Bereitgestellt wird eine Gangliosidmischung, die Ganglioside der folgenden allgemeinen Formel: (Zucker)-OCH2-CH(-NH-CO-R<1>)-CH(OH)-CH=CH-R<2>, worin (Zucker) für einen Zuckerrest steht, die Gruppe -CO-R<1> eine Acyl-Fettsäure darstellt, die in Form eines Amides an den Rest des Moleküls gebunden ist, R<l> einen geraden, gesättigten Alkylrest mit mindestens 10 C-Atomen bedeutet und R<2> einen geraden, gesättigten Alkylrest mit mindestens 10 C-Atomen oder einen geraden Alkenylrest mit mindestens 10 C-Atomen und einer, zwei oder drei Doppelbindung(en) bedeutet, enthält oder daraus besteht. Diese Mischung zeichnet sich dadurch aus, dass von den vorhandenen Gangliosiden der allgemeinen Formel I mindestens 10 Gew.-% der Gruppe -CO-R<1> und somit der Acyl-Fettsäuren eine C20:0 Fettsäure sind. Diese Mischungen verfügen über eine verbesserte biologische Aktivität.

Titel: Ganglioside mit modifizierter Acylfunktion
BESCHREIBUNG
Die Erfindung betrifft eine Gangliosidmischung, die Verwendung dieser Gangliosidmischung zur Herstellung eines diätetischen, pharmazeutischen und Nahrungs-Mittels sowie ein entsprechendes Mittel.
Ganglioside sind kompliziert aufgebaute Glycolipide, die vorwiegend auf der äußeren Oberfläche der Plasmamembran einer Zelle lokalisiert sind. Sie enthalten einen polaren, aus Zuckern sowie Zuckerderivaten und ggf. N-Acetylneuraminsäure aufgebauten Teil (Kopf) sowie mehrere unpolare, aus Fettsäuren und deren Derivate aufgebaute aliphatische Ketten.
Die Struktur der Oligosaccharidkette der Ganglioside bestimmt direkt die Zeil-Zeil Interaktion (Zelladhäsionen), Rezeptorabdeckung (z.B. IL-2, Selektine), vergleiche dazu Robb, R.J., J. Die Fettsäurezusammensetzung ist hingegen verantwortlich für die räumliche Anordnung der Moleküle in einer Membran (Microdomainbildung/ Clustering) und ist mitbestimmend für den substratspezifischen Umsatz der Glycolipide im Stoffwechsel (Enzym- Substratspezifität). Wenn in der Literatur im Zusammenhang mit Gangliosiden von deren Fettsäurezusammensetzung die Rede ist, wird häufig nicht zwischen den beiden unpolaren aliphatischen Ketten unterschieden, die beide als Fettsäuren bezeichnet werden. Diese Fettsäuren sind dabei ein struktur- und funktionsbestimmendes Element, vergleiche dazu Ladisch, S., Hasegawa, A:, Li, R., Kiso, M. Biochemistry 1999, 34, 1197-1202. Der N- Acylsphingosin Anteil von Gangliosiden (Ceramid) ist als intrazelluläres Signal vermittelndes Molekül bedeutend. Die langkettigen Acylgruppen von Gangliosiden sind in biologischen Systemen allgegenwärtige Fettsäuren und verleihen diesen Gangliosiden in ihren vielfältigen Funktionen eine hohe Bioaktivität.
Ganglioside werden enzymatisch (lysosomale Enzyme) intrazellulär zu Ceramid, dieses zu Sphingomyelin und weiter zu Sphingosin, Sphingosin- 1 P umgesetzt. Ceramid beeinflusst über den Sphingomyelin-Umsatz auch die Synthese von Diacylglycerol und Phosphoryldiacylglycerol. Diese Stoffe sind alle hochpotente signalvermittelnde Moleküle. Ganglioside und zu Ceramid umgesetzte Ganglioside bestimmen so in vielfältiger Weise den Stoffwechsel und die Zellfunktionen. Während die 3-keto gebundene Fettsäure des Gangliosides alle Stoffwechselprodukte des Ceramides z.B. Sphingosin strukturell und funktionell mitbestimmt d.h. diese aliphatische Kette bleibt im Stoffwechsel unberührt, ist die N- acylierte (Amid)= Fettsäure nur als Fettsäurekomposition des Ceramids selbst und entsprechenden Glycosylderivaten enthalten und wird im Stoffwechsel der sekundären Botenstoffe abgespalten.
Die spezifische Acylierung des Moleküls bestimmt die Potenz von Gangliosiden/ Ceramiden und deren Stoffwechselprodukten als signalvermittelnde Moleküle. Der Stoffwechsel von Gangliosid-Derivaten durch substratspezifische Enzyme (Sphingomyelinase, Ceramidase, Sphingosinacyltransferase) und die enzymatische Regulation der Ceramidaktivität als Signalmolekül (Ceramidehydrogenase,
Ceramidkinase), ist durch das Acyl-Muster als Strukturelement mitbestimmt. Zudem bedingen gesättigte Fettsäuren mit einer Kohlenstofflänge bis C 24 einen schnellen Gangliosid-Stoffwechsel (de novo Synthese/ Umsatz). Die spezifische Acylierung des Moleküls ist auch relevant für den Einfluss von Gangliosiden auf verschiedene intrazellulär, regulative Enzyme (Phosphokinase C, Phospholipase D und verschiedene Proteinphosphatasen/ -kinasen).
