäßen Mitteln beispielsweise in Anteilen von 0,1 bis 25 Gew.-%, stärker bevorzugt von 0,5 bis 15 Gew.-%, bezogen auf das gesamte Mittel, vorhanden sein.
Die erfindungsgemäßen Mittel können ebenfalls übliche Lichtschutzmittel, beispielsweise übliche UV-A- und/oder UV-B-Filter, enthalten. Eine Zusammenstellung üblicher UV-A- und UV-B-Filter, die auch in den erfindungsgemäßen Mittel eingesetzt werden können, findet sich beispielsweise in der EP-A 1 081 140. Erfindungsgemäß können selbstverständlich auch die in dieser Druckschrift erstmals offenbarten neuen Sonnenschutzfilter in den erfindungsgemäßen Mitteln eingesetzt werden.
Geeignete organische, mineralische oder modifizierte mineralische Lichtschutzfilter sind auch in der WO 01/64177 angegeben, auf die insoweit ebenfalls Bezug genommen wird.
Falls gewünscht, können die erfindungsgemäßen Mittel ebenfalls Proteinhydrolysate oder Derivate davon sowie geeignete Mono-, Oligo- oder Polysaccharide oder deren Derivate enthalten, wie es in der WO 01/64177 offenbart ist. Weitere geeignete Hilfs- und Zusatzstoffe wie Vitamine, Provitamine und Vitaminvorstufen, Alantopin, Bisabolol, Antioxidantien, Ceramide und Pseudoceramide, Triterpene, monomere Catechine, Verdickungsmittel, Pflanzenglycoside, strukturgebende Mittel (Strukturanten), Dimethylisosorbid, Lösungsmittel, Quell- und Penetrationshilfsstoffe, Parfümöle, Pigmente und Farbstoffe zum Anfärben des Mittels, Substanzen zur Einstellung des pH-Werts, Komplexbildner, Trübungsmittel, Perlglanzmittel, Treibmittel, filmbildende, emulsionsstabilisierende, verdickende oder adhäsive Polymere, insbesondere kationische, anionische sowie nicht-ionische Polymere, sind ebenfalls in der WO 01/64177 offenbart, auf die insoweit Bezug genommen wird.
Die Formulierung der erfindungsgemäßen Mittel erfolgt auf übliche Art und Weise. In den nachfolgenden Beispielen wird beispielhaft die Herstellung einer O/W-Emulsion beschrieben. Die Herstellung dieser und anderer Formulierungen ist dem Fachmann bekannt, und es kann hier auf übliche Formulierungshandbücher verwiesen werden.
Die erfindungsgemäßen Mittel sind derart formuliert, daß sie zur topischen Verabreichung geeignet sind. Die topische Verabreichung erfolgt bevorzugt mindestens einmal pro Tag,
z.B. zwei- oder dreimal am Tag. Die Behandlungsdauer beträgt in der Regel mindestens zwei Tage bis die gewünschte Wirkung erzielt wurde. Die Behandlungsdauer kann auch mehrere Wochen oder Monate betragen. Die Behandlung einer geschädigten Hautbarriere kann schon nach relativ kurzer Zeit wie 1 Tag bis 1 Woche erfolgreich beendet sein, während die Behandlung der Cellulite und der Hautalterung in der Regel 1 bis 2 Monate dauert.
Die Menge des aufzutragenden Mittels richtet sich nach der Konzentration des Wirkstoffs in dem Mittel sowie der Schwere der zu behandelnden Erkrankung bzw. dem gewünschten kosmetischen Erfolg. In der Regel ist bei einer pharmazeutischen Verwendung die anzuwendende Menge an Wirkstoff pro Anwendung höher als bei einer kosmetischen Verwendung. Eine geeignete Menge für die Anwendung hängt von der Beschaffenheit der Haut, der zu behandelnden Person sowie von der Schwere der zu behandelnden Cellulite und anderen Faktoren ab, die einem behandelnden Arzt oder Kosmetiker bekannt sind. Beispielsweise kann die Auftragung derart erfolgen, daß eine Creme auf die Haut aufgetragen wird. Eine Creme wird üblicherweise in einer Menge von 2 mg Creme / cm2 Haut aufgetragen. Für die Behandlung der Cellulite oder der Unterhaut-Fettpolster sollte eine Menge an Wirkstoff von etwa 10 μg bis 1 mg / cm2 Haut verwendet werden. Eine Creme zur Behandlung der Cellulite oder der Unterhaut-Fettpolster sollte daher 0,5 Gew. % bis 50 Gew. % Wirkstoff der Formel I enthalten. Für die Reparatur der Barrierefunktion der Haut reichen häufig schon geringere Wirkstoffmengen, wie 2 μg / cm2 Haut aus so dass die topischen Mittel eine entsprechend geringere Wirkstoffkonzentration aufweisen können. Die aufgetragene Menge ist aber nicht kritisch, und sollte sich mit einer bestimmten aufgetragenen Wirkstoffmenge kein Behandlungserfolg einstellen, kann die aufgetragene Menge ohne weiteres erhöht werden, beispielsweise, indem höher konzentrierte topische Formulierungen verwendet werden.
