Definitions
- the present invention relates to a miniaturized device intended for the transport and analysis of liquid micro-samples, in particular by optical or electrochemical methods.
- Such devices are particularly useful tools in the field of chemistry and biochemistry, as evidenced, for example, by the following publications: "Polymer microfabrication methods for microfluidic analytical applications” (Holger Becker et Al. - Electrophoresis 2000 , 21, 12-26) and “Fabrication of microfluidic Systems in poly (dimethylsiloxane)” (J. Cooper McDonald - Electrophoresis 2000, 21, 27-40).
- An upper plate provided with at least one orifice arranged so as to open into the channel.
- the bonding of the two plates uses polymerizable adhesives which must be. deposited in a sufficiently thick and uniform layer to ensure good sealing but sufficiently thin to avoid invasion of the canal. It is therefore a technology that is delicate to implement and therefore expensive.
- Document US Pat. No. 4,756,884 proposes a device of the same type in which one of the plates is provided, along the edges of the channel, with a triangular section edge forming an integral part of the plate and constituting a director. of energy which allows it to be linked to one another by ultrasonic welding. Experience has, however, shown that this welding technique is complex and that it does not guarantee the watertightness of the channel throughout its course.
- the object of the present invention is to provide a sample holder of the above type, but ensuring, for a reasonable cost price, perfect sealing of its channel.
- the invention relates to a miniaturized device intended for the transport and analysis of a liquid sample, of the type comprising: - a lower plate whose upper face is pierced with a channel intended to receive the liquid,
- the clamping and holding members are rivets which can be formed by pins formed on one face of one of the plates and taking place in holes made in the other plate.
- these holes end in an enlarged portion in which the end of the pins is crushed.
- the clamping and holding members are either screws and nuts, or self-tapping screws.
- the channel is bordered on each side by a rim embedded in the upper plate under the action of the clamping and holding members; - The channel comprises a bowl forming a reservoir, at the point where the filling orifice opens;
- This orifice has the shape of a funnel
- FIG. 1a and 1b show the lower plate of the device, respectively seen from above and in section along A-A;
- FIGS. 2a and 2b show the upper plate of the device, viewed respectively from above and in section along B-B;
- FIG. 3 and 4 are sectional views along A-A of the assembled device, respectively before and after the joining of the two plates.
- FIGS. 1a and 1b show a rigid rectangular plastic plate 10, called the bottom plate or bottom, which typically has a length of 50 mm, a width of 30 mm and a thickness of 2 mm.
- the upper face 12 of this plate is hollowed out with a channel 14 with a substantially square section, 0.08 mm side, and in the shape of a cross. Its arm 16, longer than the other three, is folded into three parallel portions. The ends of the four arms of the cross end in a bowl 18 having, typically, a diameter of 1.95 mm.
- the channel 14 can have any shape.
- the channel 14 and the four bowls 18 are bordered, on each side, by a rim 20, visible only in FIG. 1b, having initially, in the example described, a substantially rectangular section of 0.1 mm thick and 0.2 mm wide.
- the bottom 10 is pierced, all around the channel 14 and the bowls 18, with a plurality of circular holes 22 whose portion 24, opening on its underside 26, is widened.
- the holes 22, ten in number have a 2.10 mm diameter while the enlarged portions 24 have a diameter of 3 mm and a depth of 2 mm.
- FIGS. 2a and 2b show a second rectangular rigid plate 28, called the upper plate or cover, which is intended to take place on the lower plate 10, of the same dimensions as it and, like it, made of plastic.
- the underside 30 of this plate is provided with a plurality of pins 32 arranged so that, when the two plates are superimposed, they take place in the holes 22.
- these pins have a slightly frustoconical profile and an initial length greater than the thickness of the bottom 10.
- the nipples are ten in number. Typically, their length is 2.30 mm, their diameter at the base of 2 mm and their diameter at the top of 1.80 mm.
- the upper plate 28 is, moreover, pierced with four orifices 34 in the form of a funnel, the flared part of which is on the side of its upper face 36. These orifices are positioned so as to open exactly into the bowls 18 of the lower plate 10.
- the lower plate 10 with the channel 14, the cups 18, the flange 20 and the holes 22, as well as the upper plate 28, with the pins 32 and the orifices 34, are produced by injection, according to techniques known to those skilled in the art.
- the plastics used for the bottom 10 and the cover 28 can be polystyrene, polycarbonate or polymetacrylate.
- the device is intended for analysis by optical methods, it goes without saying that these materials must be transparent.
- FIG. 3 shows the cover 28 placed on the bottom 10, the lower face of the cover being in contact with the upper face 12 of the bottom and the pins 32 engaged in the holes 22. It is noted that the end of the pins 32 exceeds the face bottom 26 of bottom 10.
- the assembly is placed inverted (cover 28 at the bottom) on a work surface and a strong pressure is exerted on the bottom 10.
- the ends of the nipples 32 are then hot-crushed, using any suitable equipment known to those skilled in the art, so as to fill the enlarged portions 24 of the holes 22.
- rivets are formed which, after cooling, keep the two plates firmly joined to one another, while ensuring the watertight closure of the channel 14 and of the bowls 18.
- the rim 20 of the bottom 10 is embedded in the cover 28 and strengthens the tightness of the assembly.
- the device thus produced can then receive its liquid sample which is injected into the channel 14 through the orifice 34 corresponding to the bowl 18 of the arm 16, this bowl acting as a reservoir.
- the other orifices 34 serve for the evacuation of air during filling.
- the rivets formed by the pins 32 after crushing their ends can be replaced either by screws and nuts or by self-tapping screws, advantageously made of brass, the ends of which are taken care of to drown in the plates so avoid any excess thickness.
- a miniaturized sample holder with a particularly simple structure, guaranteeing excellent sealing of its channel and the manufacture of which does not involve costly processes (lithography, for example). Such a device therefore has a very low cost price, without affecting its quality.