Bisher bekannte biologische Funktionen von Gangliosiden sind:
• Entwicklung des Intestinaltraktes des Neugeborenen; intestinale Immunsystem-Entwicklung, vergleiche dazu Rueda, R:, Maldonado, J., Narbona, E:, Gil, A. Early human Development 1998, 53, S135- S147; Jensen, R. Lipids 1999, 34, 1243-1271.
• Anticarzinogene Wirkungen (induziert Apoptose, stoppt das Zellwachstum), vergleiche dazu Vesper, HL, Schmelz, E.-M., Nikolova-Karakashian, M.N., Dillehay, D.L., Lynch, D.V., Merril, A.H.Jr. J. Nutr.1999, 129, 1239-1250; Farooqui, A., Harrocks, L.A., Farooqui, T., Chemistry and Physics 2000, 106, 1-29.
• T-Zellen Differenzierung wird induziert (Induktion der Zellreifung); Inhibition der T-Zell Antwort, vergleiche dazu Yanagihava, K., Kato, E:, Hitomi, S., Sunamoto, J., Wada, H. Glycoconjugate J. 1999, 16, 59-65.

Die umfangreich in der Literatur beschriebenen biologischen Funktionen der Ganglioside werden vor allem mit der Zusammensetzung der Oligosaccharidkette in Verbindung gebracht. Dagegen wird dem Lipϊd- Anteil weniger funktionelle Bedeutung beigemessen, obwohl bekannt ist, dass die Bioaktivität der Ganglioside durch die
Fettsäurezusammensetzung mitbestimmt wird. Die meisten der bisher existierenden Patente befassen sich daher vorwiegend mit der Oligosaccharidkette oder geben keine Spezifikation der Fettsäuren an, man vergleiche JP 92105616. Andere machen nur Angaben für das Verhältnis der gesättigten zu ungesättigten Fettsäuren, man vergleiche JP 3101691.
Im US-Patent 5366963 22/94 wird der Zusammenhang zwischen dem Anteil der Fettsäuren und einer Kettenlänge <C18 und >C20 und der immunsuppressiven Wirkung einer Gangliosidmischung beschrieben. Dabei sind die Fettsäuren C18 und C20 ausdrücklich ausgenommen.
Es wurde nun überraschend gefunden, dass die Lipidkomponente bei den Gangliosiden eine wesentlich höhere Bedeutung hat, als dies bisher angenommen wurde.
Aufgabe der vorliegenden Erfindung ist es daher, eine Gangliosidmischung mit einer verbesserten biologischen Aktivität bereitzustellen.
Gelöst wird diese Aufgabe durch die Lehre der Ansprüche.
Die Ganglioside entsprechen dabei der folgenden allgemeinen Formel I:
(Zucker)-OCH2-CH(-NH-CO-R1)-CH(OH)-CH=CH-R2 (I)
Diese allgemeine Formel I beschreibt formelmäßig diejenigen Ganglioside, welche die Basis für die erfindungsgemäße Mischung darstellen. Es geht hier nicht um die Bereitstellung von derartigen Gangliosiden als solchen, denn die erfindungsgemäße Mischung kann sogar insgesamt aus bekannten Gangliosiden bestehen. Vielmehr besteht die Erfindung darin, dass von den Gangliosiden, welche unter die oben angegebene Formel und die im Rahmen der hier vorliegenden Unterlagen definierten Reste in dieser allgemeinen Formel fallen, die Gruppe -CO-R1 aus bestimmten Fettsäuren in bestimmten Mengen aufgebaut ist. Die allgemeine Formel I legt somit die rechnerische und stoffliche Bezugsbasis fest, auf die sich die hier gemachten stofflichen und mengenmäßigen Angaben beziehen. Es ist beispielsweise möglich, erfindungsgemäße Mischungen ausschließlich durch ein neues Vermischen bereits bekannter Ganglioside oder Gangliosidgemischen zu erhalten. Natürlich können auch Modifizierungen etc. der Ganglioside vorgenommen werden, worauf nachstehend näher eingegangen wird.
Der Rest R2 bedeutet dabei einen geraden, gesättigten Alkylrest oder einen geraden Alkenylrest, wobei beide Reste 10 oder mehr C-Atomen, beispielsweise bis zu 30 C-Atomen, besitzen. Der Rest R2 kann somit 10, 11 , 12, 13, 14, 15, 16, 17, 18, 19, 20, 21 , 22, 23, 24, 25, 26, 27, 28, 29 und 30 C-Atome aufweisen. Vorzugsweise ist die Anzahl der C-Atome geradzahlig, beispielsweise 14, 16 und 18. Der Alkenylrest kann eine, zwei oder drei Doppelbindung(en) besitzen.