Üblicherweise enthalten die erfindungsgemäßen Mittel 0,05 bis 50 Gew. % Wirkstoff der Formel I stärker bevorzugt 0,1 bis 40 Gew. % , z. b. 0,5 bis 40 Gew. %. Die Wirkstoffkonzentration wird bevorzugt derart gewählt, dass bei Auftrag einer üblichen Menge des Mittels auf die Haut 1 μg bis 2 mg Wirkstoff / cm2 Haut, stärker bevorzugt 2 μg Wirkstoff bis 1 mg Wirkstoff / cm2 Haut, z. B. 10 μg bis 500 μg Wirkstoff / cm2 Haut zur Verfügung gestellt werden.
Ein besonderer Vorteil der erfindungsgemäßen Formulierungen ist es, daß sie besonders schnell den gesunden Zustand der Haut wieder herstellen können. Insbesondere vorteilhaft sind Bodylotionen, die nach dem Waschen der Haut und dem damit verbundenen Zerstören der Hautbarriere durch Herauslösen von Barrierelipiden aufgetragen werden. Auch dieses Problem ist beispielsweise in der WO 98/32444 ausdrücklich beschrieben.
Erfindungsgemäß können die Wirkstoffe der Formel I als solche oder auch in liposomaler Form angewendet werden. Liposome werden vorteilhafterweise mit Lecithinen ohne oder mit Zusatz von Sterolen oder Phytosterolen gebildet. Die Enkapsulierung der Wirkstoffe der Formel I kann alleine oder zusammen mit anderen Wirkstoffen erfolgen.
Die erfindungsgemäßen Mittel sind insbesondere für die Behandlung des Menschen geeignet. Sie können aber auch für die Behandlung von Tieren eingesetzt werden.
Die folgenden Beispiele erläutern die Erfindung.
Beispiel 1
Herstellung von Phytansäureethylester
3,7,11 , 15-Tetramethylhexadecansäure-ethylester:
3,7,1 1 ,15-Tetramethylhexadecansäure (28,9 g, 90.0 mmol) wird in getrocknetem CHCI3 (100 ml_) gelöst, Ethanol wird im Überschuß (157,5 mmol) zusammen mit konz. H2SO4 (450 mg) zugegeben, und die Lösung wird an einer Soxhlet Apparatur mit Molekularsieb (4 Ä) vier Tage unter Rückfluß gekocht. Das Reaktionsgemisch wird dann in einem Scheidetrichter mit wäßriger 10% Natriumhydrogencarbonat-Lösung (2 x 100 mL) ausgewaschen. Die kombinierten wäßrigen Phasen werden noch einmal mit Ethylacetat (2 x 100 mL) extrahiert. Die vereinigten organischen Phasen werden dann über Natriumsulfat getrocknet, und das Lösungsmittel wird unter vermindertem Druck abdestilliert. Der Rückstand wird bei 14O0C destilliert, 3,0 x 10'1 mbar. Ausbeute: 28,6 g reine Substanz, 93% Ausbeute. - Rf (n-Hexan/Ethylacetat 9:1 ) = 0,78; 1H-NMR (400 MHz, CDCI3): δ = 4,12 (q, J = 7,2, 2H), 2,33-2,27 (m, 1 H), 2,15-2,06 (m, 1 H), 2,02-1 ,89 (m, 1 H), 1 ,59-1 ,46 (m, 1 H), 1 ,44-1 ,02 (m, 24H), 0,98-0,80 (m, 14H); MS (El): 340 (12) [M+], 115 (100) [C8H11O2 +]; IR (Film) cm"1: v = 2925, 1737, 1462, 1376, 1165, 1033, 930, 847.