Der Rest R1 bedeutet einen geraden, gesättigten Alkylrest mit 10 oder mehr C-Atomen, beispielsweise bis zu 30 C-Atomen. Der Rest R1 kann somit 10, 11 , 12, 13, 14, 15, 16, 17, 18, 19, 20, 21 , 22, 23, 24, 25, 26, 27, 28, 29 und 30 C-Atome aufweisen. Vorzugsweise ist die Anzahl der C-Atome geradzahlig, beispielsweise 18, 20, 22 und 24. Der Rest R1 stellt zusammen mit der daran gebundenen Ketogruppe eine Fettsäure dar, die in Form eines Amides an den Rest des Moleküls gebunden ist. Dieser Rest -CO-R1 wird im Rahmen der vorliegenden Unterlagen als Acyl-Fettsäuren bezeichnet.
Ein wesentlicher Aspekt der vorliegenden Erfindung liegt darin, dass bei der erfindungsgemäßen Gangliosidmischung die C20:0 Fettsäure mindestens 10 Gew.-% der Acylfettsäuren (und somit der Gruppe -CO-R1 ; siehe oben) ausmacht. Diese Angabe betrifft somit nur eine der unpolaren aliphatischen Ketten der Ganglioside. Über die andere unpolare aliphatische Kette und die Zuckereinheit ist damit keine Aussage gemacht. Diese andere aliphatische Kette kann bekannter und/oder beliebiger Natur sein. Die Angabe C20:0 bezeichnet nach der üblichen Nomenklatur eine Acylgruppe mit 20 C-Atomen (das Kohlenstoffatom der CO-Gruppe wird dabei mitgezählt) ohne Doppelbindung (:0).
Nach einer bevorzugten Ausführungsform macht die C20:0 Fettsäure 10 bis 15 Gew.-% aus. Mit dieser Bereichsangabe sind alle Zwischenwerte, alle engeren Bereiche und insbesondere alle ganzzahligen Zwischenwerte umfasst und offenbart, z. B. 1 1 , 12, 13 und 14 Gew.-%.
Nach einer bevorzugten Ausführungsform beträgt das Gew. -Verhältnis der C18:0 Acyl-Fettsäure in der Gruppe -CO-R1 zu der C20:0 Acryl- Fettsäure in der Gruppe -CO-R1 1 ,0 bis 3,0. Auch diese Bereichsangabe umfasst alle dazwischenliegenden Werte, alle engeren Bereiche und insbesondere Einzelwerte, beispielsweise 1 ,1 , 1 ,2, 1 ,3, 1 ,4, 1 ,5, 1 ,6, 1 ,7, 1 ,8, 1 ,9, 2,0, 2,1 , 2,2, 2,3, 2,4, 2,5, 2,6, 2,7, 2,8 und 2,9.
Die erfindungsgemäße Gangliosidmischung kann auf Basis tierischer (der hier verwendete Begriff 'Tier' oder 'tierisch' bezeichnet nicht den Menschen und auch keine Quellen humanen Ursprungs) und/oder pflanzlicher, einschließlich von Pilzen und Einzellern gewonnener Lipidextrakte und/ oder synthetisch hergestellter Ganglioside erhalten werden. Um die erfindungsgemäße Zusammensetzung der Acyl- Fettsäuren zu erreichen, können die Ganglioside aus den oben gegebenen Lipidextrakten modifiziert werden. Zudem ist es möglich, derartige Lipidextrakte mit Isolaten von Gangliosiden mit angereicherter Fettsäurespezifikation zu versetzen. Auch ist es möglich, derartige Lipidextrakte mit chemisch synthetisierten Gangliosiden zu versetzen. Ferner können die Ganglioside, welche die Mischung ausmachen, vollständig synthetisch hergestellt sein. Auch ist es möglich, verschiedene Arten der Modifizierungen und/ oder Herstellungen miteinander zu kombinieren. Vorzugsweise wird jedoch von nativen Gangliosiden ausgegangen. Diese können dann modifiziert werden. Vorzugsweise sind die erfindungsgemäßen Mischungen somit aufgebaut aus nativen unmodifizierten Gangliosiden, die mit Gangliosiden vermischt wurden, die ausgehend von nativen Gangliosiden erhalten und dann modifiziert wurden. Einige der Möglichkeiten der Modifizierung sind nachfolgend aufgeführt:
• Acylkettenspezifikation von nativen/ isolierten Gangliosiden/ Ceramiden durch enzymatische Abspaltung des N-Acylrestes (Sphingolipid/ Ceramid
N-Deacylase EC 351 -) und neue spezifische Fettsäureveresterung durch sauer/ alkalisch katalysierte chemische Veresterung oder spezifischer Fettsäure Derivatisierung nach Anand, J.K., Sadozai, K.K., Hakomori, S. Lipids 1996, 31 , 995-998 mittels freien Fettsäuren.
• Interesterifikation von Gangliosiden durch die Ceramid N-Deacylase EC351 mit spezifischen freien Fettsäuren, Methyl-, Ethylestern oder Triacylglyceriden bei optimiertem pH-Wert und Temperatur.
• Acylkettenspezifikation von nativen, isolierten Gangliosiden/ Ceramiden durch Deacylierung zu Lysogangliosiden nach Gasa, S., Kamio, K., Makita, A. J. Lipid Res. 1992, 33, 1079-1084, gefolgt von einer sauer/ alkalisch katalysierten chemischen Veresterung spezifischer Fettsäuren mittels freien Fettsäuren, Methyl-, Ethylestern oder Triacylglyceriden.
Auf diese Möglichkeiten der Modifizierung wird nachstehend in den Beispielen noch näher eingegangen.
Durch die Modifizierung der Acyl-Fettsäuren im erfindungsgemäßen Sinne kann die biologische Aktivität der Ganglioside erhöht werden. Diese Erhöhung der biologischen Aktivität gilt für die unterschiedlichsten Funktionen der Ganglioside. Verfügt ein Gangliosid beispielsweise über eine bestimmte biologische Aktivität, wird durch die Veränderung bzw. die Modifizierung des Lipid-Anteils, ohne dass beispielsweise die Zuckereinheit verändert wird, die entsprechende, bereits vorhandene biologische Aktivität erhöht. Dies beruht darauf, dass durch die erfindungsgemäße Modifizierung des Lipid-Anteils der Ganglioside eine verbesserte Mobilität des Moleküls in den Membranen erreicht wird . Die hohe Mobilität und die durch die erfindungsgemäße Fettsäure- Zusammensetzung induzierte räumliche Anordnung von Gangliosiden in der Membran (Clustering, Cholesterol/ Glycolipid Mikrodomainen) führen zu einer Verstärkung Rezeptor-vermittelter Signale (beeinflusst Zelloberflächen-Rezeptoren), der Aktivität von lonenkanälen (z.B.: Na/K ATPase) und der Aktivität membranständiger Enzyme (z.B. Phosphokinase C), vergleiche dazu Zeller, C.B., Marchase, R,B., Am. J. Physiol. 1992, 262, C1341-C1355.
Durch die Modifizierung des Lipid-Anteils (genauer der Acyl-Fettsäuren) kann die bereits vorhandene biologische Aktivität von Gangliosiden verstärkt werden.
Die erfindungsgemäße Gangliosidmischung kann sowohl in reiner Form als auch in Form einer Emulsion, insbesondere einer wässrigen Emulsion Anwendung finden. Die Gangliosidmischung kann auch mit anderen Lipiden zusammen eingesetzt werden, wobei davon ausgegangen wird, dass die weiteren Lipide die biologische Aktivität der Gangliosidmischung bzw. der diese ausmachenden Ganglioside nicht beeinflusst. Dies bedeutet, dass die biologische Aktivität der Ganglioside nur durch die Fettsäuren in der Lipid-Komponente des Gangliosides, aber nicht durch die Fettsäuren, die zusätzlich Anwendung finden, beeinflusst wird.
Die erfindungsgemäße Gangliosidmischung kann jedem üblichen Nahrungsmittel und auch einem Pharmazeutikum, soweit es zweckdienlich ist, zugesetzt werden. Mit anderen Worten, Gegenstand der Erfindung sind auch Nahrungs-, diätetische und pharmazeutische Mittel, die eine erfindungsgemäße Gangliosidmischung enthalten.
Zu diesen Mitteln zählen insbesondere Fertignahrungen, Nahrungssupplemente und Formelnahrungen. Unter einer Formelnahrung wird in diesem Zusammenhang eine solche verstanden, die auf Basis von tierischen (jedoch nicht humanen) und pflanzlichen Ausgangsstoffen bzw. -Produkten formuliert wurde. Die erfindungsgemäße Gangliosidmischung kann beispielsweise als Beimengungen oder Additive folgenden Produkten zugegeben werden, wobei diese Aufzählung nicht abschließend ist: Milch und Milchprodukte, Säuglings- und Kindernahrungen, Schokoriegel, Joghurtgetränke, Aufbaunahrung, Sondennahrung, Infusionslösungen und Produkten für Schwangere.
Die erfindungsgemäße Gangliosidmischung kann auch in Form eines pharmazeutischen Mittels alleine oder zusammen mit einem oder mehreren zusätzlichen Wirkstoff(en) verabreicht werden. Diese Mittel können beispielsweise als Tablette/Kapsel formuliert sein. Für die Formulierung derartiger Pharmazeutika können übliche Adjuvantien, Träger, Hilfsmittel Verdünnungsmittel, feucht-haltende Mittel, Verdickungsmittel, Geschmacksstoffe, Süßungsmittel usw. Anwendung finden.
Die pharmazeutischen Mittel können auf jede übliche Weise ( z. B. parenteral oder enteral) an einen Patienten (d.h. Mensch und Tier) verabreicht werden. Zweckmäßigerweise handelt es sich jedoch um Mittel, die zur oralen oder lingualen Verabreichung geeignet und entsprechend der Verabreichungsart formuliert sind.