Beispiel 2
Herstellung von Phvtansäure-n-butylester
3,7,11,15-Tetramethy Ihexadecansäure-n-buty lester:
3,7,11 ,15-Tetramethylhexadecansäure (10,0 g, 32,0 mmol) wird in n-Butanol im Überschuß (160,0 mmol, 11 ,9 g) gelöst. Dann wird konz. Schwefelsäure (345 mg) zugegeben, und die Lösung wird an einer Soxhlet Apparatur mit Molekularsieb (4 Ä) vier Tage unter Rückfluß gekocht. Das Reaktionsgemisch wird dann in einem Scheidetrichter mit wäßriger 10% Natriumhydrogencarbonat-Lösung (2 x 100 mL) ausgewaschen. Die kombinierten wäßrigen Phasen werden noch einmal mit Ethylacetat (2 x 100 mL) extrahiert. Die vereinigten organischen Phasen werden dann über Natriumsulfat getrocknet, und das Lösungsmittel wird unter vermindertem Druck abdestilliert. Der Rückstand wird bei 142°C destilliert, 4,4 x 10'1 mbar. Ausbeute: 9,8 g reine Substanz, 83% Ausbeute. - Rf (n-Hexan/Ethylacetat 49:1 )
= 0,48; 1H-NMR (400 MHz, CDCI3): δ = 4,07 (t, J = 6,8 Hz, 2H), 2,32-2,26 (m, 1H), 2,18- 2,06 (m, 1H), 1,99-1,90 (m, 1H), 1,66-1,57 (m, 2H), 1,55-1,46 (m, 1H), 1,43-1,01 (m, 22H), 0,97-0,90 (m, 6H), 0,88-0,82 (m, 12H); MS (El): 368 (20) [M+], 143 (100) [C8H15O2 +]: IR (Film) cm"1: v = 2925, 2869, 1736, 1462, 1378, 1166, 1022.
In den folgenden Formulierungsbeispielen beziehen sich Gew.-%-Angaben auf das Gesamtgewicht der Zusammensetzung.
Formulierungsbeispiel 1
Anti-Cellulite-Creme mit Coffein
Herstellvorschrift
Teil A und B separat auf 800C erwärmen. Unter kräftigem Rühren mit dem Ultraturax bei 13000 rpm Teil A langsam zu Teil B geben und für zwei Minuten homogenisieren. Die Emulsion auf 450C abkühlen lassen und die Zutaten des Teiles C unter langsamem Rühren zusetzen. Danach mit dem Teil D den pH-Wert auf 6.0 einstellen.
Formulierungsbeispiel 2
Anti-Cellulite-Creme
Herstellvorschrift
Teil A und B separat auf 8O0C erwärmen. Unter kräftigem Rühren mit dem Ultraturax bei 13000 rpm Teil A langsam zu Teil B geben und für zwei Minuten homogenisieren. Die Emulsion auf 450C abkühlen lassen und die Zutaten des Teiles C unter langsamem Rühren zusetzen. Danach mit dem Teil D den pH-Wert auf 6.0 einstellen.
Formulierungsbeispiel 3
Anti-Cellulite Creme
Herstellvorschrift
Teil A und B separat auf 800C erwärmen. Unter kräftigem Rühren mit dem Ultraturax bei 13000 rpm Teil A langsam zu Teil B geben und für zwei Minuten homogenisieren. Die Emulsion auf 45°C abkühlen lassen und die Zutaten des Teiles C unter langsamem Rühren zusetzen. Danach mit dem Teil D den pH-Wert auf 7.5 einstellen.
Formulierungsbeispiel 4
Slimming Lotion mit Phytansäure-n-butylester
Herstellvorschrift
Teil A auf 85 0C unter Rühren erwärmen. Wenn alles gelöst ist wird Teil B zugegeben. Dann langsam Teil C, auf 80°C erwärmt, unter kräftigem Rühren mit dem Ultraturax bei 13000 rpm einbringen. Teil D langsam ebenfalls zusetzen. Für eine Minute homogenisieren. Die Emulsion auf 4O0C abkühlen lassen und die Zutaten des Teiles E unter langsamem Rühren zusetzen. Danach mit der Kaliumhydroxidlösung den pH-Wert auf 6.0 einstellen.
Formulierungsbeispiele 5 und 6 sind Gesichtscremes mit Anti-Faltenwirkung, Formulierungsbeispiele 7 und 8 sind Cremes für empfindliche Haut, Formulierungsbeispiele 9 und 10 stellen eine Hautschutz-Bodylotion dar, und Formulierungsbeispiele 11 und 12 ein Augen-Contour-Gel.