Um festzustellen, ob eine Ganglioside aufweisende Mischung oder ein Ganglioside aufweisendes Mittel der hier beschriebenen Art eine erfindungsgemäße Gangliosidmischung enthält, ist es lediglich erforderlich, die Menge der zu der allgemeinen Formel I gehörenden Ganglioside zu ermitteln und dann zu analysieren, ob die hier gegebenen Spezifikationen bezüglich der Acylgruppe -CO-R1 erfüllt sind.
Sollten auch Verbindungen oder Ganglioside vorhanden sein, die nicht unter die allgemeine Formel I fallen, was durchaus möglich ist, dann werden diese bei der Berechnung nicht berücksichtigt.
Die Nahrungsmittel, diätetischen Mittel und pharmazeutischen Mittel, welche die erfindungsgemäße Gangliosidmischung enthaltenden, können unter anderem zur Verbesserung der Entwicklung des Intestinaltraktes, des intestinalen Immunsystems und neuronalen Systems und Behandlung destruktiven Veränderungen des Intestinaltraktes, des intestinalen Immunsystems und neuronalen Systems eingesetzt werden. Als Zielgruppen für die erfindungsgemäße Gangliosidmischung können alle Altersstufen von Neugeborenen bis zu Senioren genannt werden.
Die Erfindung wird im folgenden anhand der Beispiele näher erläutert. Dabei beschreiben die Beispiele 1 bis 4 Verfahren zur Herstellung bzw. Modifizierung der erfindungsgemäßen Ganglioside bzw.
Gangliosidmischungen, während die Beispiele 5 bis 8 Anwendungsbeispiele bzw. Formulierungsbeispiele für die erfindungsgemäßen Mittel erläutern. Bei den Gangliosiden der erfindungsgemäßen Gangliosidmischung handelt es sich um bekannte Verbindungen, oder sie können nach bekannten Verfahren hergestellt werden.
Beispiel 1 :
Extraktion und Separation aus Rohstoffen:
Glycosphingolipide werden aus natürlichen Matrizes (Ei, Soja, tierische Milch, Kolostralmilch, Buttermilch, tierische Gewebe u.a.) nach Ladisch und Gillard (Ladisch S., Gillard B. A solvent partition method for morcoscale ganglioside purification. 1984 Analytical Biochemistry 146, 220-231) mit Chloroform-Methanol extrahiert und über eine Partitionierung mit Diisopropylether-1-Buthanol-wässriger NaCI Lösung aufgereinigt. Die Auftrennung der einzelnen Gangliosidspezies mit unterschiedlicher Saccharidkomposition kann über Normalphasen HPLC nach Gazzotti (Gazzotti G., Sonnino S. Ghidoni R. Normal phase high Performance liquid chromatograhpic Separation of non-derivatized ganglioside mixtures. 1985 J. Chromatogr. 348, 371-378) erfolgen. Die Separierung und Fraktionierung von Gangliosiden bezüglich der Acylkettenspezifität wird über RP-HPLC wie folgt durchgeführt: In Wasser gelöste Ganglioside werden über eine RP-8 Column mit einem Solventensystem aus Acetonitril mit 5mM Natriumphosphat-Puffer, pH7,0 aufgetrennt. Die Aufzeichnung des Chromatogramms erfolgt über UV/VIS Detektion (Wellenlänge: 195nm) nach Wagener (Wagener R. Kobbe B., Stoffel W. Quantification of gangliosides by microbore high Performance liquid chromatography, 1996 J. Lipid Res. 37, 18231829) oder über eine Light Scattering Detektion nach Caboni (Caboni M.F., Menotta S., Lercker G. Separation of phospholipids in different foods with light- scattering detector, 1996 JAOCS 73, 1561-1566). Die erhaltenen Fraktionen werden über eine Fünfpunktkallibrierung des Chromatogramms quantifiziert. Die einzelnen Acylkettenspezifitäten werden mittels automatischem Fraktionssammler akkumuliert und können entsprechend einem Mischungsverhältnis der Acylkettenspezifikation neu zusammengestellt werden.

Glycosphingohpide werden direkt in der natürlichen Matrix (Ei, Soja, tierische Milch, Kolostralmilch, Buttermilch, tierische Gewebe u.a.) oder nach Extraktion nach Ladisch und Gillard (Ladisch S., Gillard B. A solvent partition method for morcoscale ganglioside purification. 1984 Analytical Biochemistry 146, 220-231 ) einer Transesterifikation mit spezifischen freien Fettsäuren unterzogen. Die Transresterifikation des Substrates erfolgt mittels einer Substrat-spezifischen Ceramide-N-Deacylase E.C. 3.5.1. - (Sigma-Aldrich, Inc., USA) mit spezifischen freien Fettsäuren bei optimalen pH-Werten in 0.1 % Triton X100 (N-Acylketten Hydrolyse bei pH-Wert 5.0-6.0; Kondensation des Lysospingolipids mit freien Fettsäuren bei pH-Wert 7.0) (Ito M,., Kurita T., Kita K. A novel enzyme that cleaves the N-acyl linkage of ceramides in various glycosphingolipids as well as sphingomyeline to produce their lysoforms. 1995 J. Biol. Chem. 270, 24370-24374; Kita K., Okino N., Ito M. Reverse hydrolysis reaction of a recombinant alkaline ceramidase of Pseudomonas aeruginosa. 2000 Biochim. Biophys. Acta 1485 (2-3), 111-120). Die modifizierten Glycosphingohpide können entweder direkt mit einem Mischungsverhältnis der Acylkettenspezifikation verwendet werden oder entsprechend einem Mischungsverhältnis der Acylkettenspezifikation mit anderen Komponenten neu zusammengestellt werden.
Beispiel 3:
Chemische Fettsäuremodifikation:
Glycosphingohpide werden aus natürlichen Matrizes (Ei, Soja, tierische Milch, Kolostralmilch, Buttermilch, tierische Gewebe u.a.) nach Ladisch und Gillard (Ladisch S., Gillard B. A solvent partition method for morcoscale ganglioside purification. 1984 Analytical Biochemistry 146, 220-231) extrahiert und aufgereinigt (siehe oben). Um die Acylkettenspezifikation von isolierten Gangliosiden zu verändern wird das Extrakt nach Gasa, S (Gasa S., Kamio K., Makita A. Improved preparation method for Isogangliosides. 1992 J. Lipid Res. 33, 1079- 1084) durch N-Trifluoroacetylierung des Sphingosines, gefolgt von einer N-Acetylierung und anschließender milden Verseifung, in korrespondierende dideacylierte Ganglioside (Schutzgruppe) umgesetzt. Diese werden chemisch zu Lysogangliosiden deacyliert. Die Abspaltung der Acylketten von nativen Gangliosiden kann auch über direkte chemische Hydrolyse nach Gaver und Sweeley (Gaver R.C. Sweeley C.C., Methods for methanolysis of sphingolipids and direct determination of long chain bases by gas chromatography. 1995 J. Lipid Res. 24, 1389- 1397) oder über eine saure Hydrolyse nach Andu (Kadowaki, H., Bremer, E:G:, Evans, J.E., Jungalwala, F.B., McCIuer, RH. Acetonitrile- hydrochloric acid hydrolisis of gangliosides for high Performance liquid Chromatographie analysis of their long chain bases. J. Lipid Res. 1983, 24, 1389-1397) mittels Inkubation des Substrates mit 0.5M HCI in Acetonitril (oder Methanol) und 4M H2O bei 75 °C, 2 Stunden erfolgen. Die resultierenden Lysoganglioside werden durch Anionenaustauscher- Chromatographie mittels Aminopropylphase von den freien Fettsäuren, Salzen und niedermolekularen Kontaminanten getrennt. Die neue Acylkettenspezifität der Lysoganglioside mit spezifischen, freien Fettsäuren, Methyl- oder Ethylestern wird durch sauer/ alkalisch katalysierte chemische Veresterung dargestellt. Es ist ebenfalls möglich, die resultierenden Glycosphingohpide mit spezifischen Fettsäuren chemisch nach Anand (Anand J.K., Sadozai K.K., Hakomori S.A simple method for the synthesis of ceramides and radiolabeied analogues. 1996 Lipids 31 , 995-998) spezifisch zu aeylieren. Unter Verwendung von Diethylphosphorylcyanid in Kombination von Triethylamin werden die Sphingolipid-Amine mit spezifischen Fettsäuren gekoppelt. Es ist so möglich, spezifische N-Fettsäure-Sphingolipide, Cerebroside und Ganglioside darzustellen. Die Reaktion ist frei von Razematen. Die gewünschten Produkte entstehen mit einer Ausbeute von 85 - 90 %. Vorteil dieser Methode ist es, dass die spezifisch veresterten Ganglioside in hoch reiner Form vorliegen. Die modifizierten Glycosphingohpide können entsprechend einem Mischungsverhältnis der
Acylkettenspezifikation mit anderen Komponenten neu zusammengestellt werden.
Beispiel 4:
Chemische Synthese:
Glycosphingohpide werden durch vollständige chemische Synthese nach Duclos (Duclos R.I., The total synthesis of ganglioside GM3. 2000 Carbohydr. Res. 328, 489-507) mit spezifischen Fettsäuren chemisch oder chemoenzymatisch synthetisiert. Die synthetisch rein erzeugten Glycosphingohpide mit einer definierten Fettsäurespezifität können entsprechend einem Mischungsverhältnis der Acylkettenspezifikation mit anderen Komponenten neu zusammengestellt werden.