Die Wirksamkeit der erfindungsgemäßen Formulierungen kann überprüft werden, indem Testpersonen, die an Cellulite und/oder Unterhaut-Fettpolstern leiden, eine geeignete Menge beispielsweise der Formulierung 1 oder 2 topisch verabreicht wird. Die Verabreichung erfolgt beispielsweise in einer Menge von 20 mg der Formulierung aus Formulierungsbeispiel 2 pro 10 cm2 Haut, beispielsweise dreimal täglich. Nach geeigneter Behandlungsdauer, beispielsweise nach 2 Monaten, stellt sich bei den Versuchspersonen eine deutlich sichtbare Verbesserung der Cellulite ein.
Claims
1. Mittel zur topischen Verabreichung einer Verbindung der Formel
B O (CH3)2CH-(CH2)rCH(CH3)-(CH2)3-CH(CH3)-(CH2)rC(CH3)-C-C-R
A B (I).
in der R Wasserstoff, OR1, N(OH)R1 oder NR2R3 darstellt,
R1, R2 und R3 unabhängig voneinander Wasserstoff, CrC22-Alkyl, CrC22-Alkenyl, C7-Ci2- Arylalkyl (insbesondere Benzyl, Phenethyl und Phenylpropyl), Retinyl, Tocopheryl, Ascorbyl oder einen von einer Aminosäure oder Peptid herstammenden Rest darstellt und A und B Wasserstoffatome darstellen oder A und ein Rest B eine Doppelbildung bilden und der andere Rest B ein Wasserstoffatom darstellt oder der Rest A ein Wasserstoffatom darstellt und die Reste B zusammen ein Sauerstoffatom bilden oder einer der Reste B eine Hydroxylgruppe darstellt und der andere Rest B und der Rest A Wasserstoffatome darstellen, enthaltend eine Verbindung der Formel (I) und einen pharmazeutisch und/oder kosmetisch verträglichen Träger, mit der Maßgabe, daß das Mittel kein Retinoid enthält.
2. Mittel nach Anspruch 1 , dadurch gekennzeichnet, daß es sich bei der Verbindung der Formel I um eine Verbindung der Formel Il
handelt, in der R Wasserstoff, OR1 oder NHR1, N(OH)R1,
R1 Wasserstoff, C1-C22-Alkyl, CrC22-Alkenyl, Benzyl, Phenetyl, Phenpropyl, Retinyl, Tocopheryl, Ascorbyl oder einen von einer Aminosäure oder einem Peptid herstammenden Rest darstellt und A und B entweder beide Wasserstoffatome darstellen oder zusammen eine Doppelbindung bilden.
3. Mittel nach Anspruch 2 dadurch gekennzeichnet, daß R ein Wasserstoffatom oder einen Rest OR1 darstellt, R1 ein Wasserstoffatom oder einen CrC8-Alkylrest darstellt und A und B beide Wasserstoffatome darstellen.
4. Mittel nach Anspruch 1 dadurch gekennzeichnet, daß es sich um Phytansäure handelt.
5. Mittel nach einem der Ansprüche 1 bis 4 dadurch gekennzeichnet, daß es sich bei dem Mittel um ein kosmetisches Mittel und bei dem Träger um einen kosmetisch verträglichen Träger handelt.
6. Mittel nach einem der Ansprüche 1 bis 5 dadurch gekennzeichnet, daß das Mittel einen weiteren Wirkstoff enthält, ausgewählt aus Coffein, Flavonen und Isoflavonen.
7. Mittel nach einem der Ansprüche 1 bis 6 dadurch gekennzeichnet, daß es sich um ein Haarpflegemittel handelt.
8. Mittel nach Anspruch 7 dadurch gekennzeichnet, daß es sich um ein Shampoo oder eine Haarkur handelt.
9. Mittel nach einem der Ansprüche 1 bis 4 dadurch gekennzeichnet, daß es sich bei dem Mittel um ein Arzneimittel und bei dem Träger um einen pharmazeutisch verträglichen Träger handelt.