Beispiel 5:
Herstellung von Babynahrungen:
Zur Bereitstellung von Glycosphingolipiden zur Herstellung von Babynahrung wird Buttermilchtrockenmasse verwendet. Die Fettsäureverteilung von Gangliosid-Extrakten aus Buttermilch mit einer Fettverteilung der N-Acyl-Fettsäuren von C18:0 (10-20 Gew.-%) und C20:0 (0.01- 0.5 Gew.-%) bezogen auf die Gesamtmenge der N-Acyl- Fettsäuren wird durch Mischen im Verhältnis 1 zu 6 mit einem in der Fettsäureverteilung modifizierten Rohstoff (siehe unten), mit einer Fettverteilung der N-Acyl-Fettsäuren von C18:0 (6-9 Gew.-%) und C20:0 (60- 90 Gew.-%) zu einer neuen N-Acyl-Fettsäureverteilung von C18:0 1- 3 Gew.-% und C20:0 10-15 Gew.-% eingestellt. Der in der Fettsäureverteilung von Glycosphingolipiden modifizierte Rohstoff wird, bezogen auf 100 g Feststoffgehalt einer Formelnahrung, wie folgt hergestellt: Die Glycosphingohpide, vorwiegend GM1 , GM3, GD1a, GD1 b aus Buttermilch (Uelzena, Germany) mit einem Fettanteil von 10-15%, wird nach Ladisch und Gillard (Ladisch S., Gillard B. A solvent partition method for morcoscale ganglioside purification. 1984 Analytical Biochemistry 146, 220-231 ) extrahiert und aufgereinigt. 10g Buttermilch Trockenmasse (entspricht ca. 1 Liter Milch) werden mit 50 ml Methanol und 50 ml Chloroform vermengt, 15 Minuten extrahiert und anschließend bei 4°C pelletiert. Der Überstand wird über Rotationsverdampfen oder Lyophilization auf ca. % des Ausgangsvolumens eingeengt. Darauffolgend werden die Proteinanteile der Lösung bei -20°C gefällt und pelletiert. Der Überstand wird bis zur Trockne eingeengt. Zur Partitionierung wird das Extrakt mit 60 ml Diisopropylether/1-Butanol (v/v) 60:40 resuspendiert und darauffolgend 30 ml Wasser zugegeben. Es erfolgt eine Phasentrennung. Die wässrige Phase enthält den Glycosphingolipidanteil (ca. 1 g/l) des Rohstoffs. Die Fettsäuremodifikation wird wie folgt durchgeführt: 1g Glycosphinolipid- Extrakt (ca. 0.83 mmol bei MG 1200) wird in 10 ml 20 mM Natriumphosphat-Lösung mit 0.1 % Triton X100 suspendiert, 100 U Ceramide-N-Deacylase hinzugefügt und 1-2 Stunden bei 37°C inkubiert. Die Hydrolyse der N-Acyl-Fettsäuren erfolgt bei pH 5-6. Darauffolgend wird die Lösung durch Natriumphosphatgabe auf 25 mM Natriumphosphat Endkonzentration eingestellt. Die spezifische freie Fettsäure wird im molaren Verhältnis 1 :10 zum eingesetzten Glycosphingolipid (8.3 mmol C20:0) zugegeben. Die Transesterifikation erfolgt bei 37°C bei pH 7-8 in 1 -2 Stunden. Die modifizierten Glycosphingohpide werden über eine präparative Chromatographie (RPC18) aufgereinigt. Hierbei erfolgt die Konditionierung der Phase mit Chloroform/Methanol/0.1 M KCI-Lösung (v/v/v) 3:98:74. Die Reaktionslösung wird auf die Säule appliziert. Die Säule wird mit Chloroform/Methanol/0.1 M KCI-Lösung (v/v/v) 3:98:74, Wasser und mit Methanol/Wasser (v/v) 1 :1 gewaschen. Die anschließende Elution der Glycosphingohpide erfolgt mit Ethanol und Chloroform/Methanol (v/v) 2:1. Das gewonnene Eluat wird bis zur Trockne eingeengt. Aus dem Prozess resultieren ca. 0.8 g Glycosphingohpide pro Liter Buttermilch mit einer Fettverteilung der N- Acyl-Fettsäuren von C18:0 (6-9 Gew.-%) und C20:0 (60- 90 Gew.-%) bezogen auf die Gesamtmenge der N-Acyl-Fettsäuren. Eine im Eiweiß adaptierte Säuglingsmilchnahrung (Aptamil® von Milupa) mit 11 ,8 g Eiweiß, 56,9 g Kohlenhydrate 24,9 g Fett, 2,5 g Mineralstoffen, Vitaminen und 45 mg Taurin wird auf übliche Weise in Form eines Perlates hergestellt. Die in a) beschriebene Gangliosidmischung wird in einer Menge von 0,2-500 mg/ 100 mg, bezogen auf den Feststoffgehalt der Formelnahrung zugegeben.
Beispiel 6:
Nahrungssupplemente:
Gangliosid-Extrakte aus Kuh-Kolostralmilch, Eilecithin oder Buttermilch wurden Modifikation der N-Acylfettsäureverteiiung oder durch Mischen eines Rohstoffes mit einem in der Fettsäureverteilung modifizierten Rohstoff (siehe oben) mit einer Fettverteilung der N-Acyl-Fettsäuren von zu einer Fettsäureverteilung von 1-3 Gew.-% C18.0 und 10-15 Gew.-% C20:0 bezogen auf die Gesamtmenge der N-Acyl-Fettsäuren eingestellt.
Die beschriebenen Gangliosidmischung wird in Softgel Kapseln zu 10 bis 50 mg/Kapsel formuliert.
Beispiel 7:
Produkt für Schwangere
Eine Brausetablette (Endgewicht 4,15 g) (Neovin® von Milupa wird unter Beimengung von 10 bis 50 mg der beschriebenen Gangliosidmischung auf per se bekannte Weise hergestellt. Täglich wird eine Tablette in 150 ml Wasser gelöst und getrunken.
Beispiel 8:
Produkt für Ältere und geschwächte Personen
Eine bilanzierte pulverförmige Aufbaunahrung (Dilsana® von Milupa) mit 22,5 g Eiweiß, 7,7 g Fett, 60,8 g Kohlenhydrate, 5,4 g Mineralstoffen und Vitaminen wird auf per se bekannte Weise unter Einarbeitung von 0,2- 500 mg der beschriebenen Gangliosidmischung bezogen auf 100 mg Feststoffgehalt der Formelnahrung hergestellt. Täglich werden bis zu 3 x 50 g der Nahrung in 150 ml Wasser gelöst und verabreicht.

Claims

Titel: Ganglioside mit modifizierter AcylfunktionPATENTANSPRÜCHE
1. Gangliosidmischung enthaltend Ganglioside oder bestehend aus Gangliosiden der folgenden allgemeinen Formel I: (Zucker)-OCH2-CH(-NH-CO-R1)-CH(OH)-CH=CH-R2 (I), worin
(Zucker) für einen Zuckerrest steht, die Gruppe -CO-R1 eine Acyl-Fettsäure darstellt, die in Form eines
Amides an den Rest des Moleküls gebunden ist, R1 einen geraden, gesättigten Alkylrest mit mindestens 10 C-Atomen bedeutet und
R2 einen geraden, gesättigten Alkylrest mit mindestens 10 C-Atomen oder einen geraden Alkenylrest mit mindestens 10 C-Atomen und einer, zwei oder drei Doppelbindung(en) bedeutet, dadurch g eke n nzei chn et, dass bei den in der Mischung vorhandenen Gangliosiden der allgemeinen Formel I mindestens 10 Gew.-% der Gruppe -CO-R1 und somit der Acyl-Fettsäuren eine C20:0 Fettsäure sind.
2. Gangliosidmischung nach Anspruch 1, dadurch g eke n nzeichn et, dass die C20:0 Fettsäure 10 bis 15 Gew.-% der Gruppe -CO-R1 und somit der Acyl-Fettsäuren ausmacht.
3. Gangliosidmischung nach Anspruch 1 oder 2, dadurch g eke n nzei c hn et, dass die Gruppe -CO-R1 eine C18:0 Fettsäure bedeuten kann und das
Gewichtsverhältnis dieser C18:0 Fettsäure zu der C20:0 Fettsäure der Gruppe -CO-R11,0 bis 3,0 beträgt.
4. Gangliosidmischung nach einem der vorhergehenden Ansprüche, dadurch g eke n nzeichnet, dass die Gruppe -CO-R1 eine C23:0 Fettsäure bedeuten kann, welche 10 Gew.-% oder weniger der Acyl-Fettsäuren im Lipidteil des Moleküls der Ganglioside ausmacht.
5. Gangliosidmischung nach einem der vorhergehenden Ansprüche, dadurch geken n ze i ch n et, dass die Gangliosidmischung zusammengesetzt ist einerseits aus unmodifizierten Gangliosiden, die aus natürlich vorkommenden Quellen tierischen und/oder pflanzlichen Ursprungs gewonnen wurden, und andererseits aus Gangliosiden, die ebenfalls aus natürlich vorkommenden Quellen tierischen und/oder pflanzlichen Ursprungs gewonnen wurden und dann jedoch modifiziert wurden.
6. Gangliosidmischung nach einem der vorhergehenden Ansprüche, dadurch geken n ze i ch n et , dass sie als wässrige Emulsion vorliegt.
7. Gangliosidmischung nach einem der vorhergehenden Ansprüche, dadurch g eken n ze i ch net , dass sie einen Bestandteil einer Fettmischung darstellt.
8. Gangliosidmischung nach einem der vorhergehenden Ansprüche, dadurch geke n n ze i ch net , dass sie einer Fertignahrung, einem Nahrungssupplement oder einer Formelnahrung einverleibt ist.
9. Verwendung einer Gangliosidmischung nach einem der Ansprüche 1 bis 7 zur Herstellung eines diätetischen, pharmazeutischen oder Nahrungs-Mittels.
10. Verwendung nach Anspruch 9, dadurch geken n ze i ch n et , dass es sich bei dem Nahrungs-Mittel um eine Fertignahrung, ein Nahrungssupplement oder eine Formelnahrung handelt.
11. Diätetisches, pharmazeutisches und Nahrungs-Mittel enthaltend eine Gangliosidmischung nach einem der Ansprüche 1 bis 7.