10. Verwendung einer Verbindung der Formel (I) wie in einem der Ansprüche 1 bis 4 definiert zur Herstellung eines topisch zu verabreichenden Arzneimittels oder kosmetischen Mittels zur Vorbeugung und/oder Behandlung von Cellulite, Unterhaut-Fettpolstern, Hautalterung, Zuständen, die durch eine geschädigte oder verletzte Hautbarriere hervorgerufen werden, zur Behandlung von Haar und Kopfhaut, zur Behandlung und aktiven Prävention trockener Haut und zur Stärkung der Barrierefunktion der Haut sowie zur Behandlung, Pflege und Prophylaxe von sensibler Haut und/oder zur Behandlung und Prophylaxe der Symptome einer negativen Veränderung der physiologischen Homöostase der gesunden Haut, insbesondere defizitärer, sensitiver oder hypoaktiver Hautzustände oder defizitärer, sensitiver oder hypoaktiver Zustände von Hautanhangsgebilden, entzündlicher Hautzustände sowie des atopischen Ekzems, der polymorphen Lichtdermatose, Psoriasis, Vitiligo, empfindlicher, juckender oder gereizter Haut, Veränderungen der normalen Lipidperoxidation, einer Veränderung des Ceramid-, Lipid- und Energiestoffwechsels der gesunden Haut, einer Veränderung des physiologischen transepidermalen Wasserverlustes, einer Verminderung der Hauthydratation und Abnahme des Feuchtigkeitsgehaltes der Haut, Veränderung des Natural Moisturizing Factor-Gehalts, Verminderung der Zell-Zell-Kommunikation, Mangelerscheinungen der intrazellulären DNS- Synthese, DNS-Schädigungen und Verminderung von endogenen DNS- Reparaturmechanismen, Aktivierung von Metalloproteinasen und/oder anderer Proteasen bzw. Inhibierung der entsprechenden endogenen DNS-Reparaturmechanismen und Abweichungen von den normalen post-translationalen Modifikationen von Bindegewebsbestandteilen.
11. Verwendung nach Anspruch 10, wobei es sich bei dem Arzneimittel oder kosmetischen Mittel um ein Mittel zur Behandlung oder Vorbeugung von Cellulite und/oder Unterhaut-Fettpolster handelt.
12. Verwendung nach Anspruch 10, wobei es sich bei dem Arzneimittel oder kosmetischen Mittel um ein Mittel zur Behandlung oder Vorbeugung von fettigen Haaren und/oder Schuppenbildung handelt.
13. Verwendung einer Verbindung der Formel (I) wie in einem der Ansprüche 1 bis 4 definiert zur Behandlung und Vorbeugung von Cellulite, Unterhaut-Fettpolstern, Hautalterung, Zuständen, die durch eine geschädigte oder verletzte Hautbarriere hervorgerufen werden, zur Behandlung von Haar und Kopfhaut zur Behandlung und aktiven Prävention trockener Haut und zur Stärkung der Barrierefunktion der Haut sowie zur Behandlung, Pflege und Prophylaxe von sensibler Haut und/oder zur Behandlung und Prophylaxe der Symptome einer negativen Veränderung der physiologischen Homöostase der gesunden Haut, insbesondere defizitärer, sensitiver oder hypoaktiver Hautzustände oder defizitärer, sensitiver oder hypoaktiver Zustände von Hautanhangsgebilden, entzündlicher Hautzustände sowie des atopischen Ekzems, der polymorphen Lichtdermatose, Psoriasis, Vitiligo, empfindlicher, juckender oder gereizter Haut, Veränderungen der normalen Lipidperoxidation, einer Veränderung des Ceramid-, Lipid- und Energiestoffwechsels der gesunden Haut, einer Veränderung des physiologischen transepidermalen Wasserverlustes, einer Verminderung der Hauthydratation und Abnahme des Feuchtigkeitsgehaltes der Haut, Veränderung des Natural Moisturizing Factor-Gehalts, Verminderung der Zell-Zell-Kommunikation, Mangelerscheinungen der intrazellulären DNS- Synthese, DNS-Schädigungen und Verminderung von endogenen DNS- Reparaturmechanismen, Aktivierung von Metalloproteinasen und/oder anderer Proteasen bzw. Inhibierung der entsprechenden endogenen DNS-Reparaturmechanismen und Abweichungen von den normalen post-translationalen Modifikationen von Bindegewebsbestandteilen, wobei es sich bei den Behandlungen um kosmetische Behandlungen handelt.
14. Verwendung nach Anspruch 13 zur Behandlung und Vorbeugung von Cellulite und/oder Unterhaut-Fettpolster.
15. Verwendung nach Anspruch 13 zur Behandlung und Vorbeugung von fettigen Haaren und/oder Schuppenbildung.
16. Verbindung der Formel (I) wie in Anspruch 1 definiert, in der der Rest R OR1 darstellt und R1 ein n-Propyl oder ein C4-C22-Alkylrest ist.
